Study on the Chemical Composition of Wide leaved Golden Corn Orchid
Chloranthus henryi Hemsl. is a Chloranthus Swartz plant belonging to the Chloranthus genus of the family Chloranthus. It is also known as the Four Blocks Tile, Four Heavenly Kings, Big Leaf, and Self. It is often used as a whole plant in medicine, with a pungent taste, warm nature, and toxicity. It has the effects of dispelling wind and dampness, promoting blood circulation and blood stasis, and detoxifying. It is widely used to treat rheumatism, pain, fractures, sores, swelling, and snake bites. Pharmacological studies have shown that the broad leaved golden corn orchid has anti-inflammatory, antibacterial, anti-tumor, and uterine contraction activities. Kuan Ye Jin Su Lan, as one of the commonly used Chinese herbal medicines in Jiangxi, is included in the Chinese Pharmacopoeia as the main ingredient in the preparation of Zhuanshan Feng Syrup, which is clinically used for the treatment of rheumatoid arthritis. However, the effective anti-inflammatory ingredients are not clear, which affects the clinical application and deep development of the product of Osmanthus fragrans. Therefore, in order to conduct in-depth research on the plant chemical resources of Osmanthus fragrans and search for chemical components with anti-inflammatory activity, this study investigated the chemical composition and in vitro anti-inflammatory activity of the dichloromethane fraction of the 95% ethanol extract of Osmanthus fragrans, in order to explore the anti-inflammatory active substance basis of Osmanthus fragrans.
This article applies modern chromatographic techniques to separate and study the dichloromethane fraction of the 95% ethanol extract of Platycodon grandiflorus, and obtains 19 compounds, including 6 sesquiterpenes, 4 diterpenes, 5 amides, 1 flavonoid, and 3 phenolic acids. Fifteen compounds were isolated for the first time from this plant, among which six compounds were isolated for the first time from the genus Cymbidium. As NO is one of the most important inflammatory mediators involved in the pathogenesis of various inflammatory diseases, it is widely used to evaluate anti-inflammatory activity. Therefore, based on the study of chemical composition, this article isolated compounds 1-9, 12, and 13 in large quantities, and used lipopolysaccharide (LPS) – induced RAW264.7 cells as an in vitro inflammatory model to evaluate the anti-inflammatory activity of the compounds through inhibition of nitric oxide (NO) production experiments. The results showed that terpenoid compounds 1, 5, and 7, as well as amide compounds 12 and 13, exhibited weak anti-inflammatory activity, while sesquiterpene compound 2 had a high content and showed good anti-inflammatory activity, suggesting that compound 2 may be one of the pharmacological substances underlying the anti-inflammatory effects of Platycodon grandiflorus. Therefore, the content of this study not only enriches the chemical composition information of Osmanthus fragrans, but also provides a material basis and scientific basis for expanding the anti-inflammatory medicinal scope of the folk Chinese medicine Osmanthus fragrans.
