Research on the Chemical Composition and Anti inflammatory Activity of South China Wuyu
Euodia rutaecarpa, belonging to the Rutaceae family and the genus Euodia, is a famous traditional Chinese medicine. In traditional Eastern medicine, it is a very popular multi-purpose herb used to treat gastrointestinal diseases, headaches, amenorrhea, and postpartum hemorrhage. In the study of plant chemistry, various compounds have been discovered in the fruit of Evodia rutaecarpa, including limonin, evodiamine, rutaecarpine, as well as other alkaloids and volatile oil components. Alkaloids and limonoid compounds have been proven to have excellent cardiovascular and cerebrovascular activity, anti-tumor activity, as well as anti-inflammatory and analgesic activity. Euodia Austronesis Hand Mazz, also belonging to the genus Euodia, was initially used as a counterfeit of Euodia in the medicinal market and has a high degree of similarity with it. Considering that many valuable compounds have been isolated from Eucommia ulmoides, and there are currently no reports on the chemical composition of Eucommia ulmoides in southern China, this study selected the above ground branches and leaves of Eucommia ulmoides in southern China for research.
The chemical components of Wuyu have excellent anti-inflammatory and analgesic activities. To verify whether the chemical components of South China Wuyu also have similar activities, anti-inflammatory activity related tests were conducted on the compounds. Lactate dehydrogenase (LDH) is a stable cytoplasmic enzyme present in all cells. When cellular inflammation is activated, holes are punched on the surface of the cell membrane, and lactate dehydrogenase is released into the cell culture medium through the damaged cell membrane. Therefore, detecting the release level of LDH in cell culture medium can characterize the anti-inflammatory ability of drugs. Therefore, this study used LDH method to characterize the anti-inflammatory activity of compounds, in order to discover compounds with anti-inflammatory activity and provide reference for the treatment of inflammatory related diseases.

 

In this study, 13 compounds were isolated from southern Chinese Wu Yu, and their structures were identified using techniques such as nuclear magnetic resonance spectroscopy and mass spectrometry. Among them, compounds 6, 8, 9, and 12 were isolated for the first time from plants of the Wu Yu genus, while the remaining compounds were isolated for the first time from southern Chinese Wu Yu. The LDH release inhibition activity of the compound was tested, and compound 13 showed good inhibitory effect, proving its anti-inflammatory activity and providing a reference for the development and utilization of anti-inflammatory drugs in South China Wuyu. This study provides a certain reference basis for clarifying the chemical composition and pharmacological substance basis of South China Wuyu.
