A Hippo-hoz kapcsol\u00f3d\u00f3 aktiv\u00e1l\u00f3 faktorok hi\u00e1ny\u00e1ban a nem foszforil\u00e1lt YAP a sejtmagba ker\u00fcl, \u00e9s a TEAD\/TEF csal\u00e1d transzkripci\u00f3t aktiv\u00e1l\u00f3 faktor\u00e1val k\u00f6lcs\u00f6nhat\u00e1sba l\u00e9pve szab\u00e1lyozza a c\u00e9lg\u00e9nek expresszi\u00f3j\u00e1t. Az extracellul\u00e1ris stimul\u00e1ci\u00f3s jelek egy tiszt\u00e1zatlan mechanizmuson kereszt\u00fcl aktiv\u00e1lhatj\u00e1k az Mst1\/2-t, majd egy sor kaszk\u00e1d foszforil\u00e1ci\u00f3s reakci\u00f3n kereszt\u00fcl foszforil\u00e1lj\u00e1k a t\u00f6bbi magtagot.<br \/>\nEz\u00e9rt ez a tanulm\u00e1ny a m\u00e9hnyakr\u00e1k Ca Ski sejtekre \u00f6sszpontos\u00edt, \u00e9s megvizsg\u00e1lja az RCE-4 hat\u00e1s\u00e1t \u00e9s molekul\u00e1ris mechanizmusait a m\u00e9hnyakr\u00e1k prolifer\u00e1ci\u00f3j\u00e1ra, inv\u00e1zi\u00f3j\u00e1ra \u00e9s migr\u00e1ci\u00f3j\u00e1ra a Wnt\/\u03b2-katenin \u00e9s Hippo\/YAP jel\u00e1tviteli utakon kereszt\u00fcl, elm\u00e9leti t\u00e1mogat\u00e1st ny\u00fajtva az RCE-4 j\u00f6v\u0151beli klinikai alkalmaz\u00e1s\u00e1hoz.<\/p>\n<p>Laborat\u00f3riumunk MTT-k\u00eds\u00e9rletekkel kimutatta, hogy az RCE-4 id\u0151- \u00e9s d\u00f3zisf\u00fcgg\u0151 m\u00f3don k\u00e9pes g\u00e1tolni a Ca Ski sejtek prolifer\u00e1ci\u00f3j\u00e1t. A jelen tanulm\u00e1nyban v\u00e9gzett kl\u00f3nk\u00e9pz\u00e9si k\u00eds\u00e9rlet eredm\u00e9nyei tov\u00e1bb valid\u00e1lj\u00e1k az RCE-4 hat\u00e9konys\u00e1g\u00e1t. Ezenk\u00edv\u00fcl az RCE-4 hat\u00e1s\u00e1t a Ca Ski sejtek inv\u00e1zi\u00f3j\u00e1ra \u00e9s migr\u00e1ci\u00f3j\u00e1ra karcol\u00e1sos k\u00eds\u00e9rletekkel \u00e9s Transwell-kamr\u00e1s in vitro inv\u00e1zi\u00f3s k\u00eds\u00e9rletekkel mutattuk ki. Az eredm\u00e9nyek azt mutatt\u00e1k, hogy az RCE-4 jelent\u0151sen g\u00e1tolta a Ca Ski sejtek migr\u00e1ci\u00f3s \u00e9s inv\u00e1zi\u00f3s k\u00e9pess\u00e9g\u00e9t, \u00e9s koncentr\u00e1ci\u00f3f\u00fcgg\u00e9st mutatott.<br \/>\nA Wnt\/\u03b2-katenin jel\u00e1tviteli \u00fatvonal fontos szerepet j\u00e1tszik a tumormigr\u00e1ci\u00f3ban, \u00e9s a \u03b2-katenin bejut\u00e1sa a sejtmagba a kulcs az \u00fatvonal aktiv\u00e1l\u00e1s\u00e1hoz. Az eredm\u00e9nyek azt mutatt\u00e1k, hogy az RCE-4 h\u00e1rom \u00fatvonalon kereszt\u00fcl k\u00e9pes g\u00e1tolni a \u03b2 - katenin nukle\u00e1ris bejut\u00e1s\u00e1t: g\u00e1tolja a p-GSK3 \u03b2 expresszi\u00f3j\u00e1t, el\u0151seg\u00edti a \u03b2 - katenin foszforil\u00e1ci\u00f3j\u00e1t, \u00e9s g\u00e1tolja a PAK4 expresszi\u00f3j\u00e1t, ez\u00e1ltal g\u00e1tolja a downstream g\u00e9nek, p\u00e9ld\u00e1ul az MMP2\/7\/9\/14 m\u00e1trix metalloprotein\u00e1zok transzkripci\u00f3j\u00e1t. \u00c9rdekes m\u00f3don a leg\u00fajabb kutat\u00e1sok szerint a PAK4 aktivit\u00e1s\u00e1nak g\u00e1tl\u00e1sa szinergikusan fokozhatja a PD-1 immunblokkol\u00f3 ter\u00e1pia ter\u00e1pi\u00e1s hat\u00e1s\u00e1t. A PD-1 ter\u00e1pia kev\u00e9ss\u00e9 hat\u00e1sos, ha a tumorsz\u00f6vetben nincs el\u0151zetes immunsejt-infiltr\u00e1ci\u00f3, ami egyben a PD-1 ter\u00e1pi\u00e1val szembeni rezisztencia kialakul\u00e1s\u00e1nak f\u0151 oka is. A PAK4-g\u00e1tl\u00f3k alkalmaz\u00e1sa jelent\u0151sen fokozhatja a tumorsz\u00f6vetek PD-1-blok\u00e1d ter\u00e1pi\u00e1ra adott v\u00e1lasz\u00e1t in vivo. Figyelembe v\u00e9ve az RCE-4 jelent\u0151s g\u00e1tl\u00f3 hat\u00e1s\u00e1t a PAK4-re, ez \u00faj kutat\u00e1si ir\u00e1nyt ad sz\u00e1munkra.<br \/>\nA Hippo\/YAP jel\u00e1tviteli \u00fatvonal \u00e9s a tumorsejtek inv\u00e1zi\u00f3ja k\u00f6z\u00f6tti kapcsolatot szint\u00e9n meger\u0151s\u00edtett\u00e9k, de eddig viszonylag kev\u00e9s olyan gy\u00f3gyszer l\u00e9tezik, amely a Hippo\/YAP jel\u00e1tviteli \u00fatvonalat c\u00e9lozza. 2018-ban Gill \u00e9s munkat\u00e1rsai meg\u00e1llap\u00edtott\u00e1k, hogy a YAP\/TAZ molekul\u00e1k sejtmagba val\u00f3 bejut\u00e1sa k\u00f6r\u00fcl egy speci\u00e1lis pozit\u00edv visszacsatol\u00e1si hurok van, amely \u00f6t feh\u00e9rjemolekul\u00e1b\u00f3l \u00e1ll: NUAK2, LATS1, p-LATS1, YAP\/TAZ a citoplazm\u00e1ban \u00e9s a sejtmagban. E ciklus felfedez\u00e9se \u00f6sszetettebb\u00e9 teszi a Hippo\/YAP jel\u00e1tviteli \u00fatvonalat, \u00e9s megmagyar\u00e1zza a Hippo\/YAP jel\u00e1tviteli \u00fatvonal szab\u00e1lyoz\u00e1s\u00e1nak neh\u00e9zs\u00e9g\u00e9t a tumorsejtek inv\u00e1zi\u00f3j\u00e1nak g\u00e1tl\u00e1sa \u00e9rdek\u00e9ben. A k\u00eds\u00e9rlet eredm\u00e9nyei azt mutatt\u00e1k, hogy az RCE-4 t\u00f6bb \u00fatvonalon kereszt\u00fcl k\u00e9pes g\u00e1tolni a YAP\/TAZ nukle\u00e1ris bel\u00e9p\u00e9s\u00e9t, bele\u00e9rtve az MST1 foszforil\u00e1ci\u00f3j\u00e1nak g\u00e1tl\u00e1s\u00e1t, a YAP foszforil\u00e1ci\u00f3j\u00e1nak fokoz\u00e1s\u00e1t \u00e9s a YAP expresszi\u00f3j\u00e1nak cs\u00f6kkent\u00e9s\u00e9t. A YAP\/TAZ sejtmagba val\u00f3 bejut\u00e1s\u00e1nak cs\u00f6kken\u00e9se a NUAK2 expresszi\u00f3j\u00e1nak cs\u00f6kken\u00e9s\u00e9hez vezet, \u00edgy megszak\u00edtva ezt a pozit\u00edv visszacsatol\u00e1si hurkot. A ciklus megszakad\u00e1sa a downstream feh\u00e9rj\u00e9k, p\u00e9ld\u00e1ul az intercellul\u00e1ris adh\u00e9zi\u00f3s molekula-1 (ICAM-1), a vaszkul\u00e1ris sejtadh\u00e9zi\u00f3s molekula-1 (VCAM-1), a CYR61 \u00e9s a SOX2 expresszi\u00f3j\u00e1nak cs\u00f6kken\u00e9s\u00e9hez vezet, jelent\u0151sen g\u00e1tolva a Ca Ski sejtek migr\u00e1ci\u00f3s k\u00e9pess\u00e9g\u00e9t.<br \/>\n\u00c9rdemes megjegyezni, hogy a Wnt\/\u03b2-katenin \u00e9s a Hippo\/YAP jel\u00e1tviteli \u00fatvonalak a sejtekben nem teljesen f\u00fcggetlenek egym\u00e1st\u00f3l, \u00e9s sz\u00e1mos m\u00f3don k\u00f6lcs\u00f6nhat\u00e1sba l\u00e9pnek egym\u00e1ssal. P\u00e9ld\u00e1ul a foszforil\u00e1lt YAP\/TAZ k\u00f6t\u0151dhet a \u03b2 - kateninhez, aminek k\u00f6vetkezt\u00e9ben az a citoplazm\u00e1ban marad, \u00e9s g\u00e1tolja annak transzkripci\u00f3s aktivit\u00e1s\u00e1t. Ugyanakkor a kutat\u00e1s ezen szakasz\u00e1nak vannak bizonyos korl\u00e1tai is.
