Des \u00e9tudes ant\u00e9rieures ont montr\u00e9 que les champignons appartenant au genre Trichoderma peuvent produire des m\u00e9tabolites secondaires de divers types structurels, parmi lesquels les relaxants cellulaires et les compos\u00e9s d'azaphilone sont les principaux. En m\u00eame temps, les m\u00e9tabolites secondaires des champignons Trichoderma ont un large \u00e9ventail d'activit\u00e9s biologiques, telles que les activit\u00e9s antibact\u00e9riennes, anti-inflammatoires, cytotoxiques, etc. Notre groupe de recherche se consacre depuis longtemps \u00e0 la d\u00e9couverte de m\u00e9tabolites secondaires et de leurs activit\u00e9s biologiques dans les champignons du genre Trichoderma. Nous avons isol\u00e9 divers types de substances actives avec diff\u00e9rents types de structure \u00e0 partir de m\u00e9tabolites secondaires de Trichoderma, tels que C. cochlides, C. convolutum, C. globorum CIB-160 et C. gracile. C. funicola est un pathog\u00e8ne potentiel qui a \u00e9t\u00e9 trouv\u00e9 dans des tissus cutan\u00e9s infect\u00e9s. En 1969, un compos\u00e9 de lactone macrocyclique a \u00e9t\u00e9 isol\u00e9 \u00e0 partir de Trichoderma lucidum. En 1987, Itoh et al. l'ont isol\u00e9 \u00e0 partir de C Funicola JS 525 compos\u00e9s d'ubiquinone isol\u00e9s. \u00c0 l'heure actuelle, il n'existe que deux rapports de recherche sur les m\u00e9tabolites secondaires de l'\u00e9caille de la corde, et les rapports de recherche sur l'activit\u00e9 biologique pertinente font d\u00e9faut. Cet article \u00e9tudie syst\u00e9matiquement les m\u00e9tabolites secondaires d'une souche de Trichoderma funicola CIB-604 et teste les activit\u00e9s antibact\u00e9riennes et cytotoxiques de certains compos\u00e9s. Nous pr\u00e9sentons ici la structure et l'activit\u00e9 biologique des m\u00e9tabolites secondaires produits par cette bact\u00e9rie.<\/p>\n<p>Dans cette \u00e9tude, huit m\u00e9tabolites secondaires ont \u00e9t\u00e9 isol\u00e9s de la fermentation en milieu solide du riz par une souche d'Aspergillus niger (C. funicola CIB-604), dont trois alcalo\u00efdes isoquinol\u00e9iques (1-3) et cinq compos\u00e9s st\u00e9roliques (4-8), tous isol\u00e9s d'Aspergillus niger pour la premi\u00e8re fois. Les compos\u00e9s 1-3 sont les premiers alcalo\u00efdes quinol\u00e9iniques structurellement nouveaux d\u00e9couverts par notre groupe de recherche \u00e0 partir des m\u00e9tabolites secondaires de C. indica 3.202 en Inde. Jusqu'\u00e0 pr\u00e9sent, Aspergillus niger est le deuxi\u00e8me champignon capable de produire ces nouveaux alcalo\u00efdes de quinol\u00e9ine, ce qui sugg\u00e8re qu'il pourrait contenir la m\u00eame voie de biosynth\u00e8se des alcalo\u00efdes de quinol\u00e9ine que C. indica. Cet article a r\u00e9alis\u00e9 des tests d'activit\u00e9 antibact\u00e9rienne et cytotoxique sur les compos\u00e9s 1 \u00e0 6. Les r\u00e9sultats de l'activit\u00e9 antibact\u00e9rienne ont montr\u00e9 qu'\u00e0 une concentration de 100 \u03bc g\/mL, les compos\u00e9s 1-6 ne pr\u00e9sentaient pas d'effets inhibiteurs sur Escherichia coli, Bacillus subtilis et Candida albicans ; Les r\u00e9sultats de l'activit\u00e9 cytotoxique ont montr\u00e9 que les compos\u00e9s 2 et 4 pr\u00e9sentaient une activit\u00e9 cytotoxique contre les cellules tumorales humaines SK-Hep-1, A549, HCT-116 et MCF-7, et que le compos\u00e9 4 avait un effet inhibiteur plus fort sur les cellules tumorales que le compos\u00e9 2 ; Les r\u00e9sultats de l'analyse par cytom\u00e9trie de flux ont montr\u00e9 que les compos\u00e9s 2 et 4 induisaient principalement une apoptose pr\u00e9coce dans les cellules SK-Hep-1, tandis que le compos\u00e9 2 avait peu d'effet sur l'apoptose pr\u00e9coce et tardive dans les cellules A549, tandis que le compos\u00e9 4 induisait principalement une apoptose tardive dans les cellules A549. Cet article rapporte pour la premi\u00e8re fois l'activit\u00e9 cytotoxique du compos\u00e9 2 sur les cellules tumorales. Les r\u00e9sultats de l'analyse montrent que le compos\u00e9 3 est un diast\u00e9r\u00e9o-isom\u00e8re du compos\u00e9 2, mais que son activit\u00e9 cytotoxique est significativement plus faible que celle du compos\u00e9 2, ce qui indique que la configuration relative entre les groupes hydroxyle et isopropyle a un impact significatif sur son activit\u00e9 cytotoxique. Compar\u00e9 aux compos\u00e9s 5 et 6, le compos\u00e9 4 pr\u00e9sente une activit\u00e9 cytotoxique significative. L'analyse de ses caract\u00e9ristiques structurelles r\u00e9v\u00e8le un centre actif typique - \u03b1, \u03b2 - unit\u00e9 structurelle carbonyl\u00e9e insatur\u00e9e - dans la structure mol\u00e9culaire du compos\u00e9 4, qui peut subir une r\u00e9action d'alkylation avec les groupes thiol des prot\u00e9ines dans les cellules, se liant ainsi aux cellules tumorales et les tuant.
