Les produits naturels tels que la curcumine, le resv\u00e9ratrol et les gins\u00e9nosides ont montr\u00e9 de bons effets neuroprotecteurs en r\u00e9gulant les ROS, en ayant des effets anti-inflammatoires et en inhibant l'excitotoxicit\u00e9. Ils ont des applications prometteuses dans la lutte contre les l\u00e9sions c\u00e9r\u00e9brales isch\u00e9miques et l'am\u00e9lioration des capacit\u00e9s d'apprentissage et cognitives.<br \/>\nSargentodoxa cuneta (Oliv.) Rehd Et Wils. est une plante unique en Chine, qui a des effets de d\u00e9sintoxication, de dissipation des escarboucles, de promotion de la circulation sanguine, de soulagement de la douleur et de dissipation du vent et de l'humidit\u00e9. Les composants chimiques de la vigne rouge comprennent principalement des acides ph\u00e9noliques, des ph\u00e9nylpropano\u00efdes, des lignanes, etc. Ses extraits ont des activit\u00e9s pharmacologiques telles que des effets anti-inflammatoires, antioxydants et neuroprotecteurs. Dans le cadre de recherches pr\u00e9liminaires, notre laboratoire a isol\u00e9 le compos\u00e9 glycoside ph\u00e9nylpropano\u00efde sargentol (190 mg isol\u00e9s \u00e0 partir de 6 kg de tiges de vigne) \u00e0 partir de vignes rouges sauvages, mais il existe peu de rapports sur l'activit\u00e9 pharmacologique de ce compos\u00e9. Cet article vise \u00e0 \u00e9valuer l'effet protecteur du sargentol sur le mod\u00e8le de l\u00e9sion des cellules PC12 et \u00e0 discuter de son m\u00e9canisme d'action possible sur la base de la technologie de docking mol\u00e9culaire, afin de fournir une base scientifique pour la d\u00e9couverte d'ingr\u00e9dients actifs neuroprotecteurs \u00e0 partir de sources de la m\u00e9decine traditionnelle chinoise.<\/p>\n<p>Les l\u00e9sions induites par la fis\u00e9tine sur les cellules PC12 sont couramment utilis\u00e9es pour \u00e9tablir un mod\u00e8le cellulaire de la maladie de Parkinson, tandis que les l\u00e9sions induites par le NaN3 sur les cellules PC12 sont couramment utilis\u00e9es pour le criblage pr\u00e9liminaire de compos\u00e9s anti-Alzheimer. Dans cette \u00e9tude, le sargentol n'a pas montr\u00e9 d'inhibition significative des l\u00e9sions des cellules PC12 induites par la fis\u00e9tine et le NaN3 (P&gt;0,05), mais il s'est montr\u00e9 plus sensible au mod\u00e8le de l\u00e9sion H2O2 et a eu un effet inhibiteur significatif sur les ROS intracellulaires induits par H2O2 dans les cellules PC12.<br \/>\nAfin d'\u00e9tudier plus avant le m\u00e9canisme neuroprotecteur du sargentol, le logiciel AutoDock Vina a \u00e9t\u00e9 utilis\u00e9 pour cribler virtuellement ses prot\u00e9ines cibles de liaison dans cette \u00e9tude. Consid\u00e9rant que l'effet ROS est une cause commune de dommages cellulaires ou d'apoptose dans le mod\u00e8le de cellules endommag\u00e9es par H2O2, nous avons s\u00e9lectionn\u00e9 14 prot\u00e9ines, principalement des prot\u00e9ines li\u00e9es \u00e0 l'effet ROS et \u00e0 l'inflammation, pour l'ancrage mol\u00e9culaire.<br \/>\nSur la base de l'\u00e9nergie de liaison et du rapport R, des cibles potentielles du sargentol ont \u00e9t\u00e9 identifi\u00e9es, notamment TLR2 (5D3I), iNOS (2Y37), PI3K (1E7V), Keap1 (4L7B) et IKK \u03b2 (3RZF). La voie de signalisation Keap1-Nrf2-ARE est l'un des m\u00e9canismes les plus importants de d\u00e9fense des cellules contre les dommages caus\u00e9s par les ROS. L'\u00e9nergie de liaison entre Keap1 (4L7B) et le sargentol est de -7,2 kcal\/mol, ce qui indique que le sargentol peut avoir une forte capacit\u00e9 d'activation de la prot\u00e9ine Keap1 et des effets ROS potentiels bas\u00e9s sur la voie de signalisation Keap1-Nrf2-ARE. Cette hypoth\u00e8se est coh\u00e9rente avec les donn\u00e9es de l'exp\u00e9rience 2.2. TLR2 (5D3I), iNOS (2Y37) et PI3K (1E7V) sont des prot\u00e9ines r\u00e9gulatrices importantes des ROS et de l'inflammation. Dans cet ancrage mol\u00e9culaire, leurs \u00e9nergies de liaison au sargentol \u00e9taient respectivement de -7,7, -7,7 et -7,3 kcal\/mol. En particulier pour le premier, la force de liaison a d\u00e9pass\u00e9 celle du ligand original (-7,5 kcal\/mol). Cela sugg\u00e8re que le sargentol pourrait exercer des effets neuroprotecteurs en inhibant le cycle de stress oxydatif de l'inflammation. Des \u00e9tudes ant\u00e9rieures ont montr\u00e9 que le sargentol pouvait inhiber de mani\u00e8re significative le gonflement de l'oreille chez la souris, et son activit\u00e9 anti-inflammatoire est coh\u00e9rente avec la pr\u00e9diction d'amarrage mol\u00e9culaire de cette \u00e9tude. D'apr\u00e8s l'\u00e9nergie de liaison, le sargentol a une faible capacit\u00e9 \u00e0 inhiber l'excitotoxicit\u00e9 et \u00e0 r\u00e9guler les g\u00e8nes d'apoptose.
