CPCC 203679, originaire du sol<br \/>\nLes actinobact\u00e9ries constituent une ressource importante pour la d\u00e9couverte de nouveaux antibiotiques, car elles peuvent produire une vari\u00e9t\u00e9 de produits naturels actifs avec divers types de structure. Streptomyces est le genre dominant des actinomyc\u00e8tes, avec le plus grand nombre d'esp\u00e8ces efficaces rapport\u00e9es. Il est largement r\u00e9pandu dans le sol, l'eau douce, les environnements marins et certains organismes. Streptomyces est connu pour produire une grande vari\u00e9t\u00e9 de m\u00e9tabolites secondaires actifs avec divers types de structure, et est une source importante d'antibiotiques couramment utilis\u00e9s dans la pratique clinique. Actuellement, environ deux tiers des antibiotiques utilis\u00e9s en pratique clinique sont produits par ce genre de micro-organismes. Ces derni\u00e8res ann\u00e9es, un grand nombre de m\u00e9tabolites secondaires structurellement nouveaux et biologiquement actifs ont \u00e9t\u00e9 continuellement signal\u00e9s dans le genre Streptomyces, ce qui indique que la d\u00e9couverte de nouveaux compos\u00e9s principaux \u00e0 partir de ce genre a encore un grand potentiel. Notre groupe de recherche a d\u00e9couvert une souche de Streptomyces sp. CPCC 203679 d\u00e9riv\u00e9e du sol au cours du criblage pr\u00e9liminaire de l'activit\u00e9 biologique. L'extrait brut de son bouillon de fermentation pr\u00e9sente une bonne activit\u00e9 antibact\u00e9rienne. Dans cette \u00e9tude, diverses techniques modernes de chromatographie et de spectroscopie ont \u00e9t\u00e9 utilis\u00e9es pour isoler, purifier et identifier structurellement les m\u00e9tabolites secondaires de la souche, et 9 compos\u00e9s ont \u00e9t\u00e9 obtenus (voir figure 1). Les compos\u00e9s 1-8 ont \u00e9t\u00e9 \u00e9valu\u00e9s pour leurs activit\u00e9s antibact\u00e9riennes et cytotoxiques in vitro.<\/p>\n<p>&nbsp;<\/p>\n<p>Neuf compos\u00e9s ont \u00e9t\u00e9 isol\u00e9s du bouillon de fermentation de Streptomyces sp. CPCC 203679 provenant du sol, y compris des acides gras, des alcalo\u00efdes et des dipeptides cycliques. Le compos\u00e9 1 est un nouveau compos\u00e9 d'acide gras, et le compos\u00e9 3 a \u00e9t\u00e9 isol\u00e9 pour la premi\u00e8re fois \u00e0 partir de Streptomyces provenant du sol. Les r\u00e9sultats de l'\u00e9valuation de l'activit\u00e9 pharmacologique ont montr\u00e9 que les compos\u00e9s 1 \u00e0 8 ne pr\u00e9sentaient pas d'activit\u00e9 antibact\u00e9rienne et antitumorale significative. Le compos\u00e9 2 appartient \u00e0 la classe des alcalo\u00efdes et est largement pr\u00e9sent dans les micro-organismes tels que les actinomyc\u00e8tes, les bact\u00e9ries et les champignons, comme Streptomyces Bacillus\u3001Aspergillus\u3001Alternaria\u3002 Le compos\u00e9 3 appartient \u00e0 la classe des alcalo\u00efdes indoliques et a \u00e9t\u00e9 signal\u00e9 pour la premi\u00e8re fois en 1965. Ce compos\u00e9 a \u00e9galement \u00e9t\u00e9 signal\u00e9 dans des genres tels que Bacillus, Balansia, Edwardsiella, etc. Selon les rapports de la litt\u00e9rature, le compos\u00e9 3 a des effets anti-pollution et un certain effet inhibiteur sur les larves de balanes. Sa valeur ED50 est de 18,6 \u03bc mol\/L. Les dipeptides cycliques sont les plus petits peptides cycliques dans la nature, largement pr\u00e9sents dans les produits m\u00e9taboliques des plantes, des animaux et des micro-organismes, avec des activit\u00e9s telles que les propri\u00e9t\u00e9s antibact\u00e9riennes, antitumorales, neuroprotectrices et antithrombotiques. Cette \u00e9tude n'a pas permis d'isoler des m\u00e9tabolites secondaires ayant une activit\u00e9 biologique significative, ce qui peut \u00eatre d\u00fb au fait que les groupes de g\u00e8nes biosynth\u00e9tiques des m\u00e9tabolites actifs n'ont pas \u00e9t\u00e9 activ\u00e9s dans les conditions de culture actuelles, ou qu'ils n'ont pas \u00e9t\u00e9 isol\u00e9s en raison d'une faible production de m\u00e9tabolites actifs. \u00c0 l'avenir, nous tenterons d'optimiser les conditions de culture afin d'accro\u00eetre la diversit\u00e9 structurelle des m\u00e9tabolites secondaires et de combiner davantage l'\u00e9valuation de l'activit\u00e9 antibact\u00e9rienne afin d'obtenir des m\u00e9tabolites secondaires structurellement nouveaux et significativement actifs.<\/p>","protected":false},"excerpt":{"rendered":"<p>Study on secondary metabolites of soil derived Streptomyces sp.
