Elle est utilis\u00e9e comme plante enti\u00e8re en m\u00e9decine et a pour effet d'\u00e9liminer la chaleur et de d\u00e9sintoxiquer, de favoriser la croissance musculaire et d'arr\u00eater les saignements, de disperser la stase sanguine et de r\u00e9duire les gonflements. Elle peut \u00eatre utilis\u00e9e pour traiter les blessures caus\u00e9es par les chutes, les morsures de serpent, les stases sanguines et les gonflements. Selon la litt\u00e9rature, les racines, les tiges et les feuilles du sapin de Chine contiennent de nombreuses bact\u00e9ries endophytes qui peuvent produire des m\u00e9tabolites structurellement divers lorsqu'elles sont cultiv\u00e9es in vitro ; certains champignons endophytes peuvent \u00e9galement produire de l'huperzine A pendant la fermentation et la culture in vitro. Les recherches pr\u00e9liminaires du groupe de recherche ont permis d'isoler et d'identifier de multiples champignons endophytes \u00e0 partir de Cryptomeria lanceolata. D'autres \u00e9tudes ont \u00e9t\u00e9 men\u00e9es sur leurs produits de fermentation, et une s\u00e9rie de m\u00e9tabolites terp\u00e9no\u00efdes et polyk\u00e9tides structurellement nouveaux ont \u00e9t\u00e9 isol\u00e9s et identifi\u00e9s. Certains compos\u00e9s pr\u00e9sentent des activit\u00e9s anti-inflammatoires et inhibitrices de l'ac\u00e9tylcholinest\u00e9rase significatives in vitro. C. Tenuissimum MT-35 est un champignon endophyte isol\u00e9 et purifi\u00e9 par notre groupe de recherche \u00e0 partir de Cryptomeria acutissima \u00e0 un stade pr\u00e9coce. Son produit de fermentation poss\u00e8de une certaine activit\u00e9 inhibitrice de l'ac\u00e9tylcholinest\u00e9rase. Par cons\u00e9quent, l'\u00e9quipe de recherche a isol\u00e9 et purifi\u00e9 les m\u00e9tabolites de ce champignon endophyte, en les combinant \u00e0 un criblage d'activit\u00e9 in vitro, dans l'espoir d'obtenir des m\u00e9tabolites ayant une activit\u00e9 inhibitrice de l'ac\u00e9tylcholinest\u00e9rase.<\/p>\n<p>&nbsp;<\/p>\n<p>Les micro-organismes peuvent produire des m\u00e9tabolites nouveaux et divers au cours de la fermentation et de la culture in vitro. L'isolement et l'identification des m\u00e9tabolites microbiens, combin\u00e9s \u00e0 un criblage d'activit\u00e9, devraient permettre de d\u00e9couvrir des mol\u00e9cules actives ayant un potentiel de d\u00e9veloppement. C. Tenuissimum est un champignon endophyte isol\u00e9 par notre groupe de recherche \u00e0 partir de Cryptomeria lanceolata \u00e0 un stade pr\u00e9coce. Il a \u00e9t\u00e9 rapport\u00e9 dans la litt\u00e9rature que ce champignon a \u00e9galement \u00e9t\u00e9 isol\u00e9 dans des plantes telles que Pinus wallichiana et Maytenus hookeri. Naseer, Dai et d'autres ont \u00e9tudi\u00e9 ses m\u00e9tabolites ferment\u00e9s dans un milieu g\u00e9los\u00e9 \u00e0 base de glucose de pomme de terre et ont isol\u00e9 plusieurs polyc\u00e9tones macrolides, dont certains ont une activit\u00e9 antibact\u00e9rienne. Dans cette \u00e9tude, dix m\u00e9tabolites ont \u00e9t\u00e9 isol\u00e9s et identifi\u00e9s \u00e0 partir des produits ferment\u00e9s du riz brun de Tenuissimum, dont six compos\u00e9s azot\u00e9s, deux compos\u00e9s st\u00e9ro\u00efdiens et deux compos\u00e9s polyk\u00e9tidiques. Certains de ces compos\u00e9s pr\u00e9sentent des activit\u00e9s biologiques contre l'ac\u00e9tylcholinest\u00e9rase et anti-inflammatoires. Bien que la structure du compos\u00e9 1 ait \u00e9t\u00e9 rapport\u00e9e, il n'existe pas de r\u00e9f\u00e9rence bibliographique pour ses donn\u00e9es de r\u00e9sonance magn\u00e9tique nucl\u00e9aire. Dans cet article, la r\u00e9sonance magn\u00e9tique nucl\u00e9aire bidimensionnelle (RMN 2D) et d'autres m\u00e9thodes ont \u00e9t\u00e9 utilis\u00e9es pour attribuer avec pr\u00e9cision les donn\u00e9es relatives \u00e0 l'hydrog\u00e8ne du carbone. Les r\u00e9sultats des recherches ci-dessus sont utiles pour mieux comprendre Bacillus subtilis C. La d\u00e9couverte des types structurels et des produits naturels actifs des m\u00e9tabolites tenuissimum a une certaine importance de r\u00e9f\u00e9rence. Cependant, dans l'\u00e9tude d'isolement syst\u00e9matique de cette souche, il a \u00e9t\u00e9 constat\u00e9 que les champignons endophytes \u00e9taient cultiv\u00e9s dans les conditions d\u00e9crites dans cet article et que leurs m\u00e9tabolites \u00e9taient tr\u00e8s limit\u00e9s. Les composants macrolides signal\u00e9s dans la litt\u00e9rature n'ont pas \u00e9t\u00e9 isol\u00e9s, ce qui peut \u00eatre d\u00fb \u00e0 des conditions de culture diff\u00e9rentes ou \u00e0 des plantes h\u00f4tes diff\u00e9rentes des champignons endophytes, ce qui entra\u00eene des m\u00e9tabolites diff\u00e9rents. Dans des \u00e9tudes ult\u00e9rieures, il est possible d'essayer de modifier les conditions de culture de la fermentation ou d'explorer diff\u00e9rentes m\u00e9thodes de co-culture fongique. Combin\u00e9es \u00e0 l'analyse par chromatographie liquide et spectrom\u00e9trie de masse (LC-MS), les conditions de culture appropri\u00e9es peuvent \u00eatre s\u00e9lectionn\u00e9es avant d'effectuer la s\u00e9paration guid\u00e9e par LC-MS afin d'\u00e9viter autant que possible les s\u00e9parations r\u00e9p\u00e9titives de bas niveau. D'autre part, avec le d\u00e9veloppement rapide de la technologie de s\u00e9quen\u00e7age du g\u00e9nome, la strat\u00e9gie de d\u00e9couverte de nouveaux produits naturels structurellement actifs bas\u00e9e sur la technologie d'exploration des g\u00e8nes est devenue un domaine de recherche de pointe.
