LSD1 peut activer ou inhiber la transcription des g\u00e8nes en catalysant des r\u00e9actions d'oxydation avec la flavine ad\u00e9nine dinucl\u00e9otide (FAD), en supprimant sp\u00e9cifiquement les groupes m\u00e9thyles simples et doubles de la lysine histone H3K4 et H3K9. LSD1 r\u00e9gule l'expression de prot\u00e9ines apparent\u00e9es et favorise la croissance et la m\u00e9tastase de diverses cellules tumorales en modifiant l'\u00e9tat de m\u00e9thylation des histones dans les r\u00e9gions promotrices des g\u00e8nes associ\u00e9s \u00e0 la prolif\u00e9ration tumorale et \u00e0 la m\u00e9tastase. Actuellement, il a \u00e9t\u00e9 prouv\u00e9 que LSD1 est un moteur important du cancer et une cible th\u00e9rapeutique pour diverses tumeurs. La recherche a montr\u00e9 que divers flavono\u00efdes ont une bonne activit\u00e9 inhibitrice contre LSD1, jetant ainsi les bases de la d\u00e9couverte de nouveaux compos\u00e9s anti-tumoraux. Ses graines sont couramment utilis\u00e9es dans la pratique clinique comme rem\u00e8de chinois nourrissant. Les graines de cuscute ont un go\u00fbt piquant, doux et plat, b\u00e9n\u00e9fique pour les m\u00e9ridiens du foie, des reins et de la rate. Elle a pour effet de nourrir le foie et les reins, de consolider l'essence et de r\u00e9duire l'urine, de stabiliser la grossesse, d'am\u00e9liorer la vision et d'arr\u00eater la diarrh\u00e9e. La recherche pharmacologique moderne a montr\u00e9 que le Fructus Cuscutae a une valeur m\u00e9dicinale significative dans la r\u00e9gulation du syst\u00e8me reproducteur, la lutte contre le vieillissement, la r\u00e9gulation immunitaire, la protection cardiovasculaire et c\u00e9r\u00e9brovasculaire et la r\u00e9duction du taux de sucre dans le sang. La composition chimique de la cuscute comprend principalement des flavono\u00efdes, ainsi que des compos\u00e9s tels que des lignanes, des terp\u00e8nes et des alcalo\u00efdes. Par cons\u00e9quent, afin de mieux comprendre la base de la substance pharmacologique de la m\u00e9decine traditionnelle chinoise Cuscuta chinensis et d'am\u00e9liorer l'efficacit\u00e9 de son utilisation globale des ressources, cette \u00e9tude a isol\u00e9, purifi\u00e9 et identifi\u00e9 structurellement les composants chimiques de la partie m\u00e9thanol de Cuscuta chinensis, et a test\u00e9 l'activit\u00e9 inhibitrice LSD1 des flavono\u00efdes qu'elle contient, afin de d\u00e9couvrir des compos\u00e9s principaux ayant des effets anticanc\u00e9reux.<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>Cet article a isol\u00e9 six compos\u00e9s monom\u00e8res \u00e0 partir de l'extrait de m\u00e9thanol de Cuscuta chinensis. Parmi eux, le compos\u00e9 1 est une forme rac\u00e9mique, un nouvel alcalo\u00efde \u03b2-capolin, les compos\u00e9s 2 \u00e0 5 sont des flavono\u00efdes et le compos\u00e9 6 est un compos\u00e9 lignanique. Les flavono\u00efdes totaux de Cuscuta chinensis ont une bonne activit\u00e9 antitumorale in vitro et in vivo, mais leurs ingr\u00e9dients actifs et leur m\u00e9canisme d'action ne sont pas encore clairs. Par ailleurs, l'activit\u00e9 inhibitrice de la LSD1 de la cuscute n'a pas encore \u00e9t\u00e9 rapport\u00e9e. Cet article est le premier \u00e0 \u00e9valuer l'activit\u00e9 inhibitrice du LSD1 des flavono\u00efdes 2 et 5 du Fructus Cuscutae, et les r\u00e9sultats montrent que les deux compos\u00e9s ont de forts effets inhibiteurs sur le LSD1, jetant les bases de l'am\u00e9lioration de la relation structure-activit\u00e9 de l'activit\u00e9 inhibitrice du LSD des flavono\u00efdes. En outre, la cuscute est riche en flavono\u00efdes, mais il existe peu de rapports sur les composants alcalo\u00efdes.
