D'un point de vue pharmacologique, les compos\u00e9s polyk\u00e9tidiques constituent une source importante de nouveaux m\u00e9dicaments, principalement utilis\u00e9s comme antibiotiques, immunosuppresseurs, agents antiparasitaires, agents hypocholest\u00e9rol\u00e9miants et m\u00e9dicaments antitumoraux. La fonction de chaque domaine de la polyk\u00e9tide synthase correspond \u00e0 la structure du produit final. Cette correspondance peut donc \u00eatre utilis\u00e9e pour modifier les g\u00e8nes de biosynth\u00e8se et concevoir des produits synth\u00e9tiques, cr\u00e9ant ainsi un nouveau domaine de recherche : la biologie combinatoire des polyk\u00e9tides synthases. Les chercheurs g\u00e9n\u00e8rent principalement de nouveaux produits en r\u00e9duisant, en augmentant ou en rempla\u00e7ant les modules et les domaines structurels de la polyk\u00e9tide synthase, par exemple en modifiant le groupe de g\u00e8nes de la polyk\u00e9tide synthase aveLAT-ACP chez Streptomyces avermitilis afin d'obtenir une souche g\u00e9n\u00e9tiquement modifi\u00e9e. Deux nouveaux d\u00e9riv\u00e9s de l'avermectine, la 25 m\u00e9thyl ivermectine et la 25 \u00e9thyl ivermectine, ont \u00e9t\u00e9 isol\u00e9s \u00e0 partir des m\u00e9tabolites secondaires de la souche.<\/p>\n<p>Dans la recherche pr\u00e9c\u00e9dente, notre groupe de recherche a modifi\u00e9 le g\u00e8ne milF codant pour la polyk\u00e9tide synthase dans Streptomyces bingchengensis BC-120-4 pour obtenir la souche g\u00e9n\u00e9tiquement modifi\u00e9e Streptomyces bingchengensis BCJ60. Cette souche ne peut pas coder la C-5 c\u00e9tor\u00e9ductase, ce qui bloque la synth\u00e8se de la milb\u00e9imycine A3\/A4\/B2\/B3. Gr\u00e2ce \u00e0 une modification g\u00e9n\u00e9tique cibl\u00e9e, le groupe de g\u00e8nes de biosynth\u00e8se peut subir des changements, ce qui peut activer certaines voies de biosynth\u00e8se pour obtenir de nouveaux m\u00e9tabolites. Par cons\u00e9quent, cette \u00e9tude se concentre principalement sur les m\u00e9tabolites secondaires de la souche de Streptomyces bingchengensis BCJ60 issue du g\u00e9nie g\u00e9n\u00e9tique, afin d'obtenir des antibiotiques insecticides tr\u00e8s efficaces ayant une valeur pratique.<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>Cet article \u00e9tudie syst\u00e9matiquement les m\u00e9tabolites secondaires de la bact\u00e9rie g\u00e9n\u00e9tiquement modifi\u00e9e Streptomyces bingchengensis BCJ60 et obtient 11 compos\u00e9s, dont les compos\u00e9s 1 et 2 sont deux nouveaux compos\u00e9s. Dans des \u00e9tudes ant\u00e9rieures, notre groupe de recherche a men\u00e9 des recherches pr\u00e9liminaires sur les m\u00e9tabolites secondaires de la bact\u00e9rie g\u00e9n\u00e9tiquement modifi\u00e9e Streptomyces bingchengensis BCJ60 et a obtenu 12 nouveaux compos\u00e9s, qui sont tous des milb\u00e9rellines. Cela indique que, par le biais d'une modification cibl\u00e9e des g\u00e8nes, le groupe de g\u00e8nes biosynth\u00e9tiques subit des changements, activant certaines voies de biosynth\u00e8se et obtenant de nouveaux m\u00e9tabolites, ce qui permet de mieux comprendre le m\u00e9canisme de biosynth\u00e8se des compos\u00e9s de Mirella. L'exp\u00e9rience de l'activit\u00e9 insecticide du nouveau compos\u00e9 a montr\u00e9 que les compos\u00e9s 1 et 2 pr\u00e9sentaient une activit\u00e9 de destruction significative contre les acariens cinabres et les n\u00e9matodes du bois de pin, et qu'il n'y avait pas de diff\u00e9rence significative par rapport \u00e0 la Mirbeimycin A3\/A4 commerciale.
