L'amylopectine \u00e0 cha\u00eene droite est une structure h\u00e9lico\u00efdale non ramifi\u00e9e, form\u00e9e de r\u00e9sidus de glucose reli\u00e9s uniquement par des liaisons \u03b1-1,4-glycosidiques de t\u00eate \u00e0 queue ; l'amylopectine \u00e0 cha\u00eene ramifi\u00e9e est form\u00e9e de r\u00e9sidus de glucose reli\u00e9s par des liaisons \u03b1-1,4-glycosidiques de t\u00eate \u00e0 queue dans chaque cha\u00eene, mais par des liaisons \u03b1-1,6-glycosidiques au niveau des branches.<\/p>\n<p><strong>L'\u03b1-amylase et son r\u00f4le<\/strong><\/p>\n<p>L'\u03b1-amylase, \u00e9galement connue sous le nom d'endoamylase, est une m\u00e9tallohydrolase qui n\u00e9cessite des ions calcium, qui se lient \u00e0 la prot\u00e9ine de l'enzyme pour pr\u00e9senter une activit\u00e9, qui peut \u00eatre perdue par traitement avec l'agent ch\u00e9lateur EDTA.<\/p>\n<p><strong>\u03b1- L'amylase peut hydrolyser au hasard la liaison \u03b1-1,4-glycosidique dans l'amidon, mais ne peut pas hydrolyser la liaison \u03b1-1,6-glycosidique.<\/strong><\/p>\n<p><strong>Si le substrat est un amidon \u00e0 cha\u00eene droite, l'hydrolyse produit du glucose, du maltose et du maltotriose ; si le substrat agissant est un amidon \u00e0 cha\u00eene ramifi\u00e9e, les produits de l'hydrolyse sont le glucose, le maltose, le maltotriose et l'\u03b1-dextrine contenant des liaisons \u03b1-1,6-glycosidiques contenant plus de trois r\u00e9sidus de glucose.<\/strong><\/p>\n<p>Il convient \u00e9galement de noter que les \u03b1 et \u03b2 de l'\u03b1-amylase et de la \u03b2-amylase n'indiquent aucune relation de conformation, mais sont simplement num\u00e9rot\u00e9s.<\/p>\n<p>Il existe plusieurs fa\u00e7ons de produire l'\u03b1-amylase dans la nature, et la fermentation microbienne, par exemple, est l'une des plus courantes. En pr\u00e9sence d'enzymes de d\u00e9branchement, la liaison \u03b1-1,6-glycosidique des esp\u00e8ces \u03b1-dextrine et \u03b2-dextrine est hydrolys\u00e9e et les cha\u00eenes ramifi\u00e9es sont \u00e9limin\u00e9es.<\/p>\n<p>Les cha\u00eenes droites restantes sont ensuite hydrolys\u00e9es par l'\u03b1-amylase et la \u03b2-amylase pour produire du maltose et du glucose. Cependant, les R-amylases ne peuvent pas hydrolyser directement les liaisons \u03b1-1,6-glycosidiques dans les amidons ramifi\u00e9s.<\/p>\n<p><strong>L'amylase glycosylante et son action<\/strong><\/p>\n<p>L'amylase glycosylante, ou glucoamylase, est une enzyme qui hydrolyse l'amidon en glucose, le pourcentage d'hydrolyse pouvant atteindre 100%, et est souvent utilis\u00e9e comme agent saccharifiant pour l'amidon. La glucoamylase n'est pas tr\u00e8s sp\u00e9cifique et s'applique \u00e0 de nombreuses substances. Elle peut non seulement couper la liaison \u03b1-1,4- glycosidique de l'extr\u00e9mit\u00e9 non r\u00e9ductrice de la mol\u00e9cule d'amidon, mais aussi la liaison \u03b1-1,6-glycosidique, mais l'hydrolyse de la liaison \u03b1-1,4-glycosidique est un peu plus rapide.
