Les boutons floraux s\u00e9ch\u00e9s du lotus dormant, une plante m\u00e9dicinale couramment utilis\u00e9e dans la m\u00e9decine ou\u00efghoure, ont \u00e9t\u00e9 inclus dans le volume sur la m\u00e9decine ou\u00efghoure des normes relatives aux m\u00e9dicaments du minist\u00e8re de la sant\u00e9 de la R\u00e9publique populaire de Chine. En utilisant le m\u00e9thanol 50% et l'\u00e9thanol comme solvants d'extraction, l'analyse des spectres d'empreintes digitales de 15 lots de lotus dormant a r\u00e9v\u00e9l\u00e9 l'existence de 23 pics communs \u00e0 une longueur d'onde de 273 nm. Le pic 1 pr\u00e9sentait une bonne s\u00e9paration de l'acide gallique et une grande surface de pic, il a donc \u00e9t\u00e9 utilis\u00e9 comme pic de r\u00e9f\u00e9rence. Il y a 17 pics communs \u00e0 350 nm, parmi lesquels 4 composants ont \u00e9t\u00e9 identifi\u00e9s, \u00e0 savoir l'acide tannique, le glycoside de feu d'artifice, le glycoside de nuage violet et la querc\u00e9tine. Parall\u00e8lement, la similarit\u00e9 des spectres d'empreintes digitales de 15 lots d'\u00e9chantillons a \u00e9t\u00e9 \u00e9valu\u00e9e et les r\u00e9sultats ont montr\u00e9 une bonne similarit\u00e9 avec les spectres d'empreintes digitales de contr\u00f4le. Le faible nombre de pics entra\u00eene une extraction incompl\u00e8te. L'analyse compl\u00e8te de divers facteurs a permis de d\u00e9terminer la m\u00e9thode d'extraction optimale en utilisant le m\u00e9thanol comme solvant.<\/p>\n<p>S\u00e9lection de la longueur d'onde de d\u00e9tection. Cette \u00e9tude a utilis\u00e9 un d\u00e9tecteur \u00e0 barrettes de diodes pour effectuer un balayage complet de la longueur d'onde (190-800nm) sur le spectre de l'empreinte digitale de la fleur de lotus. Sur la base du spectre 3D pr\u00e9sent\u00e9, on peut constater que la s\u00e9paration des pics chromatographiques est \u00e9lev\u00e9e, que la sym\u00e9trie des pics est bonne, que la ligne de base est stable et que la capacit\u00e9 des pics est importante dans les conditions de longueur d'onde de 273nm et 350nm. Le spectre obtenu de la solution \u00e0 tester peut afficher des informations plus riches. C'est pourquoi les longueurs d'onde de 273 nm et 350 nm ont \u00e9t\u00e9 choisies pour les mesures.<\/p>\n<p>S\u00e9lection des conditions chromatographiques. L'influence du m\u00e9thanol et de l'ac\u00e9tonitrile en tant que phases mobiles sur le spectre d'empreintes digitales des fleurs de lotus a \u00e9t\u00e9 \u00e9tudi\u00e9e. Lorsque le m\u00e9thanol et l'ac\u00e9tonitrile sont utilis\u00e9s comme phases mobiles pour une analyse isocratique, l'efficacit\u00e9 de la s\u00e9paration est m\u00e9diocre. Afin d'obtenir une meilleure s\u00e9paration de chaque pic chromatographique, l'\u00e9lution en gradient est choisie. Si l'on utilise le m\u00e9thanol et l'ac\u00e9tonitrile comme phase mobile, il est possible d'obtenir davantage de pics chromatographiques dans le chromatogramme des empreintes digitales. En comparant les chromatogrammes obtenus avec deux phases mobiles diff\u00e9rentes, on a constat\u00e9 que les performances de s\u00e9paration de l'ac\u00e9tonitrile \u00e9taient meilleures que celles du m\u00e9thanol. L'ac\u00e9tonitrile a donc \u00e9t\u00e9 choisi comme phase mobile pour cette \u00e9tude.<br \/>\n\u00c9valuation des m\u00e9thodes d'analyse. L'analyse des spectres d'empreintes digitales de 15 lots de fleurs de lotus endormies a r\u00e9v\u00e9l\u00e9 l'existence de 23 pics communs \u00e0 une longueur d'onde de 273 nm. Le pic 1 pr\u00e9sentait une bonne s\u00e9paration de l'acide gallique et une grande surface de pic, il a donc \u00e9t\u00e9 utilis\u00e9 comme pic de r\u00e9f\u00e9rence. La combinaison de l'empreinte digitale HPLC et de l'analyse de grappes, de l'ACP, de l'OPLS-DA peut \u00eatre utilis\u00e9e pour le regroupement et l'analyse de la discrimination des composants des mati\u00e8res m\u00e9dicinales d'origines et de lots diff\u00e9rents. Cette \u00e9tude a utilis\u00e9 des m\u00e9thodes de reconnaissance des formes chimiques pour effectuer des analyses en grappes, des ACP et des sch\u00e9mas de classification OPLS-DA sur l'herbe m\u00e9dicinale de Nelumbo nucifera. Les r\u00e9sultats d'analyse des trois m\u00e9thodes \u00e9taient coh\u00e9rents. Quinze lots de Nelumbo nucifera ont \u00e9t\u00e9 divis\u00e9s en quatre cat\u00e9gories \u00e0 273 nm, les diff\u00e9rences se refl\u00e9tant principalement dans les pics 17, 1, 4, 15, 20, 23, 3, 6, 2 et 21. Parmi eux, le pic 1 correspond \u00e0 l'acide gallique, le pic 15 \u00e0 la pu\u00e9rarine et le pic 20 \u00e0 la querc\u00e9tine. \u00c0 350 nm, il est divis\u00e9 en trois cat\u00e9gories, les diff\u00e9rences se refl\u00e9tant principalement dans la s\u00e9lection des pics 14, 1, 13, 7, 8, 11, 10 et 2. Le pic 14 correspond \u00e0 la querc\u00e9tine, le pic 7 au glycoside de feu d'artifice et le pic 8 au glycoside de nuage violet. Ces composants peuvent servir de composants caract\u00e9ristiques principaux pour distinguer et identifier la qualit\u00e9 des mati\u00e8res m\u00e9dicinales de lotus provenant de diff\u00e9rentes zones de production.
