Les m\u00e9tabolites secondaires d\u00e9riv\u00e9s des actinomyc\u00e8tes repr\u00e9sentent environ 45% des m\u00e9tabolites secondaires d\u00e9riv\u00e9s des micro-organismes et pr\u00e9sentent une s\u00e9rie d'activit\u00e9s biologiques, notamment des effets antitumoraux, antibact\u00e9riens, anti-inflammatoires, antiviraux et antiangiog\u00e9niques. Les antibiotiques d\u00e9riv\u00e9s des actinomyc\u00e8tes repr\u00e9sentent 70% des antibiotiques naturels signal\u00e9s, et certains ont \u00e9t\u00e9 largement utilis\u00e9s dans la pratique clinique, comme la vancomycine et l'azithromycine. Les actinobact\u00e9ries ont toujours \u00e9t\u00e9 consid\u00e9r\u00e9es comme une bonne source de m\u00e9tabolites secondaires nouvellement actifs provenant de sources microbiennes.<br \/>\nLes r\u00e9gions polaires comprennent g\u00e9n\u00e9ralement les p\u00f4les Nord et Sud et leurs sous-r\u00e9gions, caract\u00e9ris\u00e9es par des temp\u00e9ratures basses, la lumi\u00e8re du jour polaire et un fort rayonnement ultraviolet. Les micro-organismes polaires ont d\u00e9velopp\u00e9 une r\u00e9gulation g\u00e9n\u00e9tique et des fonctions m\u00e9taboliques uniques pour s'adapter aux conditions polaires extr\u00eames, ce qui leur permet de produire des m\u00e9tabolites secondaires structurellement nouveaux. Ces derni\u00e8res ann\u00e9es, les chercheurs ont d\u00e9couvert de nombreux m\u00e9tabolites secondaires structurellement uniques et hautement actifs chez les actinomyc\u00e8tes polaires. Cet article passe en revue les progr\u00e8s de la recherche sur les m\u00e9tabolites secondaires et les activit\u00e9s pharmacologiques des actinomyc\u00e8tes polaires de 1999 \u00e0 2021. Selon leur type de structure chimique, ils peuvent \u00eatre class\u00e9s en peptides, alcalo\u00efdes, terp\u00e8nes, macrolides, polyc\u00e9tones, macrolactames et autres cat\u00e9gories. Le tableau 1 pr\u00e9sente un r\u00e9sum\u00e9 des nouveaux m\u00e9tabolites secondaires. La plupart des m\u00e9tabolites secondaires des actinomyc\u00e8tes polaires ont \u00e9t\u00e9 isol\u00e9s dans le genre Streptomyces (74,5%), tandis qu'une petite partie a \u00e9t\u00e9 isol\u00e9e dans le genre pseudo Nocardia (14,2%), le genre Bacillus thermophile (2,8%), le genre Nocardia like (0,9%) et d'autres genres (7,7%) (figure 8, \u00e0 gauche). Selon la classification des types de structures chimiques, parmi les 104 m\u00e9tabolites secondaires, 21 (20,2%) sont des compos\u00e9s peptidiques, 9 (8,7%) sont des alcalo\u00efdes, 11 (10,6%) sont des terp\u00e8nes, 15 (14,4%) sont des macrolides, 19 (18,3%) sont des polyc\u00e9tones, 6 (5,8%) sont des macrolactames, et 23 (22,1%) sont d'autres types de compos\u00e9s. Les principaux types structurels sont les peptides, les polyc\u00e9tones, les macrolides, etc. Parmi eux, les m\u00e9tabolites secondaires pr\u00e9sentant des structures tr\u00e8s rares comprennent les nitroporosines A (29) et B (30), l'antartin (41), la g\u00e9phyromycine (41), etc. 60), les saliniquinones G-I (72-74), etc. La plupart des 104 m\u00e9tabolites secondaires ont de bonnes activit\u00e9s pharmacologiques telles que l'antibact\u00e9rien, l'antifongique, l'antiviral et la cytotoxicit\u00e9. Ils se caract\u00e9risent principalement par une activit\u00e9 antitumorale, qui inhibe la leuc\u00e9mie, le cancer du c\u00f4lon, le cancer \u00e9pidermique, le my\u00e9lome, le cancer du col de l'ut\u00e9rus, le cancer du sein, le m\u00e9lanome, le cancer gastrique et d'autres cellules tumorales. Certains compos\u00e9s ont une activit\u00e9 importante, comme le chromophore V C-1027 qui a une cytotoxicit\u00e9 importante, les saliniquinones G \uff5e H qui ont une activit\u00e9 antibact\u00e9rienne importante, et la N - (2-hydroxyph\u00e9ny1) -2-ph\u00e9nazinamine (NHP) qui a une cytotoxicit\u00e9 et un effet antifongique importants. En outre, certains compos\u00e9s ont \u00e9galement un effet inhibiteur s\u00e9lectif. D'autres esp\u00e8ces rares ont de vastes perspectives de d\u00e9veloppement. Les types de structure chimique des m\u00e9tabolites secondaires produits par les actinomyc\u00e8tes polaires sont divers, et nombre d'entre eux ont une bonne activit\u00e9 biologique, en particulier une activit\u00e9 antitumorale, ce qui pr\u00e9sente un grand int\u00e9r\u00eat pour le d\u00e9veloppement de m\u00e9dicaments. Cependant, la recherche sur les voies de biosynth\u00e8se, les m\u00e9canismes actifs et les aspects cliniques des m\u00e9tabolites n'a pas \u00e9t\u00e9 largement men\u00e9e, et la plupart des m\u00e9tabolites secondaires isol\u00e9s des actinomyc\u00e8tes polaires sont des compos\u00e9s connus. Avec le d\u00e9veloppement de la technologie bioinformatique, davantage de s\u00e9quences g\u00e9nomiques d'actinomyc\u00e8tes polaires ont \u00e9t\u00e9 obtenues par s\u00e9quen\u00e7age, et de plus en plus de BGC d'actinomyc\u00e8tes polaires ont \u00e9t\u00e9 d\u00e9couverts et rapport\u00e9s, tels que les g\u00e8nes PKS, NRPS, les g\u00e8nes d'enzymes halog\u00e9n\u00e9es, les g\u00e8nes li\u00e9s aux d\u00e9riv\u00e9s terp\u00e9niques et oligosaccharidiques, etc. On peut constater que les actinomyc\u00e8tes polaires ont un fort potentiel pour synth\u00e9tiser des m\u00e9tabolites secondaires actifs et s'adapter aux environnements polaires, ce qui fournira des sources plus riches et plus diversifi\u00e9es de nouveaux compos\u00e9s pour le d\u00e9veloppement de nouveaux m\u00e9dicaments. En m\u00eame temps, les voies de biosynth\u00e8se des nouveaux m\u00e9tabolites secondaires seront plus faciles \u00e0 d\u00e9chiffrer et m\u00e9ritent de plus amples recherches et d\u00e9veloppements. Bien que la plupart des compos\u00e9s isol\u00e9s \u00e0 partir d'actinomyc\u00e8tes polaires soient actuellement connus, les \"g\u00e8nes silencieux\" responsables de la biosynth\u00e8se de nouveaux compos\u00e9s peuvent ne pas \u00eatre exprim\u00e9s pendant la fermentation.
