Toutefois, en raison de l'utilisation abusive de ce m\u00e9dicament, des souches r\u00e9sistantes sont apparues dans de nombreuses r\u00e9gions. \u00c0 la fin des ann\u00e9es 1980, la Chine a introduit et largement utilis\u00e9 ce type de m\u00e9dicament. Aujourd'hui, les n\u00e9matodes gastro-intestinaux des moutons domestiques ont d\u00e9velopp\u00e9 divers degr\u00e9s de r\u00e9sistance aux m\u00e9dicaments \u00e0 base d'avermectine. Startect \u00ae est un nouveau type de vermifuge oral \u00e0 courte dur\u00e9e d'action pour les moutons, d\u00e9velopp\u00e9 par Zoetis, la plus grande entreprise mondiale de m\u00e9dicaments et de vaccins pour la sant\u00e9 animale, qui peut pr\u00e9venir et r\u00e9soudre efficacement les maladies parasitaires du b\u00e9tail et de la volaille et leurs probl\u00e8mes de r\u00e9sistance aux m\u00e9dicaments. Les ingr\u00e9dients actifs de ce m\u00e9dicament sont le derquantel (10mg\/mL) et l'avermectine (1mg\/mL). La dose standard de Startect \u00ae est de 1mL\/5kg (volume de liquide\/poids corporel du mouton), et chaque mouton adulte de 50kg n\u00e9cessite environ 0,1g de Dexmedetomidine \u00e0 administrer une fois. Selon les statistiques de l'Organisation des Nations unies pour l'alimentation et l'agriculture (FAO), le cheptel ovin mondial s'\u00e9l\u00e8ve \u00e0 1,2 milliard de t\u00eates. Par cons\u00e9quent, la demande potentielle de Dequnt se chiffre en centaines de tonnes, voire plus, et la demande du march\u00e9 est \u00e9norme.<\/p>\n<p>Le dextrant est un d\u00e9riv\u00e9 chimique du m\u00e9tabolite fongique Paraherquamide A (PA), qui est principalement obtenu par r\u00e9duction du carbonyle amide et d\u00e9soxyg\u00e9nation du PA (voir figure 1). Par cons\u00e9quent, l'approvisionnement en mati\u00e8res premi\u00e8res industrielles de la Dequnte est principalement affect\u00e9 par la production de PA. Cependant, la structure chimique des produits naturels tels que le paraherquamide est complexe. Bien qu'ils puissent \u00eatre synth\u00e9tis\u00e9s chimiquement \u00e0 partir de mati\u00e8res premi\u00e8res industrielles telles que l'isopr\u00e8ne ou les \u00e9poxydes d'isopr\u00e8ne, leurs \u00e9tapes de r\u00e9action chimique peuvent atteindre 46 \u00e9tapes, ce qui entra\u00eene des co\u00fbts \u00e9lev\u00e9s et des rendements extr\u00eamement faibles (2,5%), rendant difficile leur production \u00e0 l'\u00e9chelle industrielle. La production de PA par fermentation microbienne est actuellement un moyen efficace d'obtenir ce compos\u00e9 \u00e0 grande \u00e9chelle. L'AP a d'abord \u00e9t\u00e9 isol\u00e9 \u00e0 partir du produit de fermentation d'une souche de Penicillium paraherquei, un champignon appartenant au genre Penicillium. Cependant, la puissance de l'AP produit par le Penicillium de type sauvage n'\u00e9tait pas \u00e9lev\u00e9e, ce qui a entrav\u00e9 la poursuite de la recherche exp\u00e9rimentale et de l'application industrielle. Dans la recherche sur l'application des m\u00e9tabolites fongiques, le niveau d'activit\u00e9 du m\u00e9tabolite cible d\u00e9pend non seulement de la performance de la souche elle-m\u00eame, mais surtout de l'optimisation des conditions de fermentation et de culture afin d'utiliser pleinement sa capacit\u00e9 de production. La m\u00e9thodologie de la surface de r\u00e9ponse est actuellement largement utilis\u00e9e pour optimiser la culture de la fermentation microbienne. Cette m\u00e9thode combine la conception exp\u00e9rimentale et la mod\u00e9lisation math\u00e9matique et pr\u00e9sente les avantages suivants : moins d'exp\u00e9riences, des cycles plus courts, une plus grande pr\u00e9cision dans l'obtention des \u00e9quations de r\u00e9gression, une meilleure performance pr\u00e9dictive et l'\u00e9tude de l'interaction entre plusieurs facteurs.<\/p>\n<p>Lors des travaux pr\u00e9liminaires, notre groupe de recherche a d\u00e9couvert un champignon Penicillium KWF31 d'origine marine produisant beaucoup de PA. L'exploration initiale a montr\u00e9 que sa capacit\u00e9 de fermentation \u00e9tait de 55,8 \u00b1 3,7 mg\/L dans le milieu Chatelier, ce qui ne r\u00e9pondait pas aux exigences de la production industrielle. Il s'agit d'une mati\u00e8re premi\u00e8re pour la synth\u00e8se du Dextrant, un agent commercial r\u00e9sistant aux insectes, qui a une valeur marchande \u00e9lev\u00e9e et une \u00e9norme demande sur le march\u00e9. Cependant, la souche de Penicillium de type sauvage a un faible rendement en PA, avec de multiples \u00e9tapes de synth\u00e8se chimique et un faible rendement, ce qui fait qu'il est difficile de r\u00e9pondre \u00e0 la demande du march\u00e9. Cette \u00e9tude a utilis\u00e9 le plan de Plackett Burman pour s\u00e9lectionner trois composants du milieu qui affectent de mani\u00e8re significative la production de PA par fermentation horizontale de la souche sauvage KWF31 dans des flacons \u00e0 agitation \u00e0 partir de huit facteurs : amidon soluble, glucose et extrait de levure ; leurs concentrations ont \u00e9t\u00e9 progressivement optimis\u00e9es \u00e0 l'aide d'exp\u00e9riences d'escalade et du plan de Box Behnken. Les concentrations optimales sont les suivantes : amidon soluble 31,0 g\/l, glucose 1208 g\/l et extrait de levure 14,5 g\/l. Dans ce milieu, la souche de type sauvage KWF31 a montr\u00e9 une puissance \u00e9lev\u00e9e de 243,7 mg\/L pour la production d'AP pendant la fermentation horizontale en fiole agit\u00e9e, ce qui est 3,4 fois plus \u00e9lev\u00e9 qu'avant l'optimisation (55,8 mg\/L). Le processus \u00e9tabli dans cette \u00e9tude pour la production par fermentation d'AP par le champignon marin KWF31 pr\u00e9sente une bonne reproductibilit\u00e9, une bonne stabilit\u00e9 et une bonne contr\u00f4labilit\u00e9.
