Cependant, avec l'isolement d'un grand nombre de m\u00e9tabolites secondaires, la probabilit\u00e9 de d\u00e9couvrir des antibiotiques ayant une activit\u00e9 biologique significative et des structures nouvelles \u00e0 partir de Streptomyces diminue. Ces derni\u00e8res ann\u00e9es, avec l'application g\u00e9n\u00e9ralis\u00e9e de la technologie de s\u00e9quen\u00e7age du g\u00e9nome dans le s\u00e9quen\u00e7age microbien, une grande quantit\u00e9 d'informations sur le g\u00e9nome de Streptomyces a \u00e9t\u00e9 rendue publique. L'analyse bioinformatique des donn\u00e9es du g\u00e9nome de Streptomyces a r\u00e9v\u00e9l\u00e9 qu'une grande partie des g\u00e8nes sont encore silencieux, ce qui indique qu'un grand nombre de m\u00e9tabolites secondaires potentiels n'ont pas encore \u00e9t\u00e9 d\u00e9couverts. En raison des conditions de culture conventionnelles, les groupes de g\u00e8nes biosynth\u00e9tiques des Streptomyces n'ont pas \u00e9t\u00e9 activ\u00e9s de mani\u00e8re efficace, ce qui fait que leurs voies m\u00e9taboliques uniques sont souvent peu ou pas exprim\u00e9es. Il est donc tr\u00e8s important d'explorer de nouveaux m\u00e9tabolites secondaires actifs en activant le groupe de g\u00e8nes silencieux de Streptomyces.<br \/>\nAfin d'activer l'expression des g\u00e8nes silencieux chez les Streptomyces et de d\u00e9couvrir des m\u00e9tabolites secondaires plus actifs, diverses m\u00e9thodes d'activation des groupes de g\u00e8nes silencieux ont \u00e9t\u00e9 d\u00e9velopp\u00e9es et appliqu\u00e9es jusqu'\u00e0 pr\u00e9sent. Les strat\u00e9gies couramment utilis\u00e9es peuvent \u00eatre divis\u00e9es en deux cat\u00e9gories : la premi\u00e8re est une strat\u00e9gie d'activation non dirig\u00e9e, qui incite Streptomyces \u00e0 produire diff\u00e9rents m\u00e9tabolites en modifiant la composition du milieu de culture (source de carbone, source d'azote, oligo-\u00e9l\u00e9ments, activateurs chimiques), l'environnement de croissance de la souche (temp\u00e9rature, pH, a\u00e9ration, type de r\u00e9cipient) ou la co-culture microbienne ; Un autre type est la strat\u00e9gie d'activation cibl\u00e9e, qui utilise la technologie d'\u00e9dition de g\u00e8nes (CRISPR-Cas9), l'activation cibl\u00e9e de groupes de g\u00e8nes sp\u00e9cifiques (ing\u00e9nierie des promoteurs, activation de facteurs de r\u00e9gulation sp\u00e9cifiques \u00e0 une voie, expression h\u00e9t\u00e9rologue) et d'autres moyens pour permettre l'expression de g\u00e8nes r\u00e9duits au silence chez les Streptomyces. Par rapport aux strat\u00e9gies d'activation cibl\u00e9e, les strat\u00e9gies d'activation non cibl\u00e9e ne n\u00e9cessitent pas la mise en place d'un syst\u00e8me de manipulation g\u00e9n\u00e9tique pour la souche cible, ce qui pr\u00e9sente l'avantage d'un fonctionnement simple et de la possibilit\u00e9 d'activer simultan\u00e9ment plusieurs groupes de g\u00e8nes biosynth\u00e9tiques (plut\u00f4t qu'un groupe de g\u00e8nes biosynth\u00e9tiques sp\u00e9cifique), ce qui les rend plus largement applicables. Cet article donne un bref aper\u00e7u des strat\u00e9gies d'activation non dirig\u00e9e des g\u00e8nes silencieux chez les Streptomyces, telles que la modification de la composition du milieu de culture ou des conditions de culture, les strat\u00e9gies de co-culture, l'ajout d'activateurs chimiques et la r\u00e9gulation globale (voir figure 1), et d\u00e9crit les probl\u00e8mes Actuellement, environ 60% des antibiotiques d\u00e9couverts sont produits par Streptomyces. Cependant, avec l'utilisation g\u00e9n\u00e9ralis\u00e9e des antibiotiques, les probl\u00e8mes de r\u00e9sistance et de superbact\u00e9ries deviennent de plus en plus importants. Pour r\u00e9soudre les probl\u00e8mes de r\u00e9sistance aux m\u00e9dicaments et de superbact\u00e9ries, il est urgent d'explorer de nouveaux antibiotiques. Par cons\u00e9quent, l'activation des groupes de g\u00e8nes silencieux chez Streptomyces est l'une des questions les plus importantes \u00e0 l'heure actuelle. De plus en plus de chercheurs ont remarqu\u00e9 le potentiel des Streptomyces pour d\u00e9couvrir de nouveaux compos\u00e9s et ont commenc\u00e9 \u00e0 \u00e9tudier comment stimuler l'activation des groupes de g\u00e8nes silencieux.<\/p>\n<p>Selon que le g\u00e8ne cible de l'activation est clair ou non, nous divisons les strat\u00e9gies d'activation actuelles en strat\u00e9gies d'activation dirig\u00e9e et en strat\u00e9gies d'activation non dirig\u00e9e. Les strat\u00e9gies d'activation non dirig\u00e9es comprennent les strat\u00e9gies d'activation qui modifient la composition ou les conditions du milieu de culture, les strat\u00e9gies de co-culture, l'ajout d'activateurs chimiques et la r\u00e9gulation globale. Par rapport \u00e0 certaines m\u00e9thodes bas\u00e9es sur les g\u00e8nes, telles que l'ing\u00e9nierie des ribosomes, l'ing\u00e9nierie des promoteurs, la r\u00e9gulation des facteurs de transcription, etc., les strat\u00e9gies d'activation non dirig\u00e9es ont leurs propres avantages et limites (voir tableau 4).
