L'\u00e9dition 2020 de la pharmacop\u00e9e de la R\u00e9publique populaire de Chine inclut les parties a\u00e9riennes s\u00e9ch\u00e9es de trois plantes de la famille des ast\u00e9rac\u00e9es, \u00e0 savoir Siegesbeckia orientalis L., Siegesbeckia pubescens Makino et Siegesbeckia glabrescent Makino, dans la m\u00e9decine traditionnelle chinoise sous le nom de \"Siegesbeckia\". Elle a pour effet de dissiper le vent et l'humidit\u00e9, de favoriser le d\u00e9veloppement des articulations et de d\u00e9toxifier. Elle est utilis\u00e9e pour les rhumatismes et les douleurs, les faiblesses musculaires et osseuses, les douleurs et les faiblesses de la taille et des genoux, la paralysie des membres, l'h\u00e9mipl\u00e9gie, la rub\u00e9ole et les plaies humides. Des exp\u00e9riences pharmacologiques modernes ont montr\u00e9 que l'extrait de Polygonatum sibiricum a des activit\u00e9s anti-inflammatoires, antiallergiques, antithrombotiques et antihistaminiques. Des \u00e9tudes sur la composition chimique et l'activit\u00e9 pharmacologique ont montr\u00e9 que les abondants compos\u00e9s diterp\u00e9no\u00efdes du Polygonatum sibiricum constituent la base mat\u00e9rielle de ses divers effets, notamment en ce qui concerne les aspects anti-inflammatoires, analg\u00e9siques, ainsi que l'activation et l'\u00e9limination de la stase sanguine.<br \/>\nLe darutig\u00e9nol est un compos\u00e9 diterp\u00e9no\u00efde extrait de la plante Scutellaria baicalensis. Des exp\u00e9riences pharmacologiques modernes ont montr\u00e9 que le darutig\u00e9nol peut inhiber de mani\u00e8re significative le taux maximal d'agr\u00e9gation plaquettaire et le taux d'adh\u00e9sion chez les rats, prolonger le temps de dissolution de l'euglobuline et le temps de saignement et de coagulation chez les souris, r\u00e9duire de mani\u00e8re significative la viscosit\u00e9 du sang total et la viscosit\u00e9 du plasma chez les rats mod\u00e8les de stase sanguine, am\u00e9liorer la rh\u00e9ologie sanguine anormale et inhiber la synth\u00e8se de TXB2 chez les rats mod\u00e8les de stase sanguine. Il a des effets antithrombotiques, thrombolytiques, anticoagulants et h\u00e9morh\u00e9ologiques \u00e9vidents. Le facteur de coagulation Xa (FXa) est situ\u00e9 \u00e0 l'intersection des voies de coagulation endog\u00e8ne et exog\u00e8ne et joue un r\u00f4le important dans la voie de coagulation, ce qui en fait l'un des points chauds de la recherche sur les m\u00e9dicaments anticoagulants de ces derni\u00e8res ann\u00e9es. Par cons\u00e9quent, cet article a men\u00e9 des \u00e9tudes de modification structurelle sur le compos\u00e9 actif de l'alcool de Pixiu isol\u00e9 de l'herbe de Pixiu par notre groupe de recherche \u00e0 un stade pr\u00e9coce, et a explor\u00e9 son effet sur le facteur Xa de la coagulation, afin de d\u00e9couvrir des mol\u00e9cules de premier plan ayant une meilleure activit\u00e9. C'est devenu un sujet br\u00fblant dans la recherche sur les m\u00e9dicaments anticoagulants aujourd'hui. Les produits naturels pr\u00e9sentent des avantages uniques en mati\u00e8re d'anticoagulation, et la d\u00e9couverte d'ingr\u00e9dients actifs issus de la m\u00e9decine traditionnelle chinoise avec une efficacit\u00e9 clinique pr\u00e9cise et la conduite de recherches sur les modifications structurelles peuvent constituer un moyen important de d\u00e9couvrir de nouveaux anticoagulants. L'alcool de Pixiu est un compos\u00e9 actif isol\u00e9 \u00e0 partir de l'herbe de Pixiu, un rem\u00e8de traditionnel chinois. Des \u00e9tudes ant\u00e9rieures ont montr\u00e9 qu'il poss\u00e8de une activit\u00e9 anticoagulante significative. Par cons\u00e9quent, cette \u00e9tude a men\u00e9 des recherches sur la modification structurelle de l'alcool de Pixiu et a examin\u00e9 l'activit\u00e9 inhibitrice du FXa, dans le but de d\u00e9couvrir de nouveaux inhibiteurs du FXa pr\u00e9sentant un int\u00e9r\u00eat pour le d\u00e9veloppement.<\/p>\n<p>Sur la base des caract\u00e9ristiques structurelles de la berb\u00e9rine, un anneau h\u00e9t\u00e9rocyclique a \u00e9t\u00e9 introduit dans la structure originale afin d'am\u00e9liorer l'hydrophilie et l'activit\u00e9 de ce type de compos\u00e9. Par cons\u00e9quent, plusieurs d\u00e9riv\u00e9s ont \u00e9t\u00e9 con\u00e7us et synth\u00e9tis\u00e9s en introduisant un anneau h\u00e9t\u00e9rocyclique \u00e0 cinq membres sur l'anneau A de la berb\u00e9rine. En menant des exp\u00e9riences d'inhibition in vitro du FXa sur la berb\u00e9rine et ses d\u00e9riv\u00e9s, sept compos\u00e9s pr\u00e9sentant une bonne activit\u00e9 (IC50&lt;10 \u03bc mol\/L) ont \u00e9t\u00e9 obtenus. Par rapport \u00e0 la berb\u00e9rine, la plupart des d\u00e9riv\u00e9s ont montr\u00e9 une certaine am\u00e9lioration de l&#039;activit\u00e9 inhibitrice du facteur de coagulation FXa, parmi lesquels les compos\u00e9s 9 et 11 \u00e9taient les plus significatifs, avec des valeurs IC50 de 0,71 \u00b1 0,07 et 0,22 \u00b1 0,03 \u03bc mol\/L, respectivement. Des \u00e9tudes pr\u00e9liminaires sur la relation structure-activit\u00e9 ont montr\u00e9 que l&#039;introduction de groupes m\u00e9thyl\u00e8ne sur les 15,16-diols dans la structure originale et l&#039;introduction d&#039;h\u00e9t\u00e9rocycles dans l&#039;anneau A peuvent am\u00e9liorer l&#039;activit\u00e9 des compos\u00e9s, et que l&#039;effet \u00e9lectronique sur les substituants h\u00e9t\u00e9rocycliques joue un r\u00f4le important dans l&#039;am\u00e9lioration de l&#039;activit\u00e9. D&#039;apr\u00e8s l&#039;analyse des mod\u00e8les d&#039;interaction ligand-prot\u00e9ine dans la conception de m\u00e9dicaments assist\u00e9e par ordinateur, le groupe hydroxyle sur l&#039;anneau imidazole du compos\u00e9 9 et le groupe amino sur l&#039;anneau thioph\u00e8ne du compos\u00e9 11 agissent comme des donneurs de liaisons hydrog\u00e8ne pour former des liaisons hydrog\u00e8ne avec le FXa, renfor\u00e7ant ainsi l&#039;affinit\u00e9 entre le compos\u00e9 et le r\u00e9cepteur. C&#039;est peut-\u00eatre la raison de sa forte activit\u00e9. En outre, l&#039;h\u00e9t\u00e9rocycle de soufre dans la structure du compos\u00e9 11 peut contribuer \u00e0 l&#039;am\u00e9lioration de l&#039;activit\u00e9 du compos\u00e9, et une exploration plus pouss\u00e9e est n\u00e9cessaire.
