Il s'agit d'une culture ol\u00e9agineuse importante dans la r\u00e9gion m\u00e9diterran\u00e9enne, et le Gansu et le Yunnan sont les principales zones de plantation d'olives en Chine. Les feuilles d'olivier sont un sous-produit bon march\u00e9 de la culture de l'olivier. Chaque olivier doit tailler environ 25 kg de feuilles par an, et le poids total des feuilles r\u00e9colt\u00e9es repr\u00e9sente 10% du poids total des olives. Les feuilles d'olivier contiennent divers compos\u00e9s bioactifs. Comme l'acide ol\u00e9anolique, l'acide d'aub\u00e9pine, l'acide ursolique, l'oleurop\u00e9ine, l'hydroxytyrosol, etc.<br \/>\nDes recherches ont montr\u00e9 que l'oleurop\u00e9ine est pr\u00e9sente dans toutes les parties de l'olivier, la teneur la plus \u00e9lev\u00e9e se trouvant dans les feuilles d'olivier. Dans les feuilles d'olivier s\u00e8ches, la teneur en oleurop\u00e9ine peut atteindre 10% \u00e0 17%. Comme le montre la figure 1, l'oleurop\u00e9ine s'hydrolyse pour produire de l'hydroxytyrosol, du glucose et de l'acide ol\u00e9nique. Parmi ceux-ci, l'hydroxytyrosol pr\u00e9sente une meilleure activit\u00e9 biologique que l'oleurop\u00e9ine. \u00c0 l'heure actuelle, les m\u00e9thodes d'hydrolyse de l'oleurop\u00e9ine comprennent principalement l'hydrolyse acide, l'hydrolyse alcaline et l'hydrolyse enzymatique. L'hydrolyse acide et l'hydrolyse alcaline n\u00e9cessitent des acides forts, des bases fortes et des environnements \u00e0 haute temp\u00e9rature, avec des conditions de r\u00e9action strictes. Ces derni\u00e8res ann\u00e9es, l'utilisation d'enzymes en tant que catalyseurs pour la biotransformation des sous-produits a suscit\u00e9 l'int\u00e9r\u00eat. Compar\u00e9e aux m\u00e9thodes traditionnelles d'hydrolyse acide-base, l'hydrolyse enzymatique de l'oleurop\u00e9ine pr\u00e9sente l'avantage de conditions de r\u00e9action douces et d'une moindre pollution de l'environnement. Liu et al. ont utilis\u00e9 la cellulase comme additif alimentaire pour rompre les liaisons glycosidiques et hydrolyser l'oleurop\u00e9ine afin d'obtenir de l'hydroxytyrosol. En raison de l'instabilit\u00e9 thermique de la cellulase, celle-ci ne peut pas maintenir une activit\u00e9 \u00e9lev\u00e9e en continu \u00e0 la temp\u00e9rature optimale de l'enzyme, ce qui entra\u00eene une faible efficacit\u00e9 de l'hydrolyse enzymatique. Par cons\u00e9quent, nous essayons de trouver un nouveau solvant vert pour remplacer la solution tampon, qui peut augmenter la solubilit\u00e9 de l'oleurop\u00e9ine et am\u00e9liorer la stabilit\u00e9 de la cellulase dans les r\u00e9actions \u00e0 long terme, am\u00e9liorant ainsi l'efficacit\u00e9 de l'hydrolyse enzymatique de l'oleurop\u00e9ine.<br \/>\nLes solvants eutectiques profonds (DES) sont des m\u00e9langes homog\u00e8nes et transparents form\u00e9s par agitation \u00e0 haute temp\u00e9rature d'accepteurs de liaisons hydrog\u00e8ne (HBA) et de donneurs de liaisons hydrog\u00e8ne (HBD), dont les points de fusion sont inf\u00e9rieurs \u00e0 ceux des composants individuels. En raison de ses excellentes propri\u00e9t\u00e9s physiques et chimiques, notamment sa faible inflammabilit\u00e9, sa faible volatilit\u00e9, sa facilit\u00e9 de pr\u00e9paration, sa grande solubilit\u00e9 et sa grande compatibilit\u00e9 avec les enzymes, le DES est de plus en plus utilis\u00e9 comme agent d'extraction et de solubilisation non toxique. Le plus important est que les enzymes ont tendance \u00e0 se d\u00e9sactiver dans les solvants organiques et que le DES, en tant que solvant, peut emp\u00eacher ce ph\u00e9nom\u00e8ne de se produire. Cela permet \u00e9galement au DES de remplacer les solvants organiques et d'incuber les cellulases. Le DES compos\u00e9 de HBA et de HBD naturels est appel\u00e9 NADES, qui est plus s\u00fbr et plus \u00e9cologique que le DES conventionnel. Le NADES a fait l'objet d'une grande attention en tant que solvant vert dans le domaine des r\u00e9actions catalys\u00e9es par des enzymes. Le NADES a \u00e9t\u00e9 utilis\u00e9 avec succ\u00e8s dans des r\u00e9actions enzymatiques sp\u00e9cifiques, exer\u00e7ant des effets favorables sur l'activit\u00e9 et la stabilit\u00e9 des enzymes. Les r\u00e9sultats des recherches ci-dessus d\u00e9montrent que le NADES peut devenir un bon solvant pour l'hydrolyse de l'oleurop\u00e9ine par la cellulase.<br \/>\nCette \u00e9tude a pr\u00e9par\u00e9 six types de NADES en utilisant du chlorure de choline ou de la b\u00e9ta\u00efne comme HBA et diff\u00e9rents types de polyols comme HBD, et a \u00e9tudi\u00e9 leurs effets sur l'hydrolyse de l'oleurop\u00e9ine par la cellulase. Il a \u00e9t\u00e9 constat\u00e9 que la s\u00e9lection du type et de la concentration appropri\u00e9s de NADES peut augmenter la stabilit\u00e9 de la cellulase et promouvoir l'hydrolyse de l'oleurop\u00e9inase. Dans les conditions d'une temp\u00e9rature de 50 \u2103 et d'un pH=5, le taux d'hydrolyse de l'oleurop\u00e9inase dans un syst\u00e8me de solvants 10% DES-5 (V\/V) \u00e9tait 1,7 fois celui de la solution tampon apr\u00e8s une hydrolyse de 3 heures ; le taux d'hydrolyse enzymatique apr\u00e8s 36 heures d'hydrolyse est 1,5 fois celui de la solution tampon ; sur la base de conditions d'hydrolyse enzymatique douces, efficaces et respectueuses de l'environnement, cette m\u00e9thode permet \u00e0 la cellulase d'hydrolyser efficacement l'oleurop\u00e9ine pendant une longue p\u00e9riode de temps.
