La recherche sur les polysaccharides a toujours \u00e9t\u00e9 un sujet d'actualit\u00e9, en particulier pour les algues marines, qui sont les plus appr\u00e9ci\u00e9es du public en termes de fonctionnalit\u00e9 et de s\u00e9curit\u00e9. Cependant, les recherches sur les polysaccharides des microalgues du d\u00e9sert sont relativement peu nombreuses.<\/p>\n<p>La chlorelle du d\u00e9sert est tr\u00e8s r\u00e9sistante aux sels alcalins et \u00e0 la s\u00e9cheresse, mais l'extraction de ses polysaccharides et la d\u00e9tection de son activit\u00e9 antioxydante ont fait l'objet de relativement peu de recherches. Cet article optimise le processus d'extraction des IPS des microalgues du d\u00e9sert et proc\u00e8de \u00e0 la s\u00e9paration, \u00e0 la purification et \u00e0 la d\u00e9tection de l'activit\u00e9. La m\u00e9thode CCD a \u00e9t\u00e9 utilis\u00e9e pour optimiser le processus d'extraction des IPS, et la plus grande quantit\u00e9 d'extraction d'IPS a \u00e9t\u00e9 obtenue dans les conditions suivantes : temp\u00e9rature de 80\u00b0C, dur\u00e9e d'une heure et rapport solide-liquide (g\/mL) de 1:25. Deux polysaccharides acides, IPS-2 et IPS-3, ont \u00e9t\u00e9 s\u00e9par\u00e9s avec succ\u00e8s par chromatographie ionique, avec une teneur en sulfate de (5,69 \u00b1 0,11)% et (8,46 \u00b1 0,03)%, respectivement. Par ailleurs, les \u00e9chantillons IPS, IPS-2 et IPS-3 sont principalement compos\u00e9s de glucose et de galactose. Les IPS des microalgues du d\u00e9sert ont montr\u00e9 un effet inhibiteur sur les radicaux libres DP-PH avec un IC50 de 0,011mg\/mL, qui est proche de l'IC50 du VC sur les radicaux libres DPPH \u00e0 0,007mg\/mL ; En termes de radicaux hydroxyle, les IPS des microalgues du d\u00e9sert ont montr\u00e9 un effet inhibiteur avec un IC50 de 0,737mg\/mL, qui est \u00e9galement proche de l'IC50 de 0,626mg\/mL du VC sur les radicaux hydroxyle. Cette activit\u00e9 antioxydante extr\u00eamement forte peut \u00eatre li\u00e9e \u00e0 la teneur en monosaccharides (glucose).
