Ici, les auteurs ont con\u00e7u une palette d'agitation \u00e0 paroi racl\u00e9e en forme d'ancre (cristalliseur \u00e0 surface racl\u00e9e) et ont s\u00e9lectionn\u00e9 trois types diff\u00e9rents d'IPA pour \u00e9tudier l'effet de l'agitation sur la cristallisation.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2981\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/1-1.png\" alt=\"\" width=\"473\" height=\"483\" \/><\/p>\n<p>Une vue d'ensemble des techniques de cristallisation, qui nous a ouvert les yeux !<br \/>\nLe premier \u00e9tait l'ibuprof\u00e8ne (IBP), qui a \u00e9t\u00e9 cristallis\u00e9 en ajoutant une gouttelette d'eau \u00e0 une solution d'IBP \u00e0 25 \u00b0C dans du DMSO. Lors de la cristallisation avec l'agitateur axial ST \u00e0 60 tr\/min, l'agitation n'\u00e9tait pas bonne et au bout de 25 minutes, il y avait une phase aqueuse dans la couche sup\u00e9rieure de DMSO, ce qui a donn\u00e9 un rendement \u00e9lev\u00e9 lors de l'\u00e9chantillonnage et de l'analyse au bout de 30 minutes. En continuant d'agiter et d'\u00e9chantillonner, la cristallisation en utilisant l'agitation axiale ST \u00e0 200 rpm et l'agitation par ancrage de racleur de paroi SS \u00e0 60 rpm a donn\u00e9 des rendements similaires \u00e0 30 min et 60 min.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2982\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/2-1.png\" alt=\"\" width=\"467\" height=\"232\" \/><\/p>\n<p>Comment appliquer un processus de production \u00e0 flux continu efficace dans l'industrie biopharmaceutique ? Avec partage de cas !<br \/>\nLa cro\u00fbte dure \u00e0 la surface est li\u00e9e \u00e0 la nature de l'IPA de cristallisation lui-m\u00eame ainsi qu'\u00e0 la douceur et \u00e0 la duret\u00e9 de la surface de la palette d'agitation et de la marmite de cristallisation. Avec le type de palette ST en acier inoxydable \u00e0 200 tr\/min (Fig. c), la cro\u00fbte superficielle \u00e9tait plus importante, mais avec le type d'ancre en acier inoxydable \u00e0 60 tr\/min utilisant du PTFE, la cro\u00fbte superficielle \u00e9tait moins importante.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2983\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/3-1.png\" alt=\"\" width=\"883\" height=\"512\" \/><\/p>\n<p>Le chlorhydrate de diph\u00e9nhydramine (DPH) est un cristal lamellaire qui est pr\u00e9cipit\u00e9 en refroidissant \u00e0 partir de 60 \u00b0C jusqu'\u00e0 5 \u00b0C, puis en le maintenant. La cin\u00e9tique de cristallisation du SS Anchor 60 rpm est similaire \u00e0 celle du ST 200 rpm, et les deux pr\u00e9cipitent plus rapidement que le ST Paddle 60 rpm.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2984\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/4-1.png\" alt=\"\" width=\"499\" height=\"269\" \/><\/p>\n<p>Diff\u00e9rences et liens entre les exp\u00e9riences \u00e0 petite \u00e9chelle et \u00e0 grande \u00e9chelle et la production pilote<\/p>\n<p>Les cristaux obtenus par agitation ST (Fig. b, 60 rpm ; c, 200 rpm) pr\u00e9sentaient davantage de solides grumeleux que ceux obtenus par agitation ST (Fig. a).<\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2985\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/5.png\" alt=\"\" width=\"843\" height=\"246\" \/><\/p>\n<p>La vidange a \u00e9t\u00e9 plus facile avec l'ancre SS 60 tr\/min et la palette ST 200 tr\/min, sans r\u00e9sidus de solides en morceaux, mais la ST 60 tr\/min a produit plus de cro\u00fbtes en surface et de r\u00e9sidus en morceaux lors de la vidange.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2986\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/6.png\" alt=\"\" width=\"825\" height=\"524\" \/><\/p>\n<p>Pour le chlorhydrate de fluox\u00e9tine (FLU) sous forme d'aiguille, \u00e9galement refroidi et d\u00e9cristallis\u00e9, l'ancre SS \u00e0 60 tr\/min a d\u00e9cristallis\u00e9 beaucoup plus rapidement que la palette ST.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2987\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/7.png\" alt=\"\" width=\"510\" height=\"263\" \/><\/p>\n<p>Ces trois conditions de cristallisation ont permis d'obtenir des cristaux d'UFL longs, minces et semblables \u00e0 des aiguilles, le type d'ancre SS produisant des cristaux plus longs (Fig. a) que les autres (Fig. a, ST, 60 rpm ; Fig. c, ST, 200 rpm). (Partage \u00e0 sec) Points de contr\u00f4le du processus de cristallisation industrielle et cas d'application<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2988\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/8.png\" alt=\"\" width=\"849\" height=\"241\" \/><\/p>\n<p>Aucun probl\u00e8me d'\u00e9coulement n'a \u00e9t\u00e9 rencontr\u00e9 avec le SS ancr\u00e9 \u00e0 60 rpm (Fig. a), mais les cristaux obtenus \u00e0 diff\u00e9rentes vitesses avec le ST \u00e9taient peu mobiles et ne pouvaient pas \u00eatre vers\u00e9s directement, mais seulement apr\u00e8s avoir reconstitu\u00e9 le solvant (Fig. b).<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2989\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/9.png\" alt=\"\" width=\"932\" height=\"263\" \/><\/p>\n<p>Comparaison globale :<br \/>\n1. ST Agitation \u00e0 pales 200 tr\/min favorise la rupture des cristaux, l'agglom\u00e9ration et la nucl\u00e9ation secondaire, de sorte que la distribution de la taille des cristaux est plus large.<br \/>\n2. Avec une agitation ST \u00e0 60 rpm, les cristaux de DPH atteignent une certaine taille et commencent \u00e0 pr\u00e9cipiter et \u00e0 s'agglom\u00e9rer au fond de la marmite. En raison d'une mauvaise agitation, la croissance des cristaux est affect\u00e9e par le transfert de masse, ce qui ralentit la pr\u00e9cipitation des cristaux.<br \/>\n3. Agitation par ancre SS \u00e0 60 tr\/min, car l'espace entre l'ancre et la paroi de la bouilloire est plus petit, la suspension solide est meilleure, et la CFD de la simulation des fluides montre que le cisaillement moyen du syst\u00e8me est plus petit. Moins de cristaux cass\u00e9s, comme la cristallisation de l'UFL, que dans d'autres syst\u00e8mes d'agitation avec des cristaux finement cass\u00e9s.
