Las moras verdes, tambi\u00e9n conocidas como moras amarillas, se refieren a moras inmaduras. Como medicina tradicional china, las moras verdes son m\u00e1s f\u00e1ciles de recolectar, transportar y almacenar que las moras maduras, y su contenido en antocianinas, catequinas y epicatequinas es mucho mayor que el de las moras maduras. Se han realizado estudios tanto a nivel nacional como internacional sobre la extracci\u00f3n y el aislamiento de antocianinas de diversas plantas, como semillas de uva, rosas, seda de ma\u00edz, ar\u00e1ndanos, granos de cacao, etc. Liu et al. utilizaron la resina LSA-10 para enriquecer y purificar las antocianinas del espino; Chen Wenliang et al. utilizaron la separaci\u00f3n por membrana y la filtraci\u00f3n para separar las antocianinas oligom\u00e9ricas de las semillas de uva, obteniendo un contenido de 65% antocianinas oligom\u00e9ricas, pero no obtuvieron componentes de separaci\u00f3n monom\u00e9rica; Glavnik et al. analizaron las antocianinas de la ca\u00f1a de tigre japonesa mediante cromatograf\u00eda en capa fina de alto rendimiento e identificaron mon\u00f3meros, dec\u00e1meros y antocianinas de tipo B a partir de su extracto crudo. Sin embargo, este m\u00e9todo es dif\u00edcil de preparar; Sun et al. utilizaron la cromatograf\u00eda l\u00edquida semipreparativa de alto rendimiento para separar las antocianinas de las semillas de uva, pero el pretratamiento era complejo y el rendimiento de la separaci\u00f3n era bajo; este art\u00edculo elige la cromatograf\u00eda en contracorriente de alta velocidad (HSCCC), que tiene una gran capacidad de separaci\u00f3n, puede utilizarse para la preparaci\u00f3n y separaci\u00f3n por lotes, es f\u00e1cil de manejar y tiene una alta eficiencia de separaci\u00f3n.<br \/>\nLas procianidinas (PC), como tipo de compuesto polifen\u00f3lico, est\u00e1n ampliamente presentes en las plantas. Se componen principalmente de catequinas, epicatequinas (EC), epigalocatequinas (EGC) y epicatequina galatos (ECG), y tienen una fuerte actividad antioxidante. El objetivo de este trabajo es utilizar la cromatograf\u00eda en contracorriente de alta velocidad para separar y purificar las antocianinas de las moras verdes. La cromatograf\u00eda l\u00edquida de alto rendimiento (HPLC) se utilizar\u00e1 para identificar los componentes qu\u00edmicos de cada componente y analizar su pureza; mediante el examen de la capacidad de barrido de los radicales DPPH, ABTS + radicales, y los radicales hidroxilo en cada componente, la fuerza de su capacidad antioxidante puede ser reflejada. Este trabajo ampl\u00eda el alcance del procesamiento profundo y la utilizaci\u00f3n de moras, proporciona orientaci\u00f3n para la producci\u00f3n industrial de antocianinas en moras verdes, y tiene cierto valor de referencia para el desarrollo y la utilizaci\u00f3n de moras como recursos medicinales.<\/p>\n<p>\u200d<\/p>\n<p>Utilizando la morera verde como materia prima, se obtuvo el extracto crudo mediante extracci\u00f3n, desengrasado, eliminaci\u00f3n de impurezas y enriquecimiento por extracci\u00f3n en las condiciones experimentales. El sistema de disolventes \u00f3ptimo para la separaci\u00f3n y purificaci\u00f3n de HSCCC fue agua acetato de etilo n-hexano (1:20:20, V\/V\/V). 200 mg de extracto crudo de antocianina de morera verde se separaron en cinco componentes mediante HSCCC, a saber, F1 (28,5 mg), F2 (10,43 mg), F3 (6,85 mg), F4 (10,65 mg) y F5 (13,65 mg). Cada componente conten\u00eda catequinas, antocianina B2, antocianina A1, etc. Entre ellos, F4 conten\u00eda galato de epigalocatequina, con una pureza de 95,05%; F5 contiene catequinas con una pureza de 73,75%. Todos los componentes separados tienen cierta actividad antioxidante. Utilizando Vc como control, la capacidad de barrido de radicales libres DPPH es la siguiente: Vc&gt;F5&gt;F4&gt;F2&gt;F3&gt;F1&gt;extracto crudo de morera verde; capacidad de barrido de radicales libres ABTS+: Vc&gt;F5&gt;F4&gt;F2&gt;F3&gt;F1&gt;extracto crudo de morera verde; Capacidad de eliminaci\u00f3n de radicales hidroxilo: F1&gt;F5&gt;Vc&gt;F4&gt;F2&gt;extracto crudo de morera verde&gt;F3. El HSCCC tiene una gran capacidad de preparaci\u00f3n y no contiene material de relleno en la columna cromatogr\u00e1fica. En este experimento, el extracto crudo se puede separar una vez para obtener cinco componentes, y algunos componentes tienen sustancias mon\u00f3meras de alta pureza y fuerte actividad antioxidante.
