Desde el punto de vista farmacol\u00f3gico, los compuestos polic\u00e9tidos son una fuente importante de nuevos f\u00e1rmacos, utilizados principalmente como antibi\u00f3ticos, inmunosupresores, antiparasitarios, reductores del colesterol y antitumorales. La funci\u00f3n de cada dominio de la sintasa de polic\u00e9tidos se corresponde con la estructura del producto final. Por lo tanto, esta correspondencia puede utilizarse para editar genes biosint\u00e9ticos y dise\u00f1ar productos sint\u00e9ticos, creando as\u00ed un nuevo campo de investigaci\u00f3n: la biolog\u00eda combinatoria de la sintasa de poliquetos. Los estudiosos generan principalmente nuevos productos reduciendo, aumentando o sustituyendo los m\u00f3dulos y dominios estructurales de la sintasa de polioc\u00e9ptidos, como por ejemplo modificando el grupo de genes de la sintasa de polioc\u00e9ptidos aveLAT-ACP en Streptomyces avermitilis para obtener una cepa modificada gen\u00e9ticamente. Se aislaron dos nuevos derivados de la avermectina, la 25 metil ivermectina y la 25 etil ivermectina, a partir de los metabolitos secundarios de la cepa.<\/p>\n<p>En la investigaci\u00f3n anterior, nuestro grupo de investigaci\u00f3n modific\u00f3 el gen milF que codifica la sintasa de polic\u00e9tidos en Streptomyces bingchengensis BC-120-4 para obtener la cepa modificada gen\u00e9ticamente Streptomyces bingchengensis BCJ60. Esta cepa no puede codificar la C-5 cetorreductasa, bloqueando as\u00ed la s\u00edntesis de milbeimicina A3\/A4\/B2\/B3. Mediante la modificaci\u00f3n gen\u00e9tica dirigida, el grupo de genes biosint\u00e9ticos puede sufrir cambios, lo que puede activar ciertas v\u00edas biosint\u00e9ticas para obtener nuevos metabolitos. Por lo tanto, este estudio se centra principalmente en los metabolitos secundarios de la cepa Streptomyces bingchengensis BCJ60 modificada gen\u00e9ticamente, con el fin de obtener antibi\u00f3ticos insecticidas altamente eficaces con valor pr\u00e1ctico.<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>En este art\u00edculo se estudian sistem\u00e1ticamente los metabolitos secundarios de la bacteria de ingenier\u00eda gen\u00e9tica Streptomyces bingchengensis BCJ60, y se obtienen 11 compuestos, entre los cuales los compuestos 1 y 2 son dos compuestos nuevos. En estudios anteriores, nuestro grupo de investigaci\u00f3n ha llevado a cabo investigaciones preliminares sobre los metabolitos secundarios de la bacteria modificada gen\u00e9ticamente Streptomyces bingchengensis BCJ60 y ha obtenido 12 compuestos nuevos, todos ellos milberelinas. Esto indica que mediante la modificaci\u00f3n dirigida de genes, el grupo de genes biosint\u00e9ticos sufre cambios, activando ciertas v\u00edas biosint\u00e9ticas y obteniendo nuevos metabolitos, lo que ayuda a comprender mejor el mecanismo biosint\u00e9tico de los compuestos de Mirella. El experimento de actividad insecticida del nuevo compuesto mostr\u00f3 que los compuestos 1 y 2 presentaban una actividad letal significativa contra los \u00e1caros del cinabrio y los nematodos de la madera del pino, y no hab\u00eda diferencias significativas en comparaci\u00f3n con la Mirbeimicina A3\/A4 comercial.
