En la actualidad, las avernectinas son los f\u00e1rmacos antiparasitarios m\u00e1s utilizados en la pr\u00e1ctica cl\u00ednica veterinaria en todo el mundo. Sin embargo, debido al abuso de este f\u00e1rmaco, en muchas zonas han aparecido cepas farmacorresistentes. A finales de la d\u00e9cada de 1980, China introdujo y utiliz\u00f3 ampliamente este tipo de f\u00e1rmaco, y ahora los nematodos gastrointestinales de las ovejas dom\u00e9sticas han desarrollado diversos grados de resistencia a los medicamentos con avermectina. Startect \u00ae es un nuevo tipo de antihelm\u00edntico oral de acci\u00f3n corta para ovejas desarrollado por Zoetis, la mayor empresa mundial de medicamentos y vacunas para la salud animal, que puede prevenir y resolver eficazmente las enfermedades parasitarias del ganado y las aves de corral y sus problemas de resistencia a los medicamentos. Los ingredientes activos de este medicamento son derquantel (10mg\/mL) y avermectina (1mg\/mL). La dosis est\u00e1ndar de Startect \u00ae es de 1mL\/5kg (volumen l\u00edquido\/peso corporal de la oveja), y cada oveja adulta de 50kg requiere aproximadamente 0,1g de Dexmedetomidina para ser administrada una vez. Seg\u00fan las estad\u00edsticas de la Organizaci\u00f3n de las Naciones Unidas para la Agricultura y la Alimentaci\u00f3n (FAO), el censo mundial de ovejas alcanza los 1.200 millones. Por lo tanto, la demanda potencial de Dequnt es de cientos de toneladas o incluso superior, y la demanda del mercado es enorme.<\/p>\n<p>El dextrante es un derivado qu\u00edmico del metabolito f\u00fangico Paraherquamida A (PA), que se obtiene principalmente mediante la reducci\u00f3n de amida carbonilo y la desoxigenaci\u00f3n de PA (v\u00e9ase la Figura 1). Por lo tanto, el suministro de materias primas industriales de Dequnte se ve afectado principalmente por la producci\u00f3n de PA. Sin embargo, la estructura qu\u00edmica de productos naturales como la paraherquamida es compleja. Aunque pueden sintetizarse qu\u00edmicamente a partir de materias primas industriales como el isopreno o los ep\u00f3xidos de isopreno, sus pasos de reacci\u00f3n qu\u00edmica pueden alcanzar hasta 46 pasos, lo que se traduce en costes elevados y rendimientos extremadamente bajos (2,5%), lo que dificulta su producci\u00f3n a escala industrial. La producci\u00f3n de PA mediante fermentaci\u00f3n microbiana es actualmente una forma eficaz de obtener este compuesto a gran escala. El PA se aisl\u00f3 por primera vez a partir del producto de fermentaci\u00f3n de una cepa de Penicillium paraherquei, un hongo perteneciente al g\u00e9nero Penicillium. Sin embargo, la potencia del PA producido por el Penicillium de tipo salvaje no era alta, lo que dificultaba su posterior investigaci\u00f3n experimental y su aplicaci\u00f3n industrial. En la investigaci\u00f3n de la aplicaci\u00f3n de metabolitos f\u00fangicos, el nivel de potencia del metabolito objetivo depende no s\u00f3lo del rendimiento de la propia cepa, sino, lo que es m\u00e1s importante, de la optimizaci\u00f3n de las condiciones de fermentaci\u00f3n y cultivo para aprovechar al m\u00e1ximo su capacidad de producci\u00f3n. En la actualidad, la metodolog\u00eda de superficie de respuesta se utiliza ampliamente para optimizar los cultivos de fermentaci\u00f3n microbiana. Este m\u00e9todo combina el dise\u00f1o experimental y la modelizaci\u00f3n matem\u00e1tica, y presenta las ventajas de un menor n\u00famero de experimentos, ciclos m\u00e1s cortos, mayor precisi\u00f3n en la obtenci\u00f3n de ecuaciones de regresi\u00f3n, mejor rendimiento predictivo y estudio de la interacci\u00f3n entre varios factores.<\/p>\n<p>En el trabajo preliminar, nuestro grupo de investigaci\u00f3n descubri\u00f3 un hongo Penicillium KWF31 derivado del mar con una elevada producci\u00f3n de PA. La exploraci\u00f3n inicial mostr\u00f3 que su potencia de fermentaci\u00f3n era de 55,8 \u00b1 3,7mg\/L en medio Chatelier, lo que no cumpl\u00eda los requisitos de la producci\u00f3n industrial. Es una materia prima para la s\u00edntesis del agente comercial resistente a los insectos Dextrant y tiene un alto valor comercial y una enorme demanda en el mercado. Sin embargo, la cepa de Penicillium de tipo salvaje tiene un bajo rendimiento de PA, con m\u00faltiples pasos de s\u00edntesis qu\u00edmica y bajo rendimiento, lo que dificulta satisfacer la demanda del mercado. Este estudio utiliz\u00f3 el dise\u00f1o de Plackett Burman para seleccionar tres componentes del medio que afectan significativamente a la producci\u00f3n de PA por fermentaci\u00f3n horizontal de la cepa silvestre KWF31 en matraces agitados a partir de ocho factores: almid\u00f3n soluble, glucosa y extracto de levadura; sus concentraciones se optimizaron gradualmente utilizando experimentos de escalada y el dise\u00f1o Box Behnken. Las concentraciones \u00f3ptimas optimizadas son almid\u00f3n soluble 31.0g\/L, glucosa 1208g\/L, y extracto de levadura 14.5g\/L. En este medio, la cepa de tipo salvaje KWF31 mostr\u00f3 una alta potencia de 243,7 mg\/L para la producci\u00f3n de PA durante la fermentaci\u00f3n horizontal en matraz agitado, que fue 3,4 veces mayor que antes de la optimizaci\u00f3n (55,8 mg\/L). El proceso establecido en este estudio para la producci\u00f3n por fermentaci\u00f3n de PA por el hongo marino KWF31 tiene buena reproducibilidad, estabilidad y controlabilidad.
