La fibrina es el principal componente de los co\u00e1gulos sangu\u00edneos, por lo que promover su degradaci\u00f3n es un medio habitual de prevenir y tratar las enfermedades tromb\u00f3ticas. Las enzimas fibrinol\u00edticas ex\u00f3genas, como la uroquinasa (UK) y la estreptoquinasa (SK), se utilizan ampliamente en el tratamiento cl\u00ednico de las enfermedades tromboemb\u00f3licas debido a su capacidad para hidrolizar la fibrina de forma indirecta o directa. Aunque tienen una importante eficacia trombol\u00edtica, tambi\u00e9n presentan ciertas limitaciones de aplicaci\u00f3n, como una vida media corta y efectos secundarios significativos. Por ello, la b\u00fasqueda de nuevas enzimas fibrinol\u00edticas con gran especificidad, buena seguridad y escasos efectos secundarios se ha convertido en uno de los principales temas de investigaci\u00f3n.<\/p>\n<p>Los hongos comestibles son ricos en prote\u00ednas, polisac\u00e1ridos, terpenos, amino\u00e1cidos, esteroles, nucle\u00f3sidos y otras sustancias, que tienen diversas funciones fisiol\u00f3gicas como antioxidante, antitumoral, regulaci\u00f3n inmunitaria y regulaci\u00f3n neuronal. En la actualidad, la investigaci\u00f3n sobre las enzimas fibrinol\u00edticas derivadas de hongos comestibles y sus propiedades funcionales ha recibido una amplia atenci\u00f3n. Se ha informado de que se han aislado y extra\u00eddo enzimas fibrinol\u00edticas de varios hongos comestibles, como Lentinus edodes, Cordyceps Militaris, Agrocybe aegerita, Tricholoma sejunctum, Lepista nuda, Pleurotus ferulae, etc.<\/p>\n<p>La seta de cuerno de ciervo es un hongo comestible y medicinal que se descompone en la madera y est\u00e1 ampliamente distribuido en las regiones templadas del hemisferio norte. Tiene funciones fisiol\u00f3gicas como antioxidante, mejora del sistema inmunitario, supresi\u00f3n de tumores y reducci\u00f3n de l\u00edpidos sangu\u00edneos. Actualmente, nuestro grupo de investigaci\u00f3n no ha visto ning\u00fan otro informe relacionado con la enzima fibrinol\u00edtica en el hongo cuerno de terciopelo. La secuencia N-terminal de doce amino\u00e1cidos de la enzima fibrinol\u00edtica es Gly Ala Val Thr Gln Asn Ala Pro Trp Gly Leu. Tras compararla con la base de datos NCBI, la enzima fibrinol\u00edtica se identific\u00f3 como una nueva enzima fibrinol\u00edtica. La actividad espec\u00edfica de la enzima fibrinol\u00edtica es de 4105,78U\/mg, el factor de purificaci\u00f3n es de 206,09 y la tasa de recuperaci\u00f3n de la actividad es de 30,91%. Mediante electroforesis Native PAGE y detecci\u00f3n por electroforesis SDS-PAGE, se descubri\u00f3 que el peso molecular relativo de la enzima fibrinol\u00edtica era de 30,9 kDa.
