Aqu\u00ed los autores dise\u00f1aron una paleta de agitaci\u00f3n de pared raspada tipo ancla (cristalizador de superficie raspada) y seleccionaron tres tipos diferentes de APIs para estudiar el efecto de la agitaci\u00f3n en la cristalizaci\u00f3n.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2981\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/1-1.png\" alt=\"\" width=\"473\" height=\"483\" \/><\/p>\n<p>Una mirada exhaustiva a las t\u00e9cnicas de cristalizaci\u00f3n, \u00a1esta s\u00ed que me abri\u00f3 los ojos!<br \/>\nEl primero fue el ibuprofeno (IBP), que se cristaliz\u00f3 a\u00f1adiendo una gota de agua a una soluci\u00f3n a 25 \u00b0C de IBP en DMSO. Al cristalizar con agitaci\u00f3n axial de paletas ST a 60 rpm, la agitaci\u00f3n no fue buena y a los 25 minutos hab\u00eda una fase acuosa en la capa superior de DMSO, lo que dio lugar a un alto rendimiento cuando se tomaron muestras y se analizaron a los 30 minutos. Continuando con la agitaci\u00f3n y el muestreo, la cristalizaci\u00f3n utilizando agitaci\u00f3n de flujo axial ST a 200 rpm y agitaci\u00f3n de anclaje de rascador de pared SS a 60 rpm dio como resultado rendimientos similares a los 30 min y a los 60 min.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2982\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/2-1.png\" alt=\"\" width=\"467\" height=\"232\" \/><\/p>\n<p>\u00bfC\u00f3mo aplicar un proceso de producci\u00f3n de flujo continuo eficiente en el sector biofarmac\u00e9utico? \u00a1Compartiendo casos!<br \/>\nLa costra dura en la superficie est\u00e1 relacionada con la naturaleza de la propia API de cristalizaci\u00f3n, as\u00ed como con la suavidad y dureza de la superficie de la paleta agitadora y del hervidor de cristalizaci\u00f3n. Con el tipo de paleta ST de acero inoxidable a 200 rpm (Fig. c), se produjo una costra superficial m\u00e1s severa, pero con el tipo de anclaje SS a 60 rpm utilizando PTFE, se produjo menos costra superficial.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2983\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/3-1.png\" alt=\"\" width=\"883\" height=\"512\" \/><\/p>\n<p>El clorhidrato de difenhidramina (DPH) es un cristal laminar que se precipita enfriando a partir de 60 \u00b0C hasta 5 \u00b0C y manteni\u00e9ndolo a continuaci\u00f3n.La cin\u00e9tica de cristalizaci\u00f3n del SS Anchor 60 rpm es similar a la del ST 200 rpm, y ambos precipitan m\u00e1s r\u00e1pidamente que el ST Paddle 60 rpm.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2984\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/4-1.png\" alt=\"\" width=\"499\" height=\"269\" \/><\/p>\n<p>Diferencias y conexiones entre los experimentos a peque\u00f1a y gran escala y la producci\u00f3n piloto<\/p>\n<p>Hab\u00eda m\u00e1s s\u00f3lidos grumosos en los cristales obtenidos utilizando agitaci\u00f3n de paletas ST (Fig. b, 60 rpm; c, 200 rpm) (Fig. a).<\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2985\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/5.png\" alt=\"\" width=\"843\" height=\"246\" \/><\/p>\n<p>La descarga fue m\u00e1s f\u00e1cil tanto con el SS Anchor 60rpm como con el ST Paddle 200rpm sin s\u00f3lidos en trozos residuales, pero el ST 60rpm ten\u00eda m\u00e1s costras en la superficie y residuos en trozos en la descarga.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2986\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/6.png\" alt=\"\" width=\"825\" height=\"524\" \/><\/p>\n<p>Para el clorhidrato de fluoxetina (FLU) en forma de aguja, tambi\u00e9n enfriado y descristalizado, la SS Anchor 60rpm descristaliz\u00f3 significativamente m\u00e1s r\u00e1pido que la ST Paddle.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2987\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/7.png\" alt=\"\" width=\"510\" height=\"263\" \/><\/p>\n<p>Utilizando estas tres condiciones de cristalizaci\u00f3n, se obtuvieron cristales de FLU largos, finos y en forma de aguja, y el tipo de anclaje SS produjo cristales m\u00e1s largos (Fig. a) que los dem\u00e1s (Fig. a, ST, 60 rpm; Fig. c, ST, 200 rpm). (Compartici\u00f3n en seco) Puntos de control del proceso de cristalizaci\u00f3n industrial y casos de aplicaci\u00f3n<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2988\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/8.png\" alt=\"\" width=\"849\" height=\"241\" \/><\/p>\n<p>No se encontraron problemas de vertido utilizando el SS anclado a 60 rpm (Fig. a), pero los cristales obtenidos a diferentes velocidades utilizando el ST eran poco m\u00f3viles y no pod\u00edan verterse directamente, sino s\u00f3lo despu\u00e9s de reponer el disolvente (Fig. b).<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-2989\" src=\"http:\/\/longchangextracts.com\/wp-content\/uploads\/2024\/03\/9.png\" alt=\"\" width=\"932\" height=\"263\" \/><\/p>\n<p>Comparaci\u00f3n exhaustiva:<br \/>\n1. La agitaci\u00f3n de paletas ST a 200 rpm favorece la rotura de cristales, la aglomeraci\u00f3n y la nucleaci\u00f3n secundaria, por lo que la distribuci\u00f3n del tama\u00f1o de los cristales es m\u00e1s amplia.<br \/>\n2. ST agitaci\u00f3n de paletas a 60 rpm, los cristales de DPH crecen hasta un cierto tama\u00f1o y empiezan a precipitar y aglomerarse en el fondo de la caldera. Debido a la escasa agitaci\u00f3n, el crecimiento de los cristales se ve afectado por la transferencia de masa, lo que provoca una precipitaci\u00f3n m\u00e1s lenta de los cristales.<br \/>\n3. SS ancla agitaci\u00f3n 60 rpm, porque la brecha entre el ancla y la pared de la caldera es menor, la suspensi\u00f3n s\u00f3lida es mejor, y el CFD de simulaci\u00f3n de fluidos muestra que el cizallamiento medio del sistema es menor. Menor rotura de cristales, como la cristalizaci\u00f3n de FLU, que en otros sistemas de agitaci\u00f3n con cristales finamente rotos.
