Gegenw\u00e4rtig sind Avernectine die in der klinischen Veterin\u00e4rpraxis weltweit am h\u00e4ufigsten verwendeten antiparasit\u00e4ren Arzneimittel. Aufgrund des Missbrauchs dieses Medikaments sind jedoch in vielen Gebieten arzneimittelresistente St\u00e4mme aufgetreten. In den sp\u00e4ten 1980er Jahren f\u00fchrte China diese Art von Medikamenten ein und setzte sie in gro\u00dfem Umfang ein, und inzwischen haben die Magen-Darm-Nematoden von Hausschafen einen unterschiedlichen Grad an Resistenz gegen Avermectin-Medikamente entwickelt. Startect \u00ae ist ein neuartiges orales Kurzzeit-Entwurmungsmittel f\u00fcr Schafe, das von Zoetis, dem weltweit gr\u00f6\u00dften Unternehmen f\u00fcr Tierarzneimittel und -impfstoffe, entwickelt wurde und mit dem sich parasit\u00e4re Vieh- und Gefl\u00fcgelkrankheiten und deren Resistenzprobleme wirksam verhindern und l\u00f6sen lassen. Die Wirkstoffe dieses Medikaments sind Derquantel (10mg\/mL) und Avermectin (1mg\/mL). Die Standarddosis von Startect \u00ae betr\u00e4gt 1mL\/5kg (Fl\u00fcssigkeitsvolumen\/Schafsk\u00f6rpergewicht), und f\u00fcr ein erwachsenes Schaf von 50kg werden etwa 0,1g Dexmedetomidin einmalig verabreicht. Nach den Statistiken der Ern\u00e4hrungs- und Landwirtschaftsorganisation der Vereinten Nationen (FAO) bel\u00e4uft sich der weltweite Schafbestand auf 1,2 Milliarden Tiere. Der potenzielle Bedarf an Dequnt geht daher in die Hunderte von Tonnen oder sogar dar\u00fcber hinaus, und die Marktnachfrage ist enorm.<\/p>\n<p>Dextrant ist ein chemisches Derivat des Pilzmetaboliten Paraherquamid A (PA), das haupts\u00e4chlich durch Amidcarbonylreduktion und Desoxygenierung von PA gewonnen wird (siehe Abbildung 1). Daher wird die industrielle Rohstoffversorgung von Dequnte haupts\u00e4chlich durch die PA-Produktion beeinflusst. Die chemische Struktur von Naturprodukten wie Paraherquamid ist jedoch komplex. Sie k\u00f6nnen zwar aus industriellen Rohstoffen wie Isopren oder Isopren-Epoxiden chemisch synthetisiert werden, aber ihre chemischen Reaktionsschritte k\u00f6nnen bis zu 46 Schritte umfassen, was zu hohen Kosten und extrem niedrigen Ausbeuten (2,5%) f\u00fchrt und die Herstellung im industriellen Ma\u00dfstab erschwert. Die Produktion von PA durch mikrobielle Fermentation ist derzeit ein effektiver Weg, um diese Verbindung in gro\u00dfem Ma\u00dfstab zu gewinnen. PA wurde zun\u00e4chst aus dem Fermentationsprodukt eines Stammes von Penicillium paraherquei isoliert, einem Pilz der Gattung Penicillium. Allerdings war die Potenz von PA, das vom Wildtyp-Penicillium produziert wurde, nicht hoch, was seine weitere experimentelle Forschung und industrielle Anwendung behinderte. Bei der Anwendungsforschung von Pilzmetaboliten h\u00e4ngt die Potenz des Zielmetaboliten nicht nur von der Leistung des Stammes selbst ab, sondern vor allem von der Optimierung der Fermentations- und Kultivierungsbedingungen, um die Produktionskapazit\u00e4t voll auszusch\u00f6pfen. Die Response-Surface-Methode wird derzeit h\u00e4ufig zur Optimierung mikrobieller Fermentationskulturen eingesetzt. Diese Methode kombiniert Versuchsplanung und mathematische Modellierung und hat die Vorteile, dass weniger Experimente durchgef\u00fchrt werden m\u00fcssen, die Zyklen k\u00fcrzer sind, die Genauigkeit der Regressionsgleichungen h\u00f6her ist, die Vorhersagekraft besser ist und die Wechselwirkung zwischen mehreren Faktoren untersucht werden kann.<\/p>\n<p>Im Rahmen der Vorarbeiten entdeckte unsere Forschungsgruppe einen aus dem Meer stammenden Penicillium-Pilz KWF31 mit hoher PA-Produktion. Die erste Untersuchung zeigte, dass seine Fermentationsleistung 55,8 \u00b1 3,7mg\/L unter dem Chatelier-Medium betrug, was nicht den Anforderungen der industriellen Produktion entsprach. Es ist ein Rohstoff f\u00fcr die Synthese des kommerziellen Insektenschutzmittels Dextrant und hat einen hohen Marktwert und eine gro\u00dfe Marktnachfrage. Der Wildtyp-Penicillium-Stamm liefert jedoch nur eine geringe Ausbeute an PA, da mehrere chemische Syntheseschritte erforderlich sind und die Ausbeute gering ist, was es schwierig macht, die Marktnachfrage zu decken. In dieser Studie wurden mit Hilfe des Plackett-Burman-Designs aus acht Faktoren (l\u00f6sliche St\u00e4rke, Glukose und Hefeextrakt) drei Mediumskomponenten herausgefiltert, die einen signifikanten Einfluss auf die horizontale Fermentationsproduktion von PA durch den Wildstamm KWF31 in Sch\u00fcttelkolben haben; ihre Konzentrationen wurden schrittweise mit Hilfe von Kletterexperimenten und Box-Behnken-Design optimiert. Die optimierten optimalen Konzentrationen sind l\u00f6sliche St\u00e4rke 31,0 g\/L, Glukose 1208 g\/L und Hefeextrakt 14,5 g\/L. In diesem Medium zeigte der Wildtyp-Stamm KWF31 eine hohe Potenz von 243,7 mg\/L f\u00fcr die PA-Produktion w\u00e4hrend der horizontalen Fermentation im Sch\u00fcttelkolben, was 3,4-mal h\u00f6her war als vor der Optimierung (55,8 mg\/L). Der in dieser Studie etablierte Prozess f\u00fcr die Fermentationsproduktion von PA durch den marinen Pilz KWF31 weist eine gute Reproduzierbarkeit, Stabilit\u00e4t und Kontrollierbarkeit auf.
