Fibrin ist der Hauptbestandteil von Blutgerinnseln, daher ist die F\u00f6rderung des Fibrinabbaus ein g\u00e4ngiges Mittel zur Vorbeugung und Behandlung thrombotischer Erkrankungen. Exogene fibrinolytische Enzyme wie Urokinase (UK) und Streptokinase (SK) werden in der klinischen Behandlung thromboembolischer Erkrankungen h\u00e4ufig eingesetzt, da sie Fibrin indirekt oder direkt hydrolysieren k\u00f6nnen. Obwohl sie eine betr\u00e4chtliche thrombolytische Wirksamkeit aufweisen, haben sie auch gewisse Anwendungsbeschr\u00e4nkungen wie eine kurze Halbwertszeit und erhebliche Nebenwirkungen. Daher ist die Suche nach neuen fibrinolytischen Enzymen mit hoher Spezifit\u00e4t, guter Sicherheit und geringen Nebenwirkungen zu einem aktuellen Forschungsschwerpunkt geworden.<\/p>\n<p>Speisepilze sind reich an Proteinen, Polysacchariden, Terpenen, Aminos\u00e4uren, Sterolen, Nukleosiden und anderen Stoffen, die verschiedene physiologische Funktionen haben, wie z. B. antioxidative, antitumorale, immunregulatorische und neuronale Funktionen. Gegenw\u00e4rtig wird der Erforschung von fibrinolytischen Enzymen aus Speisepilzen und ihren funktionellen Eigenschaften gro\u00dfe Aufmerksamkeit geschenkt. Es wurde berichtet, dass fibrinolytische Enzyme aus verschiedenen Speisepilzen wie Lentinus edodes, Cordyceps militaris, Agrocybe aegerita, Tricholoma sejunctum, Lepista nuda, Pleurotus ferulae usw. isoliert und extrahiert worden sind.<\/p>\n<p>Der Hirschgeweihpilz ist ein holzverrottender, essbarer und medizinischer Pilz, der in den gem\u00e4\u00dfigten Regionen der n\u00f6rdlichen Hemisph\u00e4re weit verbreitet ist. Er hat physiologische Funktionen wie Antioxidantien, St\u00e4rkung des Immunsystems, Tumorbek\u00e4mpfung und Senkung der Blutfette. In vorl\u00e4ufigen Experimenten wurde festgestellt, dass der Extrakt aus dem Fruchtk\u00f6rper des Samtgeweihpilzes eine hohe Plasminaktivit\u00e4t aufweist, die deutlich h\u00f6her ist als die von anderen Speisepilzen gemeldete Plasminaktivit\u00e4t. In diesem Artikel wird die Methode zur Abtrennung und Reinigung des fibrinolytischen Enzyms aus dem Fruchtk\u00f6rper des Samtfu\u00dfr\u00fcblings vorgestellt und eine vorl\u00e4ufige Identifizierung des Enzyms vorgenommen. Unsere Forschungsgruppe hat bisher keine anderen Berichte \u00fcber fibrinolytische Enzyme in Samtgeweihpilzen gesehen. Die N-terminale zw\u00f6lf Aminos\u00e4uresequenz des fibrinolytischen Enzyms ist Gly Ala Val Thr Gln Asn Ala Pro Trp Gly Leu. Nach einem Vergleich mit der NCBI-Datenbank wurde das fibrinolytische Enzym als ein neuartiges fibrinolytisches Enzym identifiziert. Die spezifische Aktivit\u00e4t des fibrinolytischen Enzyms betr\u00e4gt 4105,78U\/mg, der Reinigungsfaktor 206,09 und die Aktivit\u00e4tsr\u00fcckgewinnungsrate 30,91%. Durch Native PAGE-Elektrophorese und SDS-PAGE-Elektrophorese wurde festgestellt, dass das relative Molekulargewicht des fibrinolytischen Enzyms 30,9 kDa betr\u00e4gt.
