A new diterpenoid compound of iridoid in the flower buds of Daphne genkwa
Daphne Linn. is a deciduous shrub belonging to the Thymelaeaceae family and the genus Daphne Sieb The dried flower buds of et Zucc., also known as medicinal fish grass, mouse flower, playful fish flower, headache flower, dull head flower, Sichuan mulberry, and fish poison, are mainly distributed in various provinces of the Yangtze River Basin and some areas of the Yellow River Basin in China. The Chinese Pharmacopoeia records that yuanhua has a bitter and pungent taste, and a warm nature; Toxic, used for swelling and fullness, chest and abdominal hydrosalpinx, accumulation of phlegm, gas reflux, wheezing and cough, and adverse bowel movements. Topical treatment for scabies, alopecia, and frostbite. Modern chemical composition and pharmacological activity studies have shown that Daphne genkwa contains rich chemical structures such as flavonoids, coumarins, lignin, diterpenes, sterols, etc. It has a wide range of physiological activities such as anti-tumor, anti-inflammatory, analgesic, anticonvulsant, uterine excitation, anti fertility, and immune regulation. However, there are relatively few studies and reports on the topical treatment of skin diseases such as scabies, alopecia, and frostbite. Given that traditional Chinese medicine flower buds may have certain resistance to human skin pathogenic fungi, the author studied the chemical composition of the 95% ethanol extract of flower buds in order to isolate and identify natural lead molecules with anti skin pathogenic fungal activity, providing basic data for the development and utilization of anti skin pathogenic drugs for traditional Chinese medicine flower buds.

Skin fungal diseases are a global infectious disease, and the humid and hot regions in southern China such as Guangdong, Hainan, Fujian, and Hong Kong are high-risk areas for skin fungal diseases. China has abundant resources of traditional Chinese medicine, and it is necessary to search for and develop new antifungal active substances or drugs from Chinese medicinal materials. This study used chromatographic separation techniques and natural product structure identification methods to isolate and identify two diterpenoid compounds of the naringenin type from the flower buds of Daphne genkwa, which have the ability to treat scabies and alopecia. One of them is a new compound; The inhibitory activity of the compound against Microsporum canis, Trichophyton rubrum, Trichophyton rubrum, and Malassezia was determined using the MIC method. It was found that compounds 1 and 2 exhibited strong inhibitory activity against Trichophyton rubrum and Trichophyton rubrum. This article further enriches the chemical composition of yuanhua flower buds and discovers their in vitro antifungal activity, accumulating basic data for the external application and development of yuanhua flower buds.
