Derfor f\u00f8ler jeg behov for at lave en lille analyse af de tre substraters rolle.<\/p>\n<p>For at forst\u00e5 den rolle, som flere typer amylaser spiller, skal man f\u00f8rst identificere to grupper af begreber: stivelse med lige k\u00e6de og stivelse med forgrenet k\u00e6de, \u03b1-1,4-glykosidisk binding og \u03b1-1,6-glykosidisk binding.<br \/>\nDer findes to typer stivelse: stivelse med lige k\u00e6de og stivelse med forgrenet k\u00e6de. Ligek\u00e6det amylopektin er en uforgrenet spiralformet struktur, der best\u00e5r af glukoserester, som kun er forbundet med \u03b1-1,4-glykosidiske bindinger; forgrenet amylopektin er dannet af glukoserester, der er forbundet med \u03b1-1,4-glykosidiske bindinger i hver k\u00e6de, men \u03b1-1,6-glykosidiske bindinger er til stede ved forgreningspunkterne. \u03b1-Amylase og dets virkning<br \/>\n\u03b1- Amylase, ogs\u00e5 kendt som endo amylase, er en metallohydrolase, der kr\u00e6ver calciumioner, som binder sig til enzymproteinet for at udvise aktivitet, hvilket kan g\u00e5 tabt ved behandling med chelateringsmidlet EDTA.\u03b1- Amylase er en st\u00f8kiometrisk amylase, der er i stand til at randomisere sin aktivitet.<br \/>\n\u03b1- Amylase kan tilf\u00e6ldigt hydrolysere den \u03b1-1,4-glykosidiske binding i stivelse, men kan ikke hydrolysere den \u03b1-1,6-glykosidiske binding.<br \/>\nHvis substratet er stivelse med lige k\u00e6de, producerer hydrolysen glukose, maltose og maltotriose; hvis det fungerende substrat er forgrenet stivelse, er hydrolyseprodukterne glukose, maltose, maltotriose og \u03b1-dextrin, der indeholder \u03b1-1,6-glykosidiske bindinger, der indeholder mere end tre glukoserester. I n\u00e6rv\u00e6r af afgreningsenzymer hydrolyseres \u03b1-1,6-glykosidbindingen i \u03b1-dextrin- og \u03b2-dextrin-arterne, og de forgrenede k\u00e6der fjernes.<br \/>\nDe resterende lige k\u00e6der hydrolyseres derefter af \u03b1-amylase og \u03b2-amylase for at producere maltose og glukose. R-amylaser kan dog ikke direkte hydrolysere de \u03b1-1,6-glykosidiske bindinger i forgrenet stivelse.<br \/>\nGlykosylerende amylase og dens virkning<br \/>\nGlykosylerende amylase eller glucoamylase er et enzym, der hydrolyserer stivelse til glukose, hvor hydrolyseprocenten kan n\u00e5 op p\u00e5 100%, og bruges ofte som et forsukringsmiddel til stivelse.<br \/>\nGlucoamylase er ikke s\u00e6rlig specifik og kan anvendes p\u00e5 mange stoffer, den kan ikke kun klippe \u03b1-1,4-glykosidbindingen fra den ikke-reducerende ende af stivelsesmolekylet, men ogs\u00e5 klippe \u03b1-1,6-glykosidbindingen, bare at hydrolysen af \u03b1-1,4-glykosidbindingen er lidt hurtigere. Den kan ogs\u00e5 hydrolysere dextrin, maltose og glykogen.
