Den 22. juni 2020 var der 85018 bekr\u00e6ftede tilf\u00e6lde af den nye coronavirus 2019 (COVID-19) i Kina, herunder 4646 d\u00f8dsfald. Efterf\u00f8lgende blev der rapporteret om over 8,97 millioner tilf\u00e6lde af infektion i mere end 200 lande verden over, herunder USA, Italien, Spanien, Storbritannien, Frankrig, Tyskland, Iran, Schweiz, Sydkorea og Japan. De almindelige symptomer hos patienter, der er smittet med SARS-CoV-2, er feber, hoste og muskelsmerter eller tr\u00e6thed. S-proteinet i SARS-CoV-2 ligner strukturelt det i SARS-CoV, og begge har betydelig bindingsaffinitet til angiotensinkonverterende enzym 2 (ACE2) p\u00e5 overfladen af humane celler, som kommer ind i den menneskelige krop gennem ACE2-m\u00e6gling. ACE2 er bredt udtrykt i v\u00e6v og celler som lunger, ford\u00f8jelseskanalen og nyrerne og menes i \u00f8jeblikket at v\u00e6re n\u00f8glen til, at SARS-CoV-2 kan komme ind i v\u00e6rtsceller. Klinisk behandling har vist, at traditionel kinesisk medicin kombineret med vestlig medicin har betydelige terapeutiske virkninger p\u00e5 COVID-19. Indtil videre er der stadig mangel p\u00e5 specifikke l\u00e6gemidler til behandling af COVID-19. Derfor er der et presserende behov for at finde effektive antivirale l\u00e6gemidler til at bremse denne sygdom.<br \/>\nParis polyphylla-planten i Liliaceae-familien, ogs\u00e5 kendt som loppefr\u00f8ken, gr\u00e6sflodvognen og den hvide s\u00f8de lilje, har en rod og st\u00e6ngel, der kaldes Paris polyphylla. Chonglou, som er en traditionel kinesisk medicin med en lang historie af medicinsk brug, har antiinflammatoriske, h\u00e6mostatiske, bakteriedr\u00e6bende, beroligende, smertestillende, antiastmatiske og hostelindrende virkninger. Det bruges ogs\u00e5 i folkemedicinen til at behandle forskellige typer s\u00e5r, karbunkler, giftige slangebid osv. Siden 1970'erne er forskningen i aktive ingredienser i tunge bygninger fortsat. P\u00e5 nuv\u00e6rende tidspunkt er der opdaget over hundrede kemiske komponenter i de tunge bygninger, hvoraf mere end 70 er steroide saponiner. Og de steroide saponiner med h\u00f8jere indhold i Chonglou omfatter dioscin og saponiner af spirostanol. Polyphyllin I, Polyphyllin VI og Polyphyllin VII h\u00f8rer alle til kategorien dioscin (figur 1), som findes i rigelige m\u00e6ngder i Polygonatum sibiricum og bruges i vid udstr\u00e6kning til behandling af forskellige sygdomme, herunder antitumor, immunregulering, antibakterielle virkninger osv. Zhonglou saponiner er spiral steroid saponiner; Saponiner er sammensat af aglyconer og sukkerarter, glucuronsyrer eller andre organiske syrer. Det indeholder en f\u00e6lles moderkerne i sin struktur (figur 1), som best\u00e5r af seks ringe og 27 kulstofatomer, hvor A, B, C og D er steroide moderkerner (cyclopentane dihydrophenanthrene). Steroidkernen er forbundet med en furanring E og en pyranring F. Og den er ofte forbundet i form af spiroketid. I denne artikel blev molekyl\u00e6r docking-metode brugt til at udforske de aktive forbindelser i kinesisk urtemedicin Yizhihua Aesculus til behandling af det nye coronavirus SARS-CoV-2. SARS-CoV-2 binder sig hovedsageligt til angiotensin-konverterende enzym 2 (ACE2)-receptoren p\u00e5 overfladen af humane celler gennem sit S-protein. Denne unders\u00f8gelse forudsagde bindingsaffiniteten for tre typer saponiner (I, VI, VII), der er rige p\u00e5 Polygonatum sibiricum, med ACE2 gennem molekyl\u00e6r docking-simulering.<\/p>\n<p>Da SARS-CoV-2 er en nyopdaget virus, er der i \u00f8jeblikket ingen bekr\u00e6ftet eller tilg\u00e6ngelig specifik medicin. Den \u00f8konomisk effektive behandlingsstrategi er at bruge eksisterende l\u00e6gemidler. Klinisk behandling har vist, at kombinationen af traditionel kinesisk medicin og vestlig medicin har betydelige terapeutiske virkninger p\u00e5 COVID-19. Derfor er screening af eksisterende l\u00e6gemidler udvundet af traditionel kinesisk medicin en effektiv strategi. Tidligere unders\u00f8gelser har vist, at klorokin er meget effektivt til at kontrollere SARS-CoV-2-infektion in vitro. Klorokin er en 4-aminoquinolin, der ud over prim\u00e6rt at blive brugt som malariamedicin ogs\u00e5 kan h\u00e6mme viral replikation ved at reducere den terminale glykosylering af ACE2-receptorer og forstyrre bindingen af SARS-CoV-2 til ACE2-receptorer. Cheng et al. brugte molekyl\u00e6r docking til at beregne bindingsenergien mellem chloroquin og ACE2. Resultaterne viste, at chloroquin har en potentiel bindingsevne med ACE2 med en fri bindingsenergi p\u00e5 -5,70 kcal\/mol og bindingssteder p\u00e5 Leu-95, Gln-58 og Gln-102. Sammenlignet med klorokin er de tre saponiners frie bindingsenergier ved docking med ACE2 lavere. For nylig, med den globale spredning af SARS-CoV-2-infektion, har flere og flere forskere v\u00e6ret involveret i screeningen af anti-SARS-CoV-2-l\u00e6gemidler. Yan et al. screenede 38 kinesiske patenterede l\u00e6gemidler for ACE (PDB: 1R4L) og virussens vigtigste M-protease (PDB: 6LU7) gennem molekyl\u00e6r docking og opdagede 10 potentielle anti-SARS-CoV-2-l\u00e6gemidler, herunder artemisinin, rutin, glycyrrhizin, puerarin, andrographolid og matrin. Liu et al. fandt gennem netv\u00e6rksfarmakologi og molekyl\u00e6re dockingstudier, at luteolin (-26,78 kJ\/mol), quercetin (-26,36 kJ\/mol) og demethyleret icariin (-25,94 kJ\/mol) binder sig til SARS-CoV-2 3CL-hydrolase. Nogle l\u00e6gemidler kan have lavere dockingaktivitet med ACE2, men det betyder ikke, at de ikke er egnede til COVID-19-behandling, da dockingsimuleringer kun giver en reference til indledende forskning. Desuden omfatter principperne for formulering af recepter for traditionel kinesisk medicin eliminering af patogen invasion og forbedring af kroppens modstandskraft. Derfor kan vi ikke udelukke muligheden for, at nogle l\u00e6gemidler kan have en effekt p\u00e5 COVID-19 ved at regulere immunsystemet.
