Som et naturligt l\u00e6gemiddel med lav toksicitet har det antiinflammatoriske, antioxidante, blodpladeh\u00e6mmende, \u00e5reforkalkningsh\u00e6mmende og andre virkninger. Derudover er resveratrol ogs\u00e5 et lovende antitumorl\u00e6gemiddel. Det har en bred vifte af m\u00e5l, s\u00e5som mTOR JAK\u3001\u03b2-amyloid\u3001Adenylyl cyclase\u3001IKK\u03b2\u3001DNA polymerase\uff0c Det er ogs\u00e5 en specifik SIRT1-aktivator og en effektiv h\u00e6mmer af pregnan X-receptor (PXR). Nylige unders\u00f8gelser har vist, at RES har in vitro- og in vivo-h\u00e6mmende aktivitet mod forskellige tumorer, men kliniske fors\u00f8g med RES som et l\u00e6gemiddel mod kr\u00e6ft er stadig begr\u00e6nsede. Resultaterne af at teste RES i tumordyrmodeller indikerer, at RES hovedsageligt spiller en forebyggende rolle i gastrointestinale tumorer. I dyremodellen for brystkr\u00e6ft kan RES blive et kr\u00e6ftterapeutisk middel. Denne gavnlige effekt afh\u00e6nger dog af dyret og celletypen. Der er relativt f\u00e5 rapporter om RES' rolle i livmoderhalskr\u00e6ftmodellen for human HPV-virusinfektion. Denne unders\u00f8gelse brugte hovedsageligt HeLa-celler til at generere tumorer subkutant i n\u00f8gne mus og unders\u00f8gte RES' anticancereffekt in vivo. Ved at p\u00e5vise RES' rolle i livmoderhalskr\u00e6ftcellers autofagi blev mekanismen for RES' h\u00e6mning af livmoderhalskr\u00e6ft ved at fremme autofagisk celled\u00f8d udforsket fra b\u00e5de in vivo- og in vitro-perspektiver.<\/p>\n<p>Behandlingen af livmoderhalskr\u00e6ft er hovedsageligt kirurgisk med postoperativ kombination af kemoterapi og str\u00e5lebehandling. P\u00e5 grund af de st\u00e6rke bivirkninger og modtagelighed for l\u00e6gemiddelresistens hos de fleste kemoterapimidler er den kliniske behandling og prognosen for livmoderhalskr\u00e6ft begr\u00e6nset. Derfor er der et presserende behov for at finde l\u00e6gemidler med gode antitumoreffekter og lav toksicitet til den kliniske behandling. I de senere \u00e5r har et stort antal unders\u00f8gelser bekr\u00e6ftet, at RES har en god antitumorevne og er effektiv i b\u00e5de tumorforekomst og -udvikling. Forskning har vist, at v\u00e6vsprotease L medierer autofagisk og apoptotisk celled\u00f8d induceret af resveratrol i livmoderhalskr\u00e6ftceller. Resveratrol inducerer et fald i mitokondriemembranpotentialet i cellelinjer med livmoderhalskr\u00e6ft, hvilket fremmer apoptose og \u00f8ger lysosomal permeabilitet. Denne unders\u00f8gelse validerede yderligere den h\u00e6mmende effekt af resveratrol p\u00e5 livmoderhalskr\u00e6ft gennem autofagi ved hj\u00e6lp af in vitro- og in vivo-eksperimenter. For det f\u00f8rste blev det bekr\u00e6ftet gennem in vivo-eksperimenter, at RES signifikant h\u00e6mmer udviklingen af livmoderhalskr\u00e6ft. Desuden afsl\u00f8rede detektion p\u00e5 v\u00e6vsniveau, at udtrykket af autofagirelaterede proteiner reguleres af RES. Derfor spekuleres der i, at mekanismen bag RES' h\u00e6mning af livmoderhalskr\u00e6ft kan v\u00e6re relateret til forekomsten af autofagisk celled\u00f8d (ACD) induceret af RES. ACD er resultatet af overdreven autofagi i celler, og det er ogs\u00e5 en Caspase-uafh\u00e6ngig type II programmeret celled\u00f8d, der adskiller sig fra apoptose. Dens vigtigste kendetegn er tilstedev\u00e6relsen af et stort antal autofagosomer og autolysosomer i cytoplasmaet, hvor st\u00f8rstedelen af stofferne i cytoplasmaet nedbrydes, men kernen forbliver intakt. Under vedvarende stress og igangv\u00e6rende autofagi vil celler d\u00f8 p\u00e5 grund af overdreven selvudt\u00f8mning. S\u00e5danne cellers d\u00f8d udviser ofte autofagiske karakteristika, prim\u00e6rt manifesteret ved overekspression af Beclin1 og produktion af et stort antal autofagosomer og autolysosomer i cellerne. Vacuole membrane protein-1 (VMP1) er et anerkendt autofagisk protein, og dets ekspressionsniveau er negativt korreleret med tumormalignitet. Sammen med Beclin-1 fungerer det som en molekyl\u00e6r kontakt til aktivering af autofagi. VMP1's udtryk og rolle i livmoderhalskr\u00e6ft er endnu ikke blevet rapporteret.<br \/>\nGennem RT-PCR, Western blot og autofagi-analyser blev det bekr\u00e6ftet, at RES fremmer forekomsten af ACD i livmoderhalskr\u00e6ftceller. Resultaterne viste, at Beclin-1, VMP1 og LC3B, der er placeret p\u00e5 den indre membran af autofagosomer og autolysosomer, kan induceres til at blive st\u00e6rkt udtrykt af RES i livmoderhalskr\u00e6ftceller med en lignende effekt som Rapa. Rapa er en h\u00e6mmer af pattedyrs TOR (mTOR) kinase, som binder sig til FKBP12 for at h\u00e6mme mTORC1-aktivering af autofagi og derved ud\u00f8ve tumorundertrykkende effekter. Yderligere unders\u00f8gelse af autofagosomer og autofagisk flux afsl\u00f8rede, at b\u00e5de RES og Rapa kan fremme dannelsen af autofagosomer og autolysosomer. Efter kombineret brug steg autofagosomer og autolysosomer yderligere, ledsaget af et fald i P62 og mitokondrier, men kernen forblev intakt; Samtidig observerede vi, at BAFA1 kan blokere den autofagiske str\u00f8m af livmoderhalskr\u00e6ftceller for\u00e5rsaget af RES. Kombinationen af BAFA1 og RES resulterede i ophobning af autofagosomer og P62, ledsaget af et betydeligt fald i mitokondrier og \u00e5benlyse abnormiteter i kernen sammenlignet med andre grupper. BAFA1 er et makrolidantibiotikum isoleret fra Streptomyces-arter og er en specifik h\u00e6mmer af den vakuol\u00e6re type H+ATPase (V-ATPase). BAFA1 kan ramme mitokondrier, f\u00e5 apoptosefremkaldende faktorer til at flytte sig fra mitokondrier til kernen og fremme caspase-uafh\u00e6ngig apoptose. Derudover inducerer BAFA1 bindingen af Beclin-1 til Bcl-2, hvilket yderligere h\u00e6mmer autofagi og fremmer celleapoptose. Vores eksperimentelle resultater bekr\u00e6ftede ogs\u00e5 dette punkt. Ved at m\u00e5le mitokondriemembranpotentialet og TUNEL-positivitetsraten fandt vi ud af, at RES kan fremkalde et fald i mitokondriemembranpotentialet for at fremme celleapoptose og give forskellige reaktioner p\u00e5 livmoderhalskr\u00e6ftceller under autofagi-aktivering og autofagi-inhibering. Det er blevet rapporteret i litteraturen, at RES forhindrer P62-nedbrydning induceret af Rapa, s\u00e5 kombinationen af Rapa og RES kan forhindre opregulering af autofagi og fremkalde apoptose af brystkr\u00e6ftceller.
