Naringin, som er en dihydroflavonoidforbindelse, har ogs\u00e5 biologiske aktiviteter som f.eks. antibakterielle, antitumor- og antioxidantegenskaber. Tutunchi og Filippin unders\u00f8gte, om naringin kunne bruges til at bek\u00e6mpe COVID-19. Naringin kan bruges i kombination med Vc og Ve til mere effektivt at behandle cadmiuminduceret og arsenikinduceret oxidativ stressinduceret leverskade hos Wistar-rotter. P\u00e5 grund af den d\u00e5rlige lipid- og vandopl\u00f8selighed af naringin selv er dets biotilg\u00e6ngelighed imidlertid ikke h\u00f8j. Derfor har forskningen i modificering af naringin f\u00e5et stor opm\u00e6rksomhed fra alle samfundslag.<\/p>\n<p>De phenoliske hydroxylgrupper i position 7 og 4 ' samt carbonylgruppen i position 4 i naringin er relativt aktive. Derfor kan naringins struktur \u00e6ndres gennem kemiske reaktioner for at forbedre dets vandopl\u00f8selighed og dermed \u00f8ge dets biotilg\u00e6ngelighed. If\u00f8lge litteraturrapporter har hydrazidforbindelser god antioxidantaktivitet, antibakteriel aktivitet, anticanceraktivitet osv. Men p\u00e5 grund af aminogruppernes indflydelse i hydrazid har de en vis toksicitet for organismer. Derfor er modificering af hydrazid for at opn\u00e5 hydrazon et effektivt middel til at forbedre hydrazids toksicitet. Acylhydrazonforbindelser har ogs\u00e5 gode antibakterielle, antioxidante og antiproliferative egenskaber. Baseret p\u00e5 den nuv\u00e6rende forskning i struktur-aktivitetsforhold og naringinmodifikation introducerer denne artikel derfor en hydrazidgruppe i position 4 for at danne en acylhydrazon i h\u00e5b om at opn\u00e5 naringinderivater med b\u00e5de biologisk aktivitet og reduceret toksicitet af hydrazidstrukturen. Udf\u00f8r in vitro antioxidantaktivitet og cytotoksicitetseksperimenter p\u00e5 de opn\u00e5ede derivater p\u00e5 HEK293-celler for at screene for h\u00f8j antioxidantaktivitet og stoffer med lav toksicitet, hvilket giver reference til yderligere udvikling og anvendelse.<\/p>\n<p>I dette eksperiment blev 13 naringinacylhydrazonderivater syntetiseret ved hj\u00e6lp af mikrob\u00f8lgeassisterede metoder, hvoraf 12 ikke er blevet rapporteret i litteraturen. In vitro-antioxidantaktiviteten af 13 derivater blev bestemt ved hj\u00e6lp af ABTS-, FRAP- og DPPH-metoder. Antioxidantaktiviteten af de opn\u00e5ede derivater er st\u00e6rkere end naringins. Desuden er antioxidantaktiviteten af de otte derivater b, e, g, i, j, k, l og m s\u00e6rlig signifikant, og antioxidantaktiviteten af derivaterne g, i, j, k, l og m kan sammenlignes med BHT, hvilket indikerer, at indf\u00f8relsen af acylhydrazon-grupper ved 4-position carbonyl af naringin gennem reaktion effektivt kan forbedre antioxidantaktiviteten af moderforbindelsen. Der blev udf\u00f8rt en struktur-aktivitetsanalyse af derivaternes antioxidantaktivitet, som viste, at n\u00e5r R-gruppen var en elektronudtr\u00e6kkende gruppe, blev derivaternes ladningst\u00e6thed spredt, og stabiliteten af de frie radikale mellemprodukter, der blev opn\u00e5et fra reaktionen, steg, hvilket resulterede i st\u00e6rkere antioxidantaktivitet end naringin selv. Resultaterne af effekten af 8 meget aktive derivater p\u00e5 HEK293-cellers overlevelsesrate viste, at bortset fra e og m var de andre derivater ikke-toksiske for HEK293 i koncentrationsomr\u00e5det 0-25 \u03bc mol\/L. Dette giver et referencegrundlag for dosering af tils\u00e6tningsstoffer i efterf\u00f8lgende unders\u00f8gelser af forskellige derivater. Efter den indledende screening har derivaterne g, j og k derfor god antioxidantaktivitet og lav cytotoksicitet. Blandt dem har derivat j b\u00e5de st\u00e6rk antioxidantaktivitet og lav cytotoksicitet og har visse anvendelsesmuligheder. Dette giver et grundlag for yderligere udvikling og forskning.
