Den har en historie med forbrug og dyrkning p\u00e5 over 5000 til 7000 \u00e5r. Quinoa har en h\u00f8j n\u00e6ringsv\u00e6rdi, er rig p\u00e5 phenoler og flavonoler og har antioxidante, immunregulerende og kr\u00e6fth\u00e6mmende egenskaber. I begyndelsen af 1960'erne begyndte Kina at introducere quinoa-ressourcer. I de senere \u00e5r er plantagearealet med quinoa i Kina hurtigt blevet udvidet. I 2019 oversteg det ansl\u00e5ede planteareal af quinoa i Kina 20000 kvadratmeter med en samlet produktion p\u00e5 20000 til 30000 tons. Quinoaklid (dvs. det ydre fr\u00f8lag af quinoa) indeholder en stor m\u00e6ngde triterpenoide saponiner, men saponinerne har en bitter smag. F\u00f8r man spiser quinoa, fjernes det ofte ved vask eller formaling, s\u00e5 der produceres en stor m\u00e6ngde quinoaklid under selve produktionen og forarbejdningen af quinoa. Den nuv\u00e6rende anvendelse af quinoaklid er dog kun simpel forarbejdning til brug som foder. Hvis quinoaklid kan udnyttes, kan den omfattende udnyttelsesv\u00e6rdi af quinoa forbedres. Triterpene saponiner har antiinflammatoriske, antitumor-, aterosklerose- og antidiabetes-aktiviteter. Desuden er det blevet rapporteret, at albumin i quinoaklid har antioxidant- og ACE-h\u00e6mmende aktiviteter. Dens 50%-ethanol-ekstrakt har beskyttende virkninger p\u00e5 kulstoftetrachlorid-induceret leverskade og fibrose hos mus ved at aktivere antioxidant-enzymsystemet og blokere TGF - \u03b2 1-vejen. Samtidig har unders\u00f8gelser vist, at polyfenoler og saponinekstrakter fra uforarbejdet quinoa har h\u00f8j h\u00e6mmende aktivitet p\u00e5 \u03b1-glucosidase.<\/p>\n<p>P\u00e5 nuv\u00e6rende tidspunkt er der ingen relevante litteraturrapporter om ultralydsekstraktionsprocessen af totale saponiner fra quinoaklid. Dette eksperiment brugte quinoa klid som det eksperimentelle materiale til at optimere ultralydsekstraktionsprocessen af samlede saponiner. P\u00e5 grundlag af enkeltfaktoreksperimenter blev forholdet mellem v\u00e6ske og materiale, ethanolkoncentration, ultralydstid og ultralydstemperatur valgt som uafh\u00e6ngige variabler, og ekstraktionsudbyttet af totale saponiner fra quinoaklid blev brugt som responsv\u00e6rdi. Responsoverflademetoden blev brugt til at optimere ultralydsekstraktionsprocessen af totale saponiner fra quinoaklid. Dette eksperiment unders\u00f8gte ogs\u00e5 den h\u00e6mmende aktivitet af ekstraktet p\u00e5 alfa glucosidase under de optimale ekstraktionsprocesbetingelser. Samtidig blev der etableret en enzymreaktionskinetikligning for at analysere dens inhiberingskinetik, og inhiberingsmekanismen for quinoaklid ekstrakt p\u00e5 alfa glucosidase blev udforsket for at udforske udviklingspotentialet for quinoaklid som en alfa glucosidaseh\u00e6mmer og give teoretisk grundlag og datast\u00f8tte til den videre udvikling og udnyttelse af quinoaklid.<\/p>\n<p>Den optimale proces til ultralydsekstraktion af saponiner fra quinoa klid blev optimeret ved hj\u00e6lp af respons overflade metodologi, med en v\u00e6ske til materiale forhold p\u00e5 15mL\/g, 75% ethanol ultralydsekstraktion, ultralyd tid p\u00e5 1,5 timer, og ultralyd temperatur p\u00e5 45 \u2103. Det samlede saponinekstraktionsudbytte var 2,370% \u00b1 0,022%. Denne proces har enkle betingelser, nem betjening og kontrol, god stabilitet og overvinder ulemperne ved konventionelle metoder s\u00e5som lang ekstraktionstid og besv\u00e6rlige varmebehandlingstrin.<\/p>\n<p>Eksperimentet med den h\u00e6mmende aktivitet af \u03b1-glucosidase viste, at det samlede saponinekstrakt af quinoaklid havde betydeligt st\u00e6rkere h\u00e6mmende aktivitet end den kommercielt tilg\u00e6ngelige \u03b1-glucosidaseh\u00e6mmer acarbose (P&lt;0,05), og det forventes at isolere sikre og effektive \u03b1-glucosidaseh\u00e6mmere fra det. Inhiberingsmekanisme og enzymkinetikunders\u00f8gelser har vist, at det samlede saponinekstrakt af quinoaklid er en reversibel blandet inhiberingstype, som giver en reference til afklaring af mekanismen for quinoaklid-ekstrakt til reduktion af postprandial blodsukker og giver datagrundlag og teknisk st\u00f8tte til dets h\u00f8jv\u00e6rdiudnyttelse.
