PM2.5 har et stort specifikt overfladeareal, hvilket g\u00f8r det nemt at transportere giftige og skadelige stoffer, der tr\u00e6nger ind i de dybe alveoler gennem \u00e5ndedr\u00e6tssystemet og for\u00e5rsager lungebet\u00e6ndelse og -skade og potentielt udvikler sig til lungefibrose. Den betydelige stigning i fibroblaster er det vigtigste kendetegn ved lungefibrose. Kollagen ophobes i periferien af pulmonale blodkar og alveol\u00e6re septa, og fibr\u00f8st bindev\u00e6v \u00f8ges ogs\u00e5 tilsvarende, hvilket i sidste ende f\u00f8rer til en stigning i matrixh\u00e5rdhed og vanskeligheder med gasudveksling. Endoendothelial mesenchymal transition (EndMT) er en af kilderne til fibroblaster, der er kendetegnet ved det gradvise tab af f\u00e6notypemark\u00f8rer som vaskul\u00e6r endothelcelle cadherin (VE cadherin\/CDH5) og platelet endothelial cell adhesion molecule-1 (CD31\/PECAM-1) i endothelceller, og \u00f8get udtryk af f\u00e6notypemark\u00f8rer som type I kollagen (Col1), vimentin, alfa glat muskelaktin (cta2\/alfa SMA) i mesenkymale celler. Fibroblast-specifikt protein 1 (Fsp1\/S100A4) og andre proteiner. Under denne proces \u00f8ges cellernes sprednings- og migrationsevne gradvist, hvilket i sidste ende f\u00f8rer til \u00e6ndringer i cellemorfologien. I de senere \u00e5r er den molekyl\u00e6re mekanisme bag inflammationsinduceret EndMT blevet forst\u00e5et. TGF - \u03b2-vejen er en klassisk vej til inflammationsinduceret EndMT, og alle tre isoformer af TGF - \u03b2 kan formidle forekomsten af EndMT i forskellige cellelinjer, hvor TGF - \u03b2 1 spiller den mest betydningsfulde rolle. Tidligere unders\u00f8gelser har vist, at PM2,5 kan \u00f8ge lungeinflammation ved at aktivere TGF - \u03b2-signalvejen, men den specifikke virkningsmekanisme og dens indvirkning p\u00e5 lungefibrose er stadig uklar.<\/p>\n<p>\u00c6teriske olier kan udvindes af blomster, frugter, fr\u00f8, blade, st\u00e6ngler eller r\u00f8dder fra aromatiske planter ved hj\u00e6lp af destillation, ekstrudering eller opl\u00f8sningsmiddelekstraktion. De har forskellige aktiviteter som f.eks. antibakterielle, antiinflammatoriske, antioxidante og anti-aging og er blevet brugt i vid udstr\u00e6kning inden for kliniske, farmaceutiske, sundheds- og wellness-omr\u00e5der. Den \"naturlige terapi\", der bruger \u00e6teriske olier udvundet af aromatiske planter til terapeutiske medicinske form\u00e5l, kaldes aromaterapi, som kan administreres gennem eksterne metoder som aromaterapi, ind\u00e5nding, topisk anvendelse eller hydroterapi. \u00c6teriske olier af eukalyptus, pebermynte, r\u00f8gelse og gran er almindelige \u00e6teriske olier, der bruges til at behandle luftvejssygdomme. De kan opl\u00f8se slim, fjerne slim, dr\u00e6be bakterier, lindre hoste og spasmer og derved beskytte lungerne mod bet\u00e6ndelse og lindre lungebet\u00e6ndelse. Vi brugte aromaterapi til at sammens\u00e6tte de fire typer \u00e6teriske olier, der er n\u00e6vnt ovenfor, til en sammensat \u00e6terisk olie til dette eksperiment og unders\u00f8gte interventionseffekten af den sammensatte \u00e6teriske olie p\u00e5 PM2.5-induceret EndMT gennem TGF - \u03b2 1\/SMAD3\/p-SMAD3-signalvejen.<\/p>\n<p>PM2.5 er et af de mest bekymrende luftforurenende stoffer. P\u00e5 grund af sine sm\u00e5 partikler er det lettere at n\u00e5 alveolerne og blodet i menneskekroppen og derved fremskynde sygdomsudviklingen hos dem, der allerede har lidt af lungesygdomme. Endotelcellerne p\u00e5 den indre v\u00e6g af blodkarrene kommer i direkte kontakt med blod og udviser betydelig heterogenitet som reaktion p\u00e5 stimulering af fremmedlegemer i blodet. Endothelceller i EndMT-processen er en af kilderne til fibroblaster, hvilket ikke kun f\u00f8rer til en betydelig stigning i fibroblaster, men ogs\u00e5 reducerer kapill\u00e6rsengens t\u00e6thed, forv\u00e6rrer v\u00e6vshypoxi og isk\u00e6mi og fremskynder processen med lungefibrose. Forel\u00f8bige laboratoriefors\u00f8g har vist, at PM2.5 kan fremkalde EndMT i MHC-celler. Proteinekspressionsniveauerne af Acta2, Col1 og S100A4 \u00f8ges, mens ekspressionsniveauerne af CD31 og CDH5 falder, hvilket indikerer, at PM2,5 kan inducere EndMT i pulmonale mikrovaskul\u00e6re endotelceller fra mus. Det \u00f8gede proteinekspressionsniveau af S100A4 bekr\u00e6fter ogs\u00e5 den vigtige rolle, som End MT spiller i lungefibrose.<\/p>\n<p>Hovedkomponenterne i den sammensatte \u00e6teriske olie, der blev brugt i dette eksperiment, er alfapinen (16,54%), eukalyptol (16,7%) og menthol (12,86%). Eucalyptol er en tumornekrosefaktor alfa-repressor, der effektivt kan h\u00e6mme udviklingen af astma og kroniske lungeobstruktive sygdomme og har potentiale som et steroid-antiinflammatorisk l\u00e6gemiddel. Alfa-pinen har antioxidant- og antibakterielle virkninger og udviser en vis h\u00e6mmende aktivitet mod tumorcellelinjen A549. Menthol kan blokere L-type calciumkanaler i vaskul\u00e6r glat muskulatur, hvilket har en vis terapeutisk effekt p\u00e5 hjerte-kar-sygdomme. Dette eksperiment viste, at PM2.5 kan fremkalde spredning og migration af MHC-celler, s\u00e5 de n\u00e5r deres h\u00f8jdepunkt efter 48 timers behandling. Men i gruppen med \u00e6terisk olie gennemgik MHC-cellernes overordnede funktion ikke v\u00e6sentlige \u00e6ndringer p\u00e5 grund af PM2.5-induktion. Efter 36 timers intervention med sammensat \u00e6terisk olie blev proteinekspressionsniveauerne af Acta2 og Col1 reduceret betydeligt, hvilket indikerer, at sammensat \u00e6terisk olie har en god lindrende effekt p\u00e5 forekomsten af EndMT.<\/p>\n<p>Forskning har vist, at TGF - \u03b2 spiller en vigtig rolle i udviklingen, skaden og reparationen af lungerne. Det kan fremme syntesen af ekstracellul\u00e6r matrix ved at aktivere TGF - \u03b2 1-receptorer og regulere patogenesen af lungefibrose. Tidligere eksperimenter har vist, at efter behandling af MHC-celler med PM2.5 i 48 timer \u00f8ges proteinekspressionsniveauerne af TGF - \u03b2 1, TGF - \u03b2 R1 og p-SMAD3. Men efter tils\u00e6tning af SB431542 til PM2.5-gruppen i 48 timer falder ekspressionen af TGF - \u03b2 R1 og p-SMAD3. Da SMAD-proteinphosphorylering er en typisk indikator for TGF - \u03b2 1-signalvejen, kan det ses, at PM2.5 kan aktivere TGF - \u03b2 R1 ved at regulere TGF - \u03b2-signalvejen og stimulere transformationen af TGF - \u03b2 1 til en aktiv form. TGF - \u03b2 R1 aktiveres og bindes til SMAD3. Overekspression af p-SMAD3 kan fremme ekspressionen af mesenkymale cellemark\u00f8rer og kollagenakkumulering. I gruppen med den sammensatte \u00e6teriske olie blev proteinekspressionsniveauerne af TGF - \u03b2 R1 og p-SMAD3 reduceret betydeligt. Den sammensatte \u00e6teriske olie h\u00e6mmede aktiveringen af TGF - \u03b2 R1 for at mindske udtrykket af p-SMAD3 og undertrykte derved PM2.5-induceret EndMT.<\/p>\n<p>Det kan udledes, at sammensatte \u00e6teriske olier kan bremse forekomsten af EndMT induceret af PM2.5 i pulmonale endotelceller ved at h\u00e6mme udtrykket af TGF - \u03b2 1\/SMAD3\/p-SMAD3-signalvejen.
