Som f\u00f8lge heraf er de sekund\u00e6re metabolitter, de producerer, meget komplekse og forskelligartede. Marine mikrobielle kilder har et ekstremt rigt udvalg af naturlige produkter, der d\u00e6kker n\u00e6sten alle strukturelle typer af naturlige produkter og besidder en bred vifte af biologiske aktiviteter, hvilket g\u00f8r dem til en vigtig kilde til hovedforbindelser til nye naturlige l\u00e6gemidler. Studiet af marine mikroorganismers sekund\u00e6re metabolitter vil g\u00f8re det muligt at opdage blyforbindelser med nye kemiske strukturer og betydelige biologiske aktiviteter, hvilket giver vigtige modelstrukturer og l\u00e6gemiddelforstadier til forskning i og udvikling af nye l\u00e6gemidler.<\/p>\n<p>Vores forskningsgruppe har tidligere isoleret og identificeret svampe fra sedimenter p\u00e5 4314 meters dybde i Det Sydkinesiske Hav (114 \u00b0 34 \u2032 32 \u2033 \u00d8, 13 \u00b0 57 \u2032 27 \u2033 N) og opn\u00e5et en ny marin svamp Neoroussolella sp. If\u00f8lge litteraturgennemgangen er der i \u00f8jeblikket ingen rapporter om forskning i sekund\u00e6re metabolitter b\u00e5de nationalt og internationalt. Gennem HPLC-MS-analyse af r\u00e5ekstraktet fra flydende fermentering i lille skala blev det konstateret, at dets metabolitter var meget rigelige. Baseret p\u00e5 dette brugte vi normal silikagels\u00f8jle, gels\u00f8jle, h\u00f8jtydende v\u00e6skekromatografi og andre kromatografiske teknikker til systematisk at studere den kemiske sammens\u00e6tning af denne svamp.<\/p>\n<p>Neoroussolella er en ny svampesl\u00e6gt, som f\u00f8rst blev opdaget og beskrevet af Liu et al. i 2014. P\u00e5 nuv\u00e6rende tidspunkt kommer Neoroussolella-sl\u00e6gtens svampe, der er blevet isoleret og identificeret, hovedsageligt fra planter og findes i bambus, urteagtige planter og palmer, mens der kun er f\u00e5 rapporter om marine kilder, hovedsageligt koncentreret i Middelhavsomr\u00e5det, og alle kilder er fra bentiske planter. FS526-stammen, der blev brugt i denne unders\u00f8gelse, blev isoleret fra dybhavssedimenter i Det Sydkinesiske Hav. If\u00f8lge litteraturen har der ikke v\u00e6ret nogen rapporter om de sekund\u00e6re metabolitter i denne svampesl\u00e6gt.<\/p>\n<p>Denne unders\u00f8gelse identificerede for f\u00f8rste gang 15 monomere forbindelser fra de faste fermenteringsprodukter fra dybhavssvampen Neoroussolella sp. FS526 med forskellige strukturelle typer, herunder 2 terpener, 5 isocoumariner, 2 benzofuraner, 1 monoterpen og 5 phenolderivater. Alle forbindelser blev isoleret fra svampe af Neoroussolella-sl\u00e6gten for f\u00f8rste gang. Baseret p\u00e5 de strukturelle typer af disse forbindelser spekuleres der i, at phenol og dets derivater er de vigtigste karakteristiske sekund\u00e6re metabolitter i denne svamp, som hovedsageligt produceres gennem polyketidbiosyntesevejen. Forbindelse 1 har strukturelle analoger med forskellige C-9-konfigurationer. S-konfigurationen af denne strukturelle type blev f\u00f8rst opdaget af Stierle et al. fra Penicillium-svampe og navngivet Berkeleyacetal C. Denne forbindelse har h\u00e6mmende virkninger p\u00e5 ikke-sm\u00e5cellede lungecancercellelinjer. Senere opdagede Sun et al. ogs\u00e5 Berkeleyacetal C fra Penicillium purpurogenum MHZ 111. Efterf\u00f8lgende isolerede og rensede Chen et al. en ny strukturel analog med C-9 R-konfiguration fra Talaromyces amettolkiae YX1 og kaldte den amettolkolid B. Aktivitetstest blev udf\u00f8rt p\u00e5 to forskellige konfigurationer af monomere forbindelser, Berkeleyacetal C og Amestolkolid B, som begge viste st\u00e6rk NO-produktionsh\u00e6mmende aktivitet, men der blev ikke fundet nogen signifikant cytotoksisk aktivitet. Resultaterne af den biologiske aktivitetstest i denne unders\u00f8gelse viste, at forbindelse 1 har svag cytotoksisk aktivitet. Derudover har forbindelse 7 af isocoumarin st\u00e6rk antioxidantaktivitet, og dens IC50-v\u00e6rdi er t\u00e6t p\u00e5 den positive kontrol Vc, hvilket er v\u00e6rd at unders\u00f8ge yderligere.
