Unormal aktivering af HDAC'er er t\u00e6t forbundet med sygdomme som tumorigenese og unormal v\u00e6vsudvikling. De udviklede histondeacetylaseh\u00e6mmere (HDACi) er blevet anvendt i klinisk praksis. For eksempel er SAHA og Romidpesion blevet terapeutiske l\u00e6gemidler til hud- og perifere lymfocyt\u00e6re tumorer. HDAC-h\u00e6mmere har forskellige strukturer, herunder kortk\u00e6dede fedtsyrer som sm\u00f8rsyre og trichostatin A, cykliske peptider samt flavonoider som genistein og kaempferol, hvilket g\u00f8r dem til et varmt emne inden for opdagelse af nye l\u00e6gemidler. Epimedium er en traditionel kinesisk medicin, der styrker nyrer og knogler med virkninger som at styrke muskler og knogler, modst\u00e5 gigt og forbedre lever- og nyrefunktion; farmakologiske unders\u00f8gelser har afsl\u00f8ret virkningerne af Epimedium-urt p\u00e5 forbedring af seksuel funktion, regulering af hormoner, modulering af immunitet og forebyggelse og behandling af osteoporose. Epimedium-urten indeholder mere end 140 forskellige typer flavonoider, men der er kun f\u00e5 forskningsrapporter om de aktive ingredienser mod osteoporose i Epimedium baseret p\u00e5 HDAC-inhiberingsmekanismen. Denne artikel rapporterer resultaterne af vores screening og identifikation af HDAC-h\u00e6mmende aktive ingredienser i Epimedium sagittatum under vejledning af aktivitetssporing.<\/p>\n<p>Dette eksperiment fastslog, at opl\u00f8sningsmiddelstedet med st\u00e6rk h\u00e6mning af HDAC-aktivitet af 70%-ethanolekstraktet af Epimedium sagittatum var ethylacetatstedet, som blev behandlet med D101 makropor\u00f8s adsorptionsharpiks. Elueringsprodukterne af 40% og 60% ethanol var de HDAC-h\u00e6mmende aktive steder (se figur 1); elleve forbindelser blev identificeret gennem s\u00f8jlekromatografiseparation og spektralanalyse styret af aktivitet. Blandt dem viste ikke-isopentensubstituerede flavonoidglykosider (forbindelse 1) og flavonoidglykosider (forbindelser 2-10) alle st\u00e6rkere HDAC-h\u00e6mmende aktivitet end icariin, hvilket indikerer, at disse komponenter er de vigtigste aktive ingredienser i h\u00e6mning af HDAC-aktivitet i denne plante. Blandt de 9 isolerede flavonoidglykosider var 6 kaempferol-3-O-glucosid, herunder forbindelse 5 med h\u00f8jt udbytte (kaempferol-3-O-\u03b2 - D-glucopyranosid), hvilket indikerer, at kaempferol-3-O-glucosid har relativt st\u00e6rk HDAC-h\u00e6mmende aktivitet. Forbindelserne med lavere aktivitet er 7 og 8. P\u00e5 grund af forestring af en eller to hydroxylgrupper p\u00e5 sukkergruppen med coumarsyre er den HDAC-h\u00e6mmende aktivitet af 7 og 8 lavere end for isorhamnetin-3-O-glucosid (9 og 10), hvilket indikerer, at forestring af coumarsyre p\u00e5 sukkergruppen af kaempferol-3-O-glucosid kan sv\u00e6kke den HDAC-h\u00e6mmende aktivitet (se tabel 3). Men da HDAC-inhibitor-screeningskittet indeholder HDAC udvundet fra kernen af HeLa-celler, som er en blanding, der indeholder forskellige undertyper af HDAC, er der behov for yderligere forskning for at bestemme, hvilken eller hvilke HDAC-aktiviteter (funktioner) der h\u00e6mmes af flavonoiderne i Epimedium sagittatum.<\/p>\n<p>Som tidligere n\u00e6vnt kan h\u00e6mning af HDAC ogs\u00e5 opn\u00e5s ved at regulere dets udtryk i cellerne. Vores unders\u00f8gelse med HeLa-celler viste (som vist i figur 2), at kaempferol-3-O-alpha-L-actanopyranosid (3), kaempferol-3-O-beta-D-xylopyranosid (4) og kaempferol-3-O-beta-D-glucopyranosid (5) ikke havde nogen signifikant effekt p\u00e5 udtrykket af type I HDAC'er, men kunne h\u00e6mme HDAC6 signifikant i type II HDAC'er (forbindelser 3 og 4). 5) og HDAC10 (forbindelser 4 og 5) er nyopdagede flavonoide HDAC-proteinekspressionsh\u00e6mmere; Blandt dem h\u00e6mmer 4 samtidig proteinudtrykket af andre klasse II (HDAC4-7) og klasse IV (HDAC9-11) HDAC'er, og deres virkninger fortjener yderligere unders\u00f8gelse.<\/p>\n<p>HDAC spiller en vigtig rolle i celletranskriptionsregulering, cellecyklusprogression og v\u00e6kst- og udviklingsaktiviteter ved at deacetylerer centrale histoner for at undertrykke transkriptionen af specifikke gener. Selektive HDACi kan virke p\u00e5 overudtrykte HDAC'er, s\u00e5 undertrykte gener kan udtrykkes normalt og dermed spille en rolle i behandlingen af sygdomme som tumorer, gigt og v\u00e6vsdysplasi. For eksempel formidler HDAC1-2 (type I) reparation af DNA-skader og er st\u00e6rkt udtrykt i forskellige kr\u00e6ftv\u00e6v som lungekr\u00e6ft og mavekr\u00e6ft. Under str\u00e5lebehandling af tumorer kan samtidig brug af HDAC1-2-h\u00e6mmere forhindre tumorceller i at underg\u00e5 DNA-reparation og fremme apoptose af kr\u00e6ftceller. Den unormale ekspression af HDAC8 (type I) og HDAC10 (type II) er et kendetegn for progressionen og maligniteten af neuroblastom. HDAC6-8-10-inhibitoren TH34 kan inducere tumorcelleapoptose, mitoseafbrydelse og cellecyklusstop, hvilket f\u00f8rer til tumorcelled\u00f8d; Kombinationen af TH34 og retinsyre kan give en synergistisk effekt. Opregulering af HDAC4-6-7 (type II)-ekspression f\u00f8rer til nedregulering af vigtige transkriptionsfaktorer Runx2 og Qstelix under osteoblastdifferentiering, hvilket er ugunstigt for osteoblastdannelse; Opregulering af HDAC6-gen- og proteinekspression kan ogs\u00e5 inducere deformation og forkortelse af primitive cilier i osteoblaster, hvilket resulterer i et fald i antallet af cilierede celler, hvorved osteoblasters ALP-funktion h\u00e6mmes og effektivt forhindrer osteogen modning induceret af knoglemorfogenetisk protein (BMP).
