Mange strukturelt nye aktive forbindelser er blevet opdaget i svampe af Alternaria-sl\u00e6gten, hovedsageligt pyranon, terpener, steroler, aromatiske polyketoner, antrakinoner, alkaloider, peptider osv. Nogle forbindelser har biologiske aktiviteter som enzymh\u00e6mning, cytotoksicitet, svampedr\u00e6bende midler, antioxidanter og fremme af rodv\u00e6kst i en- og tokimbladede planter. Blandt dem har Alternaria-toksiner som Alternariol, Alternariol-methylether og tentoxin betydelig toksicitet. Alternaria tenuissima er en almindelig milj\u00f8svamp, der er relateret til A. Der er relativt lidt forskning i de sekund\u00e6re metabolitter i tenuissima. I vores unders\u00f8gelse fik Pan et al. A. fra dybhavssedimentkilder Nye spirocykliske hydrogenerede anthraquinon- og nitrogenholdige anthraquinonforbindelser blev isoleret fra tenuissima DEFSCOS013.<br \/>\nFor at finde nye antibiotika brugte vi Streptococcus agalactiae og Streptococcus iniae som indikatorbakterier til at screene en r\u00e6kke svampe, der var isoleret og identificeret fra tarmkanalen hos tilapia, for antibakteriel aktivitet. Denne unders\u00f8gelse brugte aktivitetssporingsmetoden til at unders\u00f8ge de aktive forbindelser i fermenteringsprodukterne fra A. tenuissima SCSIO41701 rismedium, der h\u00e6mmer v\u00e6ksten af Streptococcus agalactiae og Streptococcus iniae i tilapia.<\/p>\n<p>I denne unders\u00f8gelse identificerede vi gennem sporing af antibakteriel aktivitet, at den vigtigste aktive forbindelse i svampen A. tenuissima SCSIO41701, der h\u00e6mmer v\u00e6ksten af Streptococcus agalactiae og Streptococcus iniae i tilapia, er et h\u00f8jt indhold af aternariol (1). Gennem HPLC-analyse og tyndlagspladeanalyse fandt vi, at komponenterne Fr.6~Fr.10 indeholder en stor m\u00e6ngde forbindelser 1 og 2, hvor forbindelse 1 er den vigtigste top. Det spekuleres i, at komponenterne Fr.6~Fr.10 med bedre antibakteriel aktivitet hovedsageligt ud\u00f8ver antibakterielle virkninger gennem forbindelse 1. \u00c5rsagen til krydsfordelingen af forbindelser 1 og 2 i disse komponenter skyldes det h\u00f8je indhold af forbindelser 1 og 2, der er letopl\u00f8selige i dichlormethan-methanol-opl\u00f8sningssystemet, der bruges til vask, n\u00e5r r\u00e5ekstraktet adskilles med en normalfaset silicagels\u00f8jle. Forbindelserne 1-6 tilh\u00f8rer alle klassen af benzopyranolidforbindelser. N\u00e5r man sammenligner deres strukturer og antistreptokokaktivitet mod tilapia, spekuleres der i, at tilstedev\u00e6relsen af et konjugeret system mellem hydroxylgruppen p\u00e5 C-5-benzenringen og C7~C10a spiller en vigtig rolle i deres antibakterielle aktivitet.<br \/>\nForbindelse 1 og 2 er almindelige streptomycintoksiner, der kan p\u00e5vises i f\u00f8devarer som korn, frugt, rapsfr\u00f8, solsikkefr\u00f8, oliven osv. Forbindelse 1 og 2 har begge cytotoksisk aktivitet og kan fremkalde genetiske mutationer, hvilket f\u00f8rer til genotoksiske brud p\u00e5 DNA-strenge in vitro og h\u00e6mmer topoisomeraser I og II \u03b1. Derudover kan forbindelse 2 inducere mitokondriemutationer i humane tyktarmskr\u00e6ftceller. Derudover har forbindelse 1 god aktivitet til at h\u00e6mme Staphylococcus aureus, Streptococcus lactiae, Escherichia coli og Pseudomonas aeruginosa med MIC-v\u00e6rdier p\u00e5 8,82 \u03bc g\/mL. Det h\u00e6mmer ogs\u00e5 kraftigt aktiviteten af plantepatogene svampe som \u00e6bler\u00e5d, vandmelonvisnesyge, tomatgr\u00e5skimmel og peberphytophthora med MIC-v\u00e6rdier p\u00e5 15,63, 7,82, 1,96 og 3.91 \u03bc g\/mL; Forbindelse 2 har ogs\u00e5 antibakterielle virkninger p\u00e5 Bacillus subtilis, Pseudomonas aeruginosa, Ralstonia lactis, Pseudomonas aeruginosa og Pseudomonas aeruginosa med MIC50-v\u00e6rdier p\u00e5 henholdsvis 28,83, 34,29, 27,08, 29,21 og 30,06 \u03bc g\/mL. I denne unders\u00f8gelse fandt vi for f\u00f8rste gang, at forbindelse 1 har st\u00e6rk aktivitet i at dr\u00e6be tilapia-patogene bakterier Streptococcus agalactiae og Streptococcus dolphin og har et h\u00f8jt udbytte i stammen A. tenuissima SCSIO41701.
