De fremmer ikke kun planters stressresistens som salttolerance, t\u00f8rketolerance, kuldetolerance og insektresistens, men indeholder ogs\u00e5 rige genklynger til syntese af sekund\u00e6re metabolitter, som er blevet en vigtig kilde til screening af strukturelt forskellige naturprodukter. Den vilde Dongxiang-ris (Oryza rufipogon Griffith.) er den nordligste vilde ris i verden og findes kun i Dongxiang County i Jiangxi-provinsen. P\u00e5 grund af dens langsigtede vilde status og naturlige udv\u00e6lgelse har den fremragende egenskaber som stressresistens, sygdoms- og skadedyrsresistens, kulderesistens, t\u00f8rketolerance og h\u00f8jt udbytte. Vores forskningsgruppe gennemf\u00f8rte en systematisk unders\u00f8gelse af mangfoldigheden af endofytiske svampe i Dongxiang vilde ris i den tidlige fase og fandt, at den ikke kun indeholder rigelige planteendofytiske svampe, men ogs\u00e5 PKS-genp\u00e5visning og screeningsresultater for plantepatogeninhibering viser, at den indeholder rigelige aktive sekund\u00e6re metabolitter. Yderligere forskning er n\u00f8dvendig.<br \/>\nDenne unders\u00f8gelse fokuserer p\u00e5 en stamme af endofytisk Chaetomium globosum S10, der er isoleret fra vilde ris i Dongxiang, og udf\u00f8rer fermenteringskultur p\u00e5 den. Det blev forel\u00f8bigt konstateret, at dens fermenteringsbouillon har en vis h\u00e6mmende aktivitet p\u00e5 tumorceller. Globosum er en svamp, der tilh\u00f8rer familien Trichomycteridae og sl\u00e6gten Trichomycetes. Den har v\u00e6ret klassificeret som s\u00e5dan siden 1973, da Sekita blev overf\u00f8rt fra C Siden isoleringen af Sphingomonas A og B fra Globosum er der blevet opdaget mere end 400 strukturelt nye aktive sekund\u00e6re metabolitter i sl\u00e6gten Sphingomonas. Disse forbindelser omfatter hovedsageligt Sphingomonas, nitrogenholdige ketoner, terpener, estere, flavonoider, isocoumariner osv. Blandt dem udg\u00f8r Sphingomonas og azone-forbindelser langt st\u00f8rstedelen, mens andre forbindelser som isocoumariner sj\u00e6ldent er isoleret. Kun Yan et al. isolerede isocoumarin-forbindelsen prochaetoviridin A fra Sphingomonas, der stammer fra Ginkgo biloba. I denne unders\u00f8gelse blev tre azonforbindelser, chaetomugilin D (3), chaetomugilin S (4) og chaetoviridis A (5), samt en almindelig alkaloidforbindelse, coumarin A (6), isoleret fra bakterien i 108. Samtidig blev isocoumarinforbindelsen 6-hydroxymellein (2) og naphthoquinonforbindelsen 6-O-demethyl-5-deoxyfusarubin (7) isoleret fra svampen i sl\u00e6gten Trichoderma for f\u00f8rste gang. 6-Hydroxymellein (2) er et almindeligt biosyntetisk mellemprodukt, og mikroorganismer kan syntetisere adskillige strukturelle analoger ved at bruge det som forl\u00f8ber. Det tyder p\u00e5, at S10's genom kan indeholde relaterede polyketidsyntase (PKS)-genklynger. Yderligere udforskning af de sekund\u00e6re metabolitter i S10 kan f\u00f8re til isolering af nye strukturer og aktiviteter af forbindelser.<br \/>\nDenne unders\u00f8gelse fokuserer p\u00e5 svampen C Syv forbindelser blev isoleret og oprenset fra Globosum S10, og deres biologiske aktiviteter er blevet rapporteret i vid udstr\u00e6kning. Blandt dem kan forbindelserne 3 og 4, som er azoneforbindelser s\u00e5som chaetomugilin D (3), chaetomugilin S (4) og chaetoviri dins A (5), effektivt h\u00e6mme v\u00e6ksten af kimplanter. For eksempel har forbindelserne 3 og 4, som Wang et al. isolerede fra en Ginkgo biloba-afledt svamp, en betydelig h\u00e6mmende effekt p\u00e5 fr\u00f8planternes v\u00e6kst, og deres IC50 er lavere end for det bredspektrede herbicid glyphosat. Derfor skal de udvikles til naturlige og milj\u00f8venlige herbicider. Forbindelse 5 udviser god antifungal aktivitet og har en god h\u00e6mmende effekt p\u00e5 nukleare diskbakterier. Dens forebyggende og terapeutiske virkninger kan sammenlignes med det bredspektrede fungicid carbendazim. Globulin A (6) har ogs\u00e5 vist god anti-cancer aktivitet. Knudsen et al. fandt ud af, at forbindelse 6 isoleret fra Penicillium kan bruges som et potentielt l\u00e6gemiddel til behandling af kronisk lymfatisk leuk\u00e6mi (CLL). Under simulerede lymfoide v\u00e6vskulturbetingelser udviser det ogs\u00e5 signifikant induktion af CLL-celleapoptose. P\u00e5 nuv\u00e6rende tidspunkt er der ingen rapporter om aktiviteten af 6-O-demethyl-5-deoxyfusarubin (7), men dets analoge seglcellelinje har den effekt, at den h\u00e6mmer spredningen af blodtumorcellelinjer og \u00f8ger celleapoptose. Denne unders\u00f8gelse evaluerede de h\u00e6mmende virkninger af forbindelser p\u00e5 melanom B16-celler. Resultaterne viste, at bortset fra forbindelse 2 havde de andre seks forbindelser ingen h\u00e6mmende virkning p\u00e5 melanom B16-celler.<br \/>\nIndtil videre er 6-hydroxymellein (2) kun blevet isoleret fra de endofytiske svampe Daldinia Eschscholtzii og Aspergillus terreus i mangrover, og det har vist sig at have en h\u00e6mmende effekt p\u00e5 Arabidopsis' pollenudvikling. Derudover fandt Yang et al, at forbindelse 2 ikke har nogen h\u00e6mmende virkning p\u00e5 human brystkr\u00e6ftcellelinje MDA-MB-231 og human bugspytkirtelkr\u00e6ftcellelinje PANC-1, men kun svag h\u00e6mmende aktivitet p\u00e5 ikke-sm\u00e5cellet lungekr\u00e6ftcellelinje A549 og human leverkr\u00e6ftcellelinje BEL-7402 med IC50 p\u00e5 henholdsvis 170,6 mg \/ L og 315,0 mg \/ L. Denne unders\u00f8gelse viste f\u00f8rst, at 6-hydroxymellein (2) har god h\u00e6mmende aktivitet p\u00e5 melanom B16-celler med en IC50 p\u00e5 2,24 \u03bc mol\/L. Melanom, som en almindelig ondartet hudtumor i klinikken, har en h\u00f8j grad af malignitet, er let at metastasere, og forekomsten har en stigende tendens \u00e5r for \u00e5r.
