Omkring 85% af lungekr\u00e6ften er ikke-sm\u00e5cellet lungekr\u00e6ft (NSCLC). De vigtigste kliniske behandlinger af lungekr\u00e6ft omfatter kirurgi, kemoterapi, str\u00e5lebehandling og biologisk behandling. Selvom fuldst\u00e6ndig kirurgisk resektion er effektiv, er foruds\u00e6tningen, at sygdommen er medicinsk operabel og kan iscenes\u00e6ttes korrekt. Kemoterapimedicin har uspecifikke egenskaber og giver ofte alvorlige toksiske bivirkninger. Traditionel kinesisk medicin har spillet en unik rolle i forebyggelse og behandling af tumorer p\u00e5 grund af dens egenskaber med flere m\u00e5l, flere veje og flere effekter. Flere ingredienser i traditionel kinesisk medicin og ekstrakter heraf har vist sig at have gode antitumoreffekter, f.eks. matrin, genistein og paclitaxel, hvoraf nogle er blevet brugt i vid udstr\u00e6kning i klinisk praksis. Kunlun Snow Chrysanthemum er ogs\u00e5 kendt som Coreopsis tinctoria Nutt. Moderne farmakologiske unders\u00f8gelser har vist, at Xinjiang Kunlun Snow Chrysanthemum er rig p\u00e5 forskellige kemiske komponenter s\u00e5som polyfenoler, flavonoider og polysaccharider. Blandt dem har flavonoider vigtige farmakologiske virkninger som f.eks. s\u00e6nkning af blodlipider, regulering af blodsukker og antitumoreffekter. Men p\u00e5 grund af de mange flavonoider, de mange m\u00e5l og de mange virkningsveje er der visse vanskeligheder med at studere deres molekyl\u00e6re mekanismer til behandling af sygdomme.<br \/>\nNetv\u00e6rksfarmakologi er en forskningsmetode baseret p\u00e5 systembiologi og tv\u00e6rfaglig farmakologi, som konstruerer et interaktionsnetv\u00e6rk p\u00e5 flere niveauer af \"l\u00e6gemiddelm\u00e5lsygdom\" for at analysere l\u00e6gemiddelm\u00e5l og mekanismer p\u00e5 det overordnede niveau. Denne artikel bruger netv\u00e6rksfarmakologi kombineret med in vitro-celleeksperimenter til at analysere antitumorkomponenterne og -mekanismerne i Kunlun krysantemum total flavonoider (KCTF) p\u00e5 NSCLC, hvilket giver eksperimentel evidens for den efterf\u00f8lgende l\u00e6gemiddeludvikling og kliniske anvendelse.<\/p>\n<p>Flavonoider, som findes i mange forskellige planter, er st\u00e6rke antioxidanter, der effektivt kan eliminere frie radikaler i kroppen og har vigtige farmakologiske virkninger som f.eks. anti-tumor, blodtrykss\u00e6nkning og anti-aging. Denne unders\u00f8gelse analyserede de samlede flavonoider i Kunlun Snow Chrysanthemum ved hj\u00e6lp af HPLC-teknologi. Resultaterne viste, at det relativt h\u00f8je indhold af flavonoider omfattede epigallocatechingallat, quercetin, luteolin, catechiner osv. Talrige forskningsresultater har vist, at flavonoider som quercetin og luteolin har god antitumoraktivitet mod forskellige solide tumorer. In vitro-cellefors\u00f8g har vist, at totale flavonoider fra Kunlun Snow Chrysanthemum har en signifikant h\u00e6mmende effekt p\u00e5 spredningen af HCTI16-celler og Caco-2-celler fra menneskelig tyktarmskr\u00e6ft. Den traditionelle kinesiske medicins karakteristiske synergistiske virkninger med flere komponenter, flere m\u00e5l og flere veje g\u00f8r imidlertid moderne forskning i dens virkningsmekanisme vanskelig.<br \/>\nI 2007 foreslog Hopkins netv\u00e6rksfarmakologi, som repr\u00e6senterer forholdet mellem l\u00e6gemidler, m\u00e5l og sygdomme i et netv\u00e6rk. Brugen af netv\u00e6rksfarmakologi kan systematisk analysere komponenterne i traditionel kinesisk medicin, udforske de f\u00e6lles m\u00e5l for l\u00e6gemidler og sygdomme og yderligere analysere biologiske funktioner og signalveje. Denne holistiske og systematiske forskningsmodel er i overensstemmelse med den holistiske teori om traditionel kinesisk medicin.<br \/>\nI denne unders\u00f8gelse blev 68 komponenter i KCTF screenet ved hj\u00e6lp af TCMSP, og 891 m\u00e5l for 27 flavonoide komponenter blev forudsagt. Efter at have krydset med NSCLC-m\u00e5l blev der opn\u00e5et 217 mulige m\u00e5l for KCTF-intervention i NSCLC. Blandt dem er vigtige m\u00e5l som MAPK1, EGFR, Tp53, Akt, Bax, Bcl-2, Caspase 3 relateret til flere kemiske komponenter, hvilket indikerer et t\u00e6t synergistisk forhold mellem de aktive ingredienser i KCTF. For yderligere at analysere mekanismen for KCTF-intervention i NSCLC blev forholdet mellem de to analyseret ved at konstruere et PPI-netv\u00e6rk og berige KEGG-biologiske veje. Resultaterne viste, at PI3K Akt-signalvej, Bax\/Bcl-2 antiapoptotisk signalvej, p53-signalvej, MAPK-signalvej osv. var relateret til den antitumorale effekt af KCTF.<br \/>\nDe vigtigste virkningsmekanismer for antitumormedicin er at inducere tumorcelleapoptose, h\u00e6mme cellecyklussen og undertrykke migration. Forskning har vist, at mange planteflavonoider (s\u00e5som curcumin, anthocyaniner, epigallocatechingallat, quercetin osv.) kan forstyrre PI3K Akt-signalvejen, direkte h\u00e6mme PI3k eller blokere PI3K Akt-signalvejen, yderligere regulere udtrykket af downstream-effektormolekyler, fremskynde tumorcellecyklus, fremme celleapoptose og h\u00e6mme celleproliferation og opn\u00e5 anti-tumoreffekter. Xiang et al. fandt, at quercetin inducerer apoptose i HeLa-celler ved at nedregulere ekspressionen af PI3k, Akt og Bcl-2 i PI3K Akt-signalvejen, opregulere Bax-ekspressionen og for\u00e5rsage cellecyklusstop i G0\/G. Osmanthus-ekstrakt kan ud\u00f8ve antitumoraktivitet ved at h\u00e6mme ekspressionen af MMP-2 og MMP-9 i PI3K Akt-signalvejen, undertrykke spredningen af humane melanomceller og fremkalde deres apoptose. De eksperimentelle resultater p\u00e5 celleniveau i denne unders\u00f8gelse viste, at totale flavonoider fra Chrysanthemum morifolium signifikant h\u00e6mmede spredningen af A549-celler, fremmede celleapoptose og h\u00e6mmede cellemigration. P\u00e5 mRNA-niveau blev det bekr\u00e6ftet, at de samlede flavonoider fra Chrysanthemum morifolium opregulerede udtrykket af Bax og nedregulerede udtrykket af Bcl-2 og MMP-2.
