I \u00f8jeblikket er der rapporteret om over 400 arter. Tidligere unders\u00f8gelser har vist, at svampe, der tilh\u00f8rer sl\u00e6gten Trichoderma, kan producere sekund\u00e6re metabolitter med forskellige strukturelle typer, blandt hvilke celleafslappende midler og azafilonforbindelser er de vigtigste typer. Samtidig har sekund\u00e6re metabolitter fra Trichoderma-svampe en bred vifte af biologiske aktiviteter, s\u00e5som antibakterielle, antiinflammatoriske, cytotoksiske osv. Vores forskningsgruppe har l\u00e6nge v\u00e6ret engageret i opdagelsen af sekund\u00e6re metabolitter og deres biologiske aktiviteter i svampe af sl\u00e6gten Trichoderma. Vi har isoleret forskellige typer aktive stoffer med forskellige strukturelle typer fra sekund\u00e6re metabolitter af Trichoderma, s\u00e5som C. cochlides, C. convolutum, C. globorum CIB-160 og C. gracile. C. funicola er et potentielt patogen, som er blevet fundet i inficeret hudv\u00e6v. I 1969 blev der rapporteret om en makrocyklisk laktonforbindelse isoleret fra Trichoderma lucidum. I 1987 isolerede Itoh et al. det fra C Funicola JS 525 isolerede ubiquinonforbindelser. P\u00e5 nuv\u00e6rende tidspunkt er der kun to forskningsrapporter om de sekund\u00e6re metabolitter i tovv\u00e6rksh\u00e5rskallen, og der mangler relevante forskningsrapporter om biologisk aktivitet. Denne artikel unders\u00f8gte systematisk de sekund\u00e6re metabolitter fra en stamme af Trichoderma funicola CIB-604 og testede de antibakterielle og cytotoksiske aktiviteter af nogle forbindelser. Her vil vi rapportere om strukturen og den biologiske aktivitet af de sekund\u00e6re metabolitter, der produceres af denne bakterie.<\/p>\n<p>I denne unders\u00f8gelse blev otte sekund\u00e6re metabolitter isoleret fra faststoffermentering af ris af en stamme af Aspergillus niger (C. funicola CIB-604), herunder tre isoquinolinalkaloider (1-3) og fem sterolforbindelser (4-8), som alle blev isoleret fra Aspergillus niger for f\u00f8rste gang. Forbindelserne 1-3 var de f\u00f8rste strukturelt nye quinolinalkaloider, som vores forskningsgruppe opdagede fra de sekund\u00e6re metabolitter af C. indica 3.202 i Indien. Indtil videre er Aspergillus niger den anden svamp, der er i stand til at producere s\u00e5danne nye kinolinalkaloider, hvilket tyder p\u00e5, at den kan indeholde den samme kinolinalkaloid-biosyntesevej som C. indica. Denne artikel udf\u00f8rte antibakterielle og cytotoksiske aktivitetstest p\u00e5 forbindelserne 1-6. Resultaterne af den antibakterielle aktivitet viste, at forbindelserne 1-6 ved en koncentration p\u00e5 100 \u03bc g\/mL ikke udviste h\u00e6mmende virkninger p\u00e5 Escherichia coli, Bacillus subtilis og Candida albicans; Resultaterne af den cytotoksiske aktivitet viste, at forbindelserne 2 og 4 udviste cytotoksisk aktivitet mod humane tumorceller SK-Hep-1, A549, HCT-116 og MCF-7, og at forbindelsen 4 havde en st\u00e6rkere h\u00e6mmende virkning p\u00e5 tumorceller end forbindelsen 2; Resultaterne af flowcytometrianalyse viste, at forbindelser 2 og 4 hovedsageligt inducerede tidlig apoptose i SK-Hep-1-celler, mens forbindelse 2 havde ringe effekt p\u00e5 tidlig og sen apoptose i A549-celler, mens forbindelse 4 hovedsageligt inducerede sen apoptose i A549-celler. Denne artikel rapporterer for f\u00f8rste gang den cytotoksiske aktivitet af forbindelse 2 p\u00e5 tumorceller. Analyseresultaterne viser, at forbindelse 3 er en diastereomer af 2, men dens cytotoksiske aktivitet er signifikant lavere end forbindelse 2, hvilket indikerer, at den relative konfiguration mellem hydroxyl- og isopropylgrupper har en betydelig indvirkning p\u00e5 dens cytotoksiske aktivitet. Sammenlignet med forbindelserne 5 og 6 udviser forbindelse 4 betydelig cytotoksisk aktivitet. Analyse af dets strukturelle egenskaber afsl\u00f8rer et typisk aktivt center - \u03b1, \u03b2 - um\u00e6ttet carbonylstrukturenhed - i den molekyl\u00e6re struktur af forbindelse 4, som kan gennemg\u00e5 alkyleringsreaktion med thiolgrupperne i proteiner i celler og derved binde sig til tumorceller og dr\u00e6be dem.
