If\u00f8lge forskningsdata var antallet af diabetespatienter p\u00e5 verdensplan i 2017 851 millioner, og mere end 90% af dem var type 2-diabetes (T2DM). Derfor unders\u00f8ger dette eksperiment dets hypoglyk\u00e6miske effekt og relaterede handlingsveje ved at udvinde de samlede alkaloider af Sophora alopecuroides med henblik p\u00e5 at udnytte den medicinske ressourcev\u00e6rdi af Sophora alopecuroides fuldt ud og give en ny m\u00e5de at t\u00e6nke p\u00e5 til forskning i l\u00e6gemidler mod diabetes.<\/p>\n<p>Type 2-diabetes (T2DM) er en kompleks kronisk forstyrrelse af glukosemetabolismen, og dens forekomst har v\u00e6ret stigende. De samlede alkaloider fra Sophora alopecuroides er alkaloider udvundet fra l\u00e6geplanten Sophora alopecuroides, som er det generelle navn p\u00e5 en r\u00e6kke alkaloider, herunder sophocarpine, sophoridine, matrine osv. Derfor blev der i dette eksperiment udvundet totale alkaloider fra de overjordiske dele af Sophora alopecuroides for at unders\u00f8ge dens hypoglyk\u00e6miske effekt.<br \/>\nMembrantranslokationen af GLUT4-celler i kroppen p\u00e5virkes af flere signalveje, og en vigtig vej, der p\u00e5virker GLUT4-translokationen, er den insulinmedierede signaleffektvej. Insulin kan binde sig til insulinreceptoren (IR) og regulere glykogensyntese, glukoseoptagelse og nedbrydning gennem phosphatidylinositol-3-kinase\/proteinkinase B (PI3K\/AKT)-vejen og derved ud\u00f8ve en regulerende effekt p\u00e5 blodsukkeret. Dette eksperiment fokuserede p\u00e5 L6-celler og fandt ingen signifikant \u00e6ndring i AKT-phosphoryleringsniveauer efter TASA-behandling, hvilket indikerer, at TASA's hypoglyk\u00e6miske effekt ikke p\u00e5virker AKT-vejen. Skeletmusklers glukoseoptagelse kan ogs\u00e5 aktiveres og udnyttes gennem den insulinuafh\u00e6ngige AMPK-vej. Mekanismen er, at AMPK-aktivering for\u00e5rsager fosforylering af TBC1D1, som igen opregulerer udtrykket af downstream-signalmolekylet GLUT4 og fremmer dets transport fra cellen til cellemembranen, hvilket \u00f8ger absorptionen og udnyttelsen af glukose i cellen.
