Nogle studier fors\u00f8ger at belyse forholdet mellem oxidativ stress, produktion\/frigivelse af adipocytokiner, hepatisk insulinresistens, fedtophobning og aktivering af det medf\u00f8dte immunsystem i patogenesen af NAFLD. Ved at integrere bioinformatisk analyse og forskellige databaseinformationssystemer unders\u00f8ges den farmakologiske mekanisme for totale flavonoider i sorte gojib\u00e6r til behandling af ikke-alkoholisk fedtleversygdom, hvilket giver nye metoder og ideer til yderligere belysning af den terapeutiske mekanisme for NAFLD og den farmakologiske mekanisme for flavonoider i sorte gojib\u00e6r.<\/p>\n<p>Oxidativt stress og inflammation f\u00f8rer til NAFLD for\u00e5rsaget af hypertriglycerid\u00e6mi. Sorte gojib\u00e6r indeholder forskellige flavonoider som f.eks. vandopl\u00f8selige antioxidanter og anthocyaniner. Forskning har vist, at anthocyaniner fra sorte gojib\u00e6r kan reducere ophobningen af triglycerider i leveren betydeligt, mindske inflammation, \u00f8ge GSH Px-aktiviteten og s\u00e6nke MDA-niveauet. Det kan reducere produktionen af ROS og inflammation, \u00f8ge oxidationen af fedtsyrer i leveren og reducere syntesen af fedtsyrer og dermed forbedre NAFLD, der er fremkaldt af fedtfattig kost.<br \/>\nI denne unders\u00f8gelse downloadede vi NAFLD-mikroarrays fra GEO-databasen og sammenlignede genekspressionsprofilerne mellem NAFLD og normale kontroller og opn\u00e5ede 1741 opregulerede differentielt udtrykte gener og 1634 nedregulerede differentielt udtrykte gener. Der blev konstrueret et WGCNA-genkospressionsnetv\u00e6rk, og 24 n\u00f8glegener til behandling af NAFLD med totale flavonoider fra Lycium barbarum blev opn\u00e5et ved at krydse med de opn\u00e5ede m\u00e5l for aktive ingredienser. Proanthocyanidin B2 regulerer negativt genekspressionen af NLRP3.<br \/>\nVed at analysere sammenh\u00e6ngen mellem Hub-gener og immunceller fandt vi ud af, at Hub-gener er t\u00e6t forbundet med immunceller som makrofager M1-celler, makrofager M2-celler, monocytter, gamma delta T-celler, aktiverede mastceller, hvilende mastceller, umodne B-celler og hvilende CD4+memory T-celler. Forskning har vist, at lipidinduceret levercellestimulering udl\u00f8ser frigivelse af ekstracellul\u00e6re vesikler, der indeholder tumornekrosefaktorrelateret apoptoseinducerende ligand (TRAIL), som igen aktiverer makrofager til at producere IL-1 \u03b2 og IL-6.
