Svampenes forskelligartede levesteder og primitive evolution\u00e6re status g\u00f8r det muligt for dem at producere og akkumulere et stort antal strukturelt nye sekund\u00e6re metabolitter med forskellige biologiske aktiviteter som antitumor, antiparasit\u00e6re, antivirale, antibakterielle, antiinflammatoriske og immunregulerende. De er den prim\u00e6re kilde til marine naturprodukter. Det Sydkinesiske Hav er stort og et af de omr\u00e5der i verden, hvor svampe er koncentreret. I mange \u00e5r har svampe fra Det Sydkinesiske Hav v\u00e6ret et varmt emne inden for studiet af marine naturprodukters kemi. Allerede i 1933 begyndte professor Bergmann fra Yale University i USA at studere den kemiske sammens\u00e6tning af svampe og fik steroidkomponenter fra dem. I 1951 isolerede han nukleosidforbindelser som spongothymidin og spongouridin fra svampen Phyllospongia sp; det senere antivirale l\u00e6gemiddel adenosin og kr\u00e6ftl\u00e6gemidlet cytarabin er syntetiske analoger af spongothimidin. Laulimalid, pelorusid A, salicylihalimid A, varioliner og dictyodendriner fra svampeforskning er indg\u00e5et i pr\u00e6kliniske studier som antitumorl\u00e6gemidler.<br \/>\nForskningsobjektet i denne artikel er Phyllospongia sp., en svamp, der tilh\u00f8rer Demospongiae-klassen, Dictyoceratida-ordenen og Spongidae-familien. P\u00e5 nuv\u00e6rende tidspunkt har mange forskningsgrupper i ind- og udland gennemf\u00f8rt en r\u00e6kke unders\u00f8gelser af denne svamp og har isoleret mange strukturelt nye sekund\u00e6re metabolitter, herunder sesquiterpener, steroider, diphenylethere, glycerolipider og ceramider. De fleste af dem indeholder unikke tetracykliske eller pentacykliske sesquiterpener og steroider af skalarantypen. Der er dog kun f\u00e5 rapporter om deres biologiske aktivitet. For bedre at kunne belyse de bioaktive komponenter i Phyllosponia sp. blev der i dette eksperiment anvendt forskellige moderne kromatografiske og spektroskopiske teknikker til at unders\u00f8ge de forbindelser, der blev isoleret fra petroleumsether- og ethylacetatdelene af ekstraktet, og til at screene for deres cytotoksiske aktivitet. Dette berigede sammens\u00e6tningen af Phyllosponia sp. og lagde grunden til udviklingen og udnyttelsen af aktive komponenter i denne svamp i fremtiden.<br \/>\nbillede<br \/>\nbillede<br \/>\nI denne artikel blev de kemiske bestanddele af Phyllospongia sp. indsamlet fra Xisha-\u00f8erne i Det Sydkinesiske Hav unders\u00f8gt. Otte steroider blev isoleret fra petroleumsether og dichlormethan for f\u00f8rste gang, og deres cytotoksicitet over for MCF-7 (humane brystkr\u00e6ftceller), HT-29 (humane tyktarmskr\u00e6ftceller) og Hep-2 (humane larynxkr\u00e6ftepitelceller) blev testet ved hj\u00e6lp af MTT-metoden. Blandt dem var forbindelserne 1, 3, 4, 6 og 8 svage over for humane tyktarmskr\u00e6ftceller HT-29 med IC50 st\u00f8rre end 83 \u03bc mol\/L; Forbindelserne 1, 2 og 7 viste signifikant cytotoksisk aktivitet over for MCF-7-celler med IC50-v\u00e6rdier p\u00e5 henholdsvis 8,8, 10,3 og 3,9 \u03bc mol\/L; Forbindelse 2 udviser god cytotoksisk aktivitet over for Hep-2 med en IC50 p\u00e5 10,6 \u03bc mol\/L. De eksperimentelle resultater viste, at den cytotoksiske aktivitet af forbindelse 7 var bedre end den positive kontrolmedicin cisplatin-gruppen. P\u00e5 grund af det lave indhold af denne forbindelse og manglen p\u00e5 litteratur, der rapporterer betydelig aktivitet, har den potentialet til at udvikle sig til et nyt antitumorl\u00e6gemiddel gennem strukturel modifikation eller organisk syntese.
