Tidligere rapporter har vist, at makrofager kan fungere som m\u00e5lceller for polysaccharider og ofte bruges som ideelle cellemodeller til at evaluere den immunmodulerende aktivitet af polysaccharider. Liu et al. fandt, at polysaccharider fra Panax ginseng-blomster kan forbedre makrofagernes immunaktivitet ved at \u00f8ge makrofagernes fagocytiske evne og frigive immunfaktorer. Derudover har forskning vist, at Ganoderma lucidum-polysaccharider og Poria cocos-polysaccharider ogs\u00e5 har en aktiverende effekt p\u00e5 makrofager.<\/p>\n<p>P\u00e5 nuv\u00e6rende tidspunkt fokuserer forskningen i polysakkariders biologiske aktivitet hovedsageligt p\u00e5 r\u00e5 polysakkarider, som har komplekse komponenter og ikke er befordrende for at belyse polysakkariders virkningsmekanisme. Den neutrale r\u00f8de test viste, at AHP-III kan forbedre den fagocytiske evne hos RAW264.7-celler, og der er en signifikant forskel inden for det eksperimentelle koncentrationsomr\u00e5de. RT qPCR- og ELISA-eksperimenter har vist, at AHP-III \u00f8ger ekspressionsniveauerne af TNF - \u03b1 og IL-6-gener og derved \u00f8ger frigivelsen af disse cytokiner. I ELISA-eksperimentet var frigivelsen af TNF - \u03b1-cytokiner i den blanke gruppe lavere end standardkurvens minimumst\u00e6rskel, og frigivelsen var relativt lille, hvilket resulterede i et beregnet resultat p\u00e5 0; RT qPCR-eksperimenter viste, at ekspressionsniveauet for iNOS-genet steg med stigningen i l\u00e6gemiddeldosering, og der var en signifikant forskel sammenlignet med den blanke gruppe; I mellemtiden blev det if\u00f8lge Griess-testen fundet, at NO-sekretionen i den behandlede gruppe steg signifikant sammenlignet med den blanke gruppe. Ovenst\u00e5ende eksperimentelle resultater indikerer, at AHP-III kan fremme cytokinfrigivelse og spille en immunmodulerende rolle ved at \u00f8ge ekspressionen af cytokingener.<\/p>\n<p>NF-KB har l\u00e6nge v\u00e6ret betragtet som en typisk pro-inflammatorisk signalvej, i h\u00f8j grad baseret p\u00e5 aktivering af pro-inflammatoriske cytokiner som interleukin og tumornekrosefaktor, som kan formidle inflammatoriske reaktioner gennem makrofager. RAW264.7-makrofager stimuleres af inflammatoriske faktorer som TNF-\u03b1 og IL-6, som aktiverer IKK-kinase i NF-KB-signalvejen. Fosforyleret IKK kan fosforylere IkBa og frigive NF-KB-dimerer, hvilket \u00f8ger fosforyleringsniveauet af NF-KB-signalvejsrelaterede proteiner. Resultaterne af Western Blot-eksperimentet viste, at efter AHP-III-stimulering af RAW264.7-makrofager blev fosforyleringsniveauerne af p65, IKK \u03b2 og IkB \u03b1 i NF-KB signifikant \u00f8get.
