Merr. De har en st\u00e6rk milj\u00f8m\u00e6ssig tilpasningsevne, t\u00f8rkeresistens og tolerance over for ufrugtbarhed og kan plantes de fleste steder, selv i saltholdig alkalisk jord. Sorte b\u00f8nner har et bredt planteomr\u00e5de og en relativt koncentreret regional udbredelse i Kina. De plantes ofte i Heilongjiang, Anhui, Shanxi og andre steder med rigelige ressourcer og forskellige typer. Sorte b\u00f8nner er et ber\u00f8mt traditionelt l\u00e6gemiddel og spiseligt landbrugsprodukt, der er rigt p\u00e5 um\u00e6ttede fedtsyrer, proteiner, vitaminer, polysaccharider, isoflavoner og andre bioaktive stoffer. Fr\u00f8skallen p\u00e5 sorte b\u00f8nner er sortr\u00f8d eller purpurr\u00f8d, skinnende, tynd og sk\u00f8r, g\u00e5r let i stykker og indeholder forskellige aktive stoffer som kostfibre, vitaminer, pigmenter, polysaccharider, flavonoider osv. Anthocyaninerne i fr\u00f8skallen p\u00e5 sorte b\u00f8nner giver ikke kun sorte b\u00f8nner farve, men har ogs\u00e5 en r\u00e6kke sundhedsm\u00e6ssige virkninger p\u00e5 menneskekroppen, s\u00e5som anti-oxidation, tarmberoligelse, leverbeskyttelse, forebyggelse af diabetes, blodsukker, blodlipider, forbedring af hukommelsen, synsbeskyttelse, antitumor, forebyggelse og behandling af astma osv.<br \/>\nDen gradvise forbedring af teknologien til udvinding af anthocyaniner har skabt et grundlag for den industrielle anvendelse, s\u00e5 den kan bruges i vid udstr\u00e6kning i industrier som ny kosmetik, sundhedspleje, f\u00f8devarer og naturlige farvestoffer. De traditionelle metoder til udvinding af anthocyaniner fra sorte b\u00f8nners skind omfatter prim\u00e6rt vandekstraktion og ekstraktion med organiske opl\u00f8sningsmidler. Disse metoder har dog ulemper som lang ekstraktionstid, h\u00f8jt opl\u00f8sningsmiddelforbrug, lav ekstraktionshastighed og d\u00e5rlig stabilitet af anthocyaniner. I de senere \u00e5r er metoder som ultralydsassisteret ekstraktion, mikrob\u00f8lgeassisteret ekstraktion og ionisk v\u00e6skeekstraktion blevet udforsket i unders\u00f8gelsen af anthocyaniner i sorte b\u00f8nners fr\u00f8kappe p\u00e5 grund af deres egenskaber ved kort ekstraktionstid og h\u00f8jere ekstraktionshastighed. Tidligere unders\u00f8gelser har vist, at den enzymatisk assisterede ekstraktion af bioaktive stoffer som anthocyaniner har fordelene ved lavt energiforbrug, h\u00f8j omkostningseffektivitet og h\u00f8j stabilitet og aktivitet af de ekstraherede produkter. Der har dog ikke v\u00e6ret nogen rapporter om enzymatisk ekstraktion af anthocyaniner fra skind af sorte b\u00f8nner. Denne unders\u00f8gelse anvendte enzymatisk assisteret ekstraktion af anthocyaniner fra sorte b\u00f8nneskind og optimerede ekstraktionsprocessen af anthocyaniner ved hj\u00e6lp af responsoverflademetodologi med anthocyaninindhold som indikator. Samtidig blev der udf\u00f8rt in vitro-eksperimenter for at analysere antioxidantaktiviteten af ekstraktet af anthocyaniner fra sorte b\u00f8nneskind for at give teoretiske og tekniske referencer til udvinding og omfattende udvikling og udnyttelse af anthocyaniner fra sorte b\u00f8nneskind.<\/p>\n<p>I dette eksperiment blev ethanol brugt som ekstraktionsopl\u00f8sning for at optimere processen med at udvinde anthocyaniner fra fr\u00f8skallen p\u00e5 sorte b\u00f8nner ved hj\u00e6lp af en bioenzymassisteret metode. Den enzymatisk assisterede udvinding af planteaktive ingredienser har de fordele, at den er gr\u00f8n, forureningsfri, ugiftig og uskadelig. Den bygger hovedsageligt p\u00e5 det faktum, at de mest effektive aktive ingredienser findes i plantecellernes cytoplasma og kan nedbrydes biologisk af cellulaser, alfa-amylase og andre enzymer p\u00e5 cellev\u00e6gscellulose for at opn\u00e5 m\u00e5let om at nedbryde cellev\u00e6ggens t\u00e6tte struktur, \u00f8ge cellev\u00e6ggens permeabilitet og intercellul\u00e6re barrierer og maksimere diffusionen og frigivelsen af effektive ingredienser fra cellecytoplasmaet til ekstraktionsmediet og dermed effektivt udvinde bioaktive stoffer.<\/p>\n<p>Baseret p\u00e5 enkeltfaktoreksperimenter udledes det optimale ekstraktionsskema gennem optimering af responsoverfladen og valideres. Resultaterne viste, at regressionen af responsoverflademodellen var meget signifikant (P0,05). Modellen kan bruges n\u00f8jagtigt til at forudsige m\u00f8nsteret af \u00e6ndringer i indholdet af anthocyaniner udvundet fra sorte b\u00f8nneskind med forskellige eksperimentelle faktorer. De optimale procesbetingelser opn\u00e5et gennem optimering er: sammensat enzym (cellulase 400U\/g+\u03b1 - amylase 50U\/g), enzymatisk hydrolysetemperatur p\u00e5 50 \u00b0C, v\u00e6ske\/materiale-forhold p\u00e5 26:1 ml\/g, ethanolvolumenfraktion p\u00e5 64% og enzymatisk hydrolysetid p\u00e5 59 minutter. Under disse fors\u00f8gsbetingelser var det ekstraherede anthocyaninindhold 2,019 mg\/g.<br \/>\nIndholdet af anthocyaniner er 2,019 mg\/g. Forel\u00f8bige unders\u00f8gelser af antioxidantaktiviteten af anthocyaniner i sort sojab\u00f8nnes fr\u00f8kappe indikerer, at anthocyaniner i sort sojab\u00f8nnes fr\u00f8kappe har god antioxidantaktivitet og udviser et dosis-respons-forhold. Inden for fors\u00f8gsomr\u00e5det er dens reduktionsevne og evne til at fjerne superoxidanionradikaler lidt lavere end ascorbinsyre, men dens evne til at fjerne nitritioner, DPPH-radikaler og ABTS-radikaler er st\u00e6rkere end ascorbinsyre, hvilket er i overensstemmelse med forskningsresultaterne fra Xu, Wu og andre. Dannelsen af anthocyaniners antioxidantkapacitet skyldes deres konjugerede struktur, og anthocyaninernes stabilitet og reaktivitet p\u00e5virkes af graden og placeringen af hydroxylering og methylering p\u00e5 B-ringen, hvilket igen p\u00e5virker anthocyaninernes antioxidantaktivitet. Forskning har vist, at cyanidin-3-O-glucosid (C3G) er hovedbestanddelen af anthocyaniner i sorte b\u00f8nners skind, med phenoliske hydroxylgrupper i strukturen. Det kan fungere som hydroxyldonor, fange og binde frie radikaler til at danne en stabil struktur, afslutte forekomsten af frie radikale k\u00e6dereaktioner og udvise antioxidantkapacitet.<br \/>\nEksperimenter har vist, at anthocyaniner i fr\u00f8skallen fra sorte b\u00f8nner har h\u00f8j medicinsk v\u00e6rdi og brede anvendelsesmuligheder. Den enzymatisk assisterede ekstraktion af naturlige aktive ingredienser er imidlertid en kompleks fast-v\u00e6ske-faseoverf\u00f8rselsproces, og opl\u00f8sningen af mere cellul\u00e6rt indhold kan f\u00f8re til samtidig indf\u00f8ring af flere urenheder i r\u00e5ekstrakten. Indholdet af anthocyaniner i fr\u00f8skallen af sorte b\u00f8nner er relativt h\u00f8jt, og dets r\u00e5ekstrakt af anthocyaniner udviser en vis antioxidantkapacitet, hvilket har potentiale til at udvikle nye anthocyaninressourcer. Anthocyaniner er ikke de eneste antioxidanter i fr\u00f8skallen fra sorte b\u00f8nner.
