Forskning har vist, at d\u00f8deligheden for livmoderhalskr\u00e6ftpatienter i Kina stiger med 3,35% om \u00e5ret, med en 10-\u00e5rig v\u00e6kstrate p\u00e5 29,9%, og tendensen til forekomst blandt unge bliver stadig tydeligere. CSC'er er en lille population, der findes i tumormikromilj\u00f8et, og forskning har rapporteret, at stamceller fra livmoderhalskr\u00e6ft (CCSC'er) kan identificeres og isoleres fra livmoderhalskr\u00e6ftceller. Kr\u00e6ftstamceller er uf\u00f8lsomme over for de fleste kemoterapimidler og er hoved\u00e5rsagen til tilbagefald af kr\u00e6ft hos patienter i helbredelsesfasen, hvilket alvorligt p\u00e5virker patienternes 2-5-\u00e5rige overlevelsesrate efter str\u00e5lebehandling og kemoterapi. Derfor har s\u00f8gning og screening efter nye strukturer, h\u00f8j anticanceraktivitet, lav toksicitet og tumorstamcelleh\u00e6mmende aktivitet af blyforbindelser mod livmoderhalskr\u00e6ft fra naturlige aktive ingredienser betydelig forskningsm\u00e6ssig betydning i innovativ l\u00e6gemiddelforskning.<\/p>\n<p>Lakrids, som er en medicinsk og spiselig ressource, har en lang historie med medicinsk brug i Kina. Selv om der findes forskellige lakridssorter, omfatter den kinesiske farmakop\u00e9 (2020-udgaven, del 1) to oprindelige plantearter, nemlig Glycyrrhiza inflata Bat. og Glycyrrhiza glabra L., hvoraf Glycyrrhiza inflata L. er vidt udbredt og har rigelige ressourcer i det sydlige Xinjiang, Gansu og andre nordvestlige regioner. Den kemiske struktur af chalconer i forskellige lakridssorter varierer. Forskning har bekr\u00e6ftet, at licochalcone A (LicoA) har antitumor-, antibakterielle, antiinflammatoriske, hepatobeskyttende og immunmodulerende virkninger, men der er kun f\u00e5 rapporter om dets regulering af livmoderhalskr\u00e6ftrelaterede tumorstamcellemark\u00f8rer. P\u00e5 baggrund af indledende grundforskning har vores forskningsgruppe forbedret et traditionelt hospitalspr\u00e6parat til behandling af cervikal erosion ved at tils\u00e6tte LicoA, der indeholder lakridsekstrakt. Vi er i gang med at udvikle vaginale brusetabletter, der har potentiale til at forebygge livmoderhalskr\u00e6ft og kan bruges til vaginal antibakteriel, antiinflammatorisk og cervikal erosionsbehandling. I screeningen af naturlige aktive ingrediensers anticanceraktivitet blev det konstateret, at LicoA, en komponent i lakridschalcone, har betydelig aktivitet i livmoderhalskr\u00e6ftcellers spredning, apoptose og cellecyklus. Derudover har det betydelige regulerende effekter p\u00e5 mark\u00f8rer for livmoderhalskr\u00e6fttumorstamceller (Bcl-2, ALDH1A1, OCT-4, UHRF1, BIRC7, BIRC5) gener og cyclinafh\u00e6ngige kinase 4 (CDK4) gener. Ved at bruge computerst\u00f8ttet l\u00e6gemiddeldesign (CADD) og andre metoder til at forudsige dets mulige m\u00e5l blev effekten af LicoA p\u00e5 mRNA-ekspressionsniveauerne af ovenn\u00e6vnte gener unders\u00f8gt, hvilket gav en mere omfattende reference til yderligere at belyse LicoA's anticanceraktivitet p\u00e5 livmoderhalskr\u00e6ftceller og udforske dets mulige molekyl\u00e6re mekanismer.<\/p>\n<p>P\u00e5 nuv\u00e6rende tidspunkt er kirurgisk resektion stadig den vigtigste behandling af livmoderhalskr\u00e6ft i klinisk praksis, kombineret med kemoterapi og str\u00e5lebehandling som adjuverende terapi. Forslaget om tumorstamcelleteorien har givet en ny fortolkning af forekomsten, udviklingen og behandlingen af livmoderhalskr\u00e6fttumorer. Forskning har bekr\u00e6ftet, at 11 tumorstamceller ikke kun er grund\u00e5rsagen til tumorcellernes tolerance over for kemoradioterapi, men ogs\u00e5 en af \u00e5rsagerne til tumorcellernes metastase og tilbagefald. Derfor er det en ny forskningsretning at bruge naturlige aktive ingredienser til sikkert og effektivt at gribe ind i tumorstamceller og derved reducere muligheden for livmoderhalskr\u00e6ft og forhindre tilbagefald af livmoderhalskr\u00e6ft efter behandling.<br \/>\nNuv\u00e6rende forskning har bekr\u00e6ftet, at lakrids har anti-tumor, antibakterielle, anti-inflammatoriske, hepatobeskyttende og immunmodulerende virkninger. Xinjiang-regionen er rig p\u00e5 lakridsressourcer, og lakridschalcone A findes hovedsageligt i r\u00f8dder og st\u00e6ngler. Vores forskningsgruppe udviklede og etablerede et detektions- og identifikationssystem for chalcone A i opsvulmet lakrids under den indledende forskning og ans\u00f8gte om et brugsmodelpatent relateret til instrumentudstyr (patentnummer: ZL201721685479.2). Baseret p\u00e5 dette blev LicoA-komponenterne udvundet og fremstillet. Mekanismen for LicoA's antitumoreffekt kan v\u00e6re ved at inducere tumorcelleapoptose, men der er relativt lidt forskning i dens regulerende effekt p\u00e5 livmoderhalskr\u00e6fttumorstamcellemark\u00f8rer. Resultaterne af denne unders\u00f8gelse viste, at Xinjiang swollen fruit licorice chalcone A har en signifikant h\u00e6mmende effekt p\u00e5 spredningen af livmoderhalskr\u00e6ftceller og udviser koncentrations- og tidsafh\u00e6ngige effekter. Det kan fremkalde apoptose i SiHa-celler og kan blokere cellecyklussen i S- og G2\/M-faserne og har en betydelig nedreguleringseffekt p\u00e5 livmoderhalskr\u00e6fttumorstamcellemark\u00f8rer.<\/p>\n<p>Under normale omst\u00e6ndigheder danner retinoblastom (Rb) et kompleks med E2F-transkriptionsfaktorer i G1-fasen af cellecyklussen og h\u00e6mmes. CDK4 kan fosforylere Rb-komplekset, hvilket f\u00f8rer til frigivelse af E2F for at transkribere de n\u00f8dvendige gener for at komme ind i S-fasen af cellecyklussen fra G1-fasen. Uden denne fosforyleringsaktivitet blokeres cellerne i G1-fasen. I denne unders\u00f8gelse blev CDK4-genet signifikant h\u00e6mmet ved koncentrationer p\u00e5 25 og 50 \u03bcg\/mL af glycyrrhizinsyre-chalkon A. Udtrykket af stamcellemark\u00f8rerne ALDH1A1 og OCT4 er opreguleret i livmoderhalskr\u00e6ft og dens forstadier, is\u00e6r i perifert blod, hvilket tyder p\u00e5, at ALDH1A1 og OCT4 kan bruges som biomark\u00f8rer til tidlig p\u00e5visning af livmoderhalskr\u00e6ft eller overv\u00e5gning af patientbehandling. ALDH1A1 er involveret i oxidationen af retinol til retinsyre i cytoplasmaet, som derefter overf\u00f8res til kernen og initierer transkription af gener, der er involveret i tidlig stamcelledifferentiering, hvilket fremmer tumorstamcelleproliferation. Octomer binding protein 4 (OCT-4) er en transkriptionsfaktor, der udtrykkes i embryonale og voksne stamceller, og som er forbundet med celleproliferation, differentiering, pluripotens og selvfornyelse. Hvis man sl\u00e5r OCT-4 fra i embryonale stamceller, f\u00f8rer det til differentiering og tab af stamcelleegenskaber. Glycyrrhizinsyre-chalkon A kan ogs\u00e5 nedregulere mRNA-ekspressionsniveauerne for de to ovenn\u00e6vnte mark\u00f8rer (ALDH1A1, OCT4) betydeligt. Derudover er de udvalgte tumormark\u00f8rer Bcl-2, UHRF1, BIRC7, BIRC5 osv. i denne unders\u00f8gelse alle st\u00e6rkt udtrykt i livmoderhalskr\u00e6ftceller, og deres mRNA-ekspressionsniveauer nedreguleres ogs\u00e5 i varierende grad af glycyrrhizinsyre-chalkon A.
