Gr\u00f8nne morb\u00e6r, ogs\u00e5 kendt som gule morb\u00e6r, henviser til umodne morb\u00e6r. Som traditionel kinesisk medicin er gr\u00f8nne morb\u00e6r lettere at plukke, transportere og opbevare end modne morb\u00e6r, og deres indhold af anthocyaniner, catechiner og epicatechiner er meget h\u00f8jere end i modne morb\u00e6r. Der har v\u00e6ret unders\u00f8gelser b\u00e5de herhjemme og internationalt om udvinding og isolering af anthocyaniner fra forskellige planter som f.eks. vindruekerner, roser, majssilke, traneb\u00e6r, kakaob\u00f8nner osv. Liu et al. brugte LSA-10 harpiks til at berige og rense anthocyaniner fra hvidtj\u00f8rn; Chen Wenliang et al. brugte membranseparation og filtrering til at adskille oligomere anthocyaniner fra vindruekerner, hvilket resulterede i et indhold af 65% oligomere anthocyaniner, men opn\u00e5ede ikke monomerseparationskomponenter; Glavnik et al. analyserede anthocyaninerne i japansk tigerr\u00f8r ved hj\u00e6lp af h\u00f8jtydende tyndlagskromatografi og identificerede monomerer, decamerer og anthocyaniner af B-typen fra dets r\u00e5ekstrakt. Denne metode er imidlertid vanskelig at forberede; Sun et al. brugte semiforberedende h\u00f8jtydende v\u00e6skekromatografi til at adskille anthocyaniner fra druekerner, men forbehandlingen var kompleks, og adskillelsesudbyttet var lavt; Denne artikel v\u00e6lger h\u00f8jhastigheds modstr\u00f8mskromatografi (HSCCC), som har en stor adskillelseskapacitet, kan bruges til batchforberedelse og adskillelse, er let at betjene og har h\u00f8j adskillelseseffektivitet.<br \/>\nProcyanidiner (PC), som er en type polyfenoliske forbindelser, findes i stor udstr\u00e6kning i planter. De best\u00e5r hovedsageligt af catechiner, epicatechiner (EC), epigallocatechiner (EGC) og epicatechingallater (ECG) og har en st\u00e6rk antioxidantaktivitet. Dette arbejde har til form\u00e5l at bruge h\u00f8jhastigheds modstr\u00f8mskromatografi til at adskille og oprense anthocyaniner fra gr\u00f8nne morb\u00e6r. H\u00f8jtydende v\u00e6skekromatografi (HPLC) vil blive brugt til at identificere de kemiske komponenter i hver komponent og analysere deres renhed; ved at unders\u00f8ge rensningsevnen for DPPH-radikaler, ABTS+-radikaler og hydroxylradikaler i hver komponent kan styrken af deres antioxidantkapacitet afspejles. Dette arbejde udvider omfanget af dyb forarbejdning og udnyttelse af morb\u00e6r, giver retning for den industrielle produktion af anthocyaniner i gr\u00f8nne morb\u00e6r og har en vis referencev\u00e6rdi for udviklingen og udnyttelsen af morb\u00e6r som medicinske ressourcer.<\/p>\n<p>\u200d<\/p>\n<p>Med gr\u00f8n morb\u00e6r som r\u00e5materiale blev r\u00e5ekstraktet opn\u00e5et gennem ekstraktion, affedtning, fjernelse af urenheder og ekstraktionsberigelse under de eksperimentelle forhold. Det optimale opl\u00f8sningsmiddelsystem til HSCCC-separation og -oprensning var n-hexan-ethylacetatvand (1:20:20, V\/V\/V). 200 mg r\u00e5ekstrakt af anthocyaniner fra gr\u00f8nne morb\u00e6r blev adskilt i fem komponenter ved hj\u00e6lp af HSCCC, nemlig F1 (28,5 mg), F2 (10,43 mg), F3 (6,85 mg), F4 (10,65 mg) og F5 (13,65 mg). Hver komponent indeholdt catechiner, anthocyanin B2, anthocyanin A1 osv. Blandt dem indeholdt F4 epigallocatechingallat med en renhed p\u00e5 95,05%; F5 indeholder catechiner med en renhed p\u00e5 73,75%. De adskilte komponenter har alle en vis antioxidantaktivitet. Med Vc som kontrol er evnen til at fjerne frie DPPH-radikaler som f\u00f8lger: Vc&gt;F5&gt;F4&gt;F2&gt;F3&gt;F1&gt; r\u00e5ekstrakt af gr\u00f8nne morb\u00e6r; ABTS+-fri radikalopfangningsevne: Vc&gt;F5&gt;F4&gt;F2&gt;F3&gt;F1&gt; r\u00e5ekstrakt af gr\u00f8n morb\u00e6r; Hydroxylradikal rensningsevne: F1&gt;F5&gt;Vc&gt;F4&gt;F2&gt; r\u00e5ekstrakt af gr\u00f8nne morb\u00e6r&gt;F3. HSCCC har en stor forberedelseskapacitet og intet pakkemateriale i den kromatografiske s\u00f8jle. I dette eksperiment kan r\u00e5ekstraktet adskilles en gang for at opn\u00e5 fem komponenter, og nogle komponenter har monomere stoffer med h\u00f8j renhed og st\u00e6rk antioxidantaktivitet.
