Genetik, fedme, manglende motion og rygning er risikofaktorer for at udvikle tyktarmskr\u00e6ft. P\u00e5 nuv\u00e6rende tidspunkt er det hovedsageligt kirurgisk behandling, der anvendes i klinisk praksis ved tyktarmskr\u00e6ft. Vi fandt gennem flowcytometrianalyse, at acetylrhamnosid dosisafh\u00e6ngigt kan p\u00e5virke cellecyklussen i tyktarmskr\u00e6ft HCT116-celler, hvilket for\u00e5rsager cellecyklusstop i G1-fasen og \u00f8ger celleapoptosehastigheden betydeligt.<\/p>\n<p>Forskning har bekr\u00e6ftet, at Wnt-signalvejen er st\u00e6rkt korreleret med udviklingen af tyktarmskr\u00e6ft. Blandt dem er \u03b2 - catenin et n\u00f8glemolekyle i den klassiske Wnt-vej, og E-cadherin\/\u03b2 - catenin-komplekset hj\u00e6lper med at stabilisere intercellul\u00e6r adh\u00e6sion og derved reducere celleinvasion og metastase. I tumorceller kan \u03b2 - catenin aktivere gentranskription relateret til kr\u00e6ftcelleproliferation, cyklus, apoptose, migration, invasion og l\u00e6gemiddelresistens, hvilket f\u00f8rer til ondartet spredning af tumorer. Desuden har \u03b2-catenin vist sig at v\u00e6re et lovende m\u00e5l for kr\u00e6ftforebyggelse og -behandling. Mange naturprodukter kan fungere som h\u00e6mmere af \u03b2-catenin-signalering, prim\u00e6rt gennem fosforylering, ubiquitinering og h\u00e6mning af dets nukleare translokation. Naturprodukth\u00e6mmere har vist gode forebyggende og terapeutiske effekter i forskellige tumormodeller in vitro og in vivo. \u03b2-catenin kan fremme ekspressionen af downstream-molekylet Myc i kernen. Myc tilh\u00f8rer onkogenfamilien og koder for essentielle nukleare transkriptionsfaktorer i kernen, hovedsageligt involveret i regulering af cellev\u00e6kst, proliferation, differentiering, cyklus, metabolisme, apoptose osv. Hos pattedyr omfatter Myc-familien af proteiner tre kategorier: c-Myc, n-Myc og l-Myc. c-Myc er forbundet med genomisk ustabilitet og tumorigenese. I hvilende celler er ekspressionsniveauet for c-Myc lavt, og n\u00e5r cellen g\u00e5r ind i cellecyklussen, \u00f8ges ekspressionen af c-Myc hurtigt flere gange. Derudover blev der fundet signifikant opregulering af c-Myc-protein i ca. 70% af tyktarmskr\u00e6ft, hvilket indikerer en positiv sammenh\u00e6ng mellem c-Myc og maligniteten af tyktarmskr\u00e6ft.<\/p>\n<p>I dette eksperiment blev high-throughput transkriptomsekventering brugt til at identificere 4129 forskelligt udtrykte gener i HCT116-celler induceret af acetylrhamnosid, herunder 1921 opregulerede gener og 2208 nedregulerede gener. De forskelligt udtrykte gener var hovedsageligt relateret til cellev\u00e6kst, cellecyklus, celleinvasion, celleimmunitet, oxidativ stress, cellemetabolisme, molekyl\u00e6r transport og celled\u00f8dsveje. Efter at have analyseret de forskelligt udtrykte gener i kombination med KEGG-databasen blev det konstateret, at acetylrhamnosid signifikant p\u00e5virkede Wnt-signalvejen i HCT116-celler. Den signifikante nedregulering af \u03b2-catenin og c-Myc blev bekr\u00e6ftet gennem genekspression og proteindetektion af acetylrhamnosid og kontrolgruppen. Kombinationen af acetylrhamnosid og Wnt-h\u00e6mmeren XAV939 h\u00e6mmede yderligere v\u00e6ksten af HCT116-tyktarmscancerceller, hvilket indikerer, at Wnt\/\u03b2-catenin-vejen ogs\u00e5 medierer HCT116's l\u00e6gemiddelresistens. H\u00e6mning af Wnt\/\u03b2 - catenin-stien kan \u00f8ge acetylrhamnosids antitumoraktivitet betydeligt.<\/p>\n<p>Denne unders\u00f8gelse viste, at acetylrhamnosid effektivt kan h\u00e6mme spredningsaktiviteten af tyktarmskr\u00e6ftcellelinjen HCT116.
