CPCC 203679, der stammer fra jord.<br \/>\nActinobakterier er en vigtig ressource for opdagelsen af nye antibiotika, da de kan producere en r\u00e6kke aktive naturprodukter med forskellige strukturelle typer. Streptomyces er den dominerende sl\u00e6gt af actinomyceter med det h\u00f8jeste antal rapporterede effektive arter. Den er vidt udbredt i jord, ferskvand, havmilj\u00f8er og nogle organismer. Streptomyces er kendt for at producere en lang r\u00e6kke aktive sekund\u00e6re metabolitter med forskellige strukturelle typer og er en vigtig kilde til almindeligt anvendte antibiotika i klinisk praksis. I \u00f8jeblikket produceres omkring to tredjedele af de antibiotika, der bruges i klinisk praksis, af denne sl\u00e6gt af mikroorganismer. I de senere \u00e5r er der l\u00f8bende blevet rapporteret om et stort antal strukturelt nye og biologisk aktive sekund\u00e6re metabolitter fra sl\u00e6gten Streptomyces, hvilket indikerer, at opdagelsen af nye hovedforbindelser fra denne sl\u00e6gt stadig har et stort potentiale. Vores forskningsgruppe opdagede en stamme af Streptomyces sp. CPCC 203679, der stammede fra jord, under den indledende screening af biologisk aktivitet. R\u00e5ekstrakten af dens fermenteringsbouillon har god antibakteriel aktivitet. I denne unders\u00f8gelse blev forskellige moderne kromatografiske og spektroskopiske teknikker brugt til forel\u00f8bigt at isolere, oprense og strukturelt identificere stammens sekund\u00e6re metabolitter, og der blev opn\u00e5et 9 forbindelser (se figur 1). Forbindelserne 1-8 blev evalueret for deres in vitro antibakterielle og cytotoksiske aktiviteter.<\/p>\n<p>&nbsp;<\/p>\n<p>Ni forbindelser blev isoleret fra g\u00e6ringsbouillon fra Streptomyces sp. CPCC 203679, der stammer fra jord, herunder fedtsyrer, alkaloider og cykliske dipeptider. Forbindelse 1 er en ny fedtsyreforbindelse, og forbindelse 3 blev isoleret for f\u00f8rste gang fra Streptomyces fra jorden. Resultaterne af evalueringen af den farmakologiske aktivitet viste, at forbindelserne 1-8 ikke udviste signifikant antibakteriel og antitumoraktivitet. Forbindelse 2 tilh\u00f8rer alkaloidklassen og er bredt til stede i mikroorganismer som actinomyceter, bakterier og svampe, s\u00e5som Streptomyces Bacillus\u3001Aspergillus\u3001Alternaria\u3002 Forbindelse 3 tilh\u00f8rer indolalkaloidklassen og blev f\u00f8rst rapporteret i 1965. Denne forbindelse er ogs\u00e5 blevet rapporteret i sl\u00e6gter som Bacillus, Balansia, Edwardsiella osv. If\u00f8lge litteraturrapporter har forbindelse 3 antiforureningseffekter og har en vis h\u00e6mmende effekt p\u00e5 larver af rankef\u00f8dder. Dens ED50 er 18,6 \u03bc mol\/L. Cykliske dipeptider er de mindste cykliske peptider i naturen og findes i vid udstr\u00e6kning i metaboliske produkter fra planter, dyr og mikroorganismer med aktiviteter som antibakterielle, antitumor-, neurobeskyttende og antitrombotiske egenskaber. I denne unders\u00f8gelse blev der ikke isoleret sekund\u00e6re metabolitter med betydelig biologisk aktivitet, hvilket kan skyldes, at de biosyntetiske genklynger for aktive metabolitter ikke blev aktiveret under de nuv\u00e6rende dyrkningsforhold, eller at de ikke blev isoleret p\u00e5 grund af lav produktion af aktive metabolitter. I fremtiden vil vi fors\u00f8ge at optimere dyrkningsbetingelserne for at \u00f8ge den strukturelle mangfoldighed af sekund\u00e6re metabolitter og yderligere kombinere evaluering af antibakteriel aktivitet for at opn\u00e5 strukturelt nye og signifikant aktive sekund\u00e6re metabolitter.<\/p>","protected":false},"excerpt":{"rendered":"<p>Study on secondary metabolites of soil derived Streptomyces sp.
