Svampe fra mangrover producerer nye og meget aktive sekund\u00e6re metabolitter p\u00e5 grund af deres unikke livsmilj\u00f8, og de har ydet et vigtigt bidrag til opdagelsen af vigtige stoffer som forskellige antiinflammatoriske l\u00e6gemidler, nye antibiotika, svampemidler og l\u00e6gemidler mod kr\u00e6ft. Aspergillus fumigatus er en saprofytisk n\u00e6ringssvamp, der hovedsageligt lever i jord. Det er den mest almindelige patogene bakterie af invasiv aspergillose, og dens sporer og nogle metaboliske komponenter overf\u00f8res let gennem luften for at for\u00e5rsage humane lungesygdomsinfektioner, s\u00e5som svampeallergisk astma, variant reaktiv bronkopulmonal aspergillose og invasiv pulmonal aspergillose. Aspergillus-sl\u00e6gten er udbredt over hele verden med stor overlevelsesevne, st\u00e6rk forplantningsevne og mange forskellige metabolitter. If\u00f8lge litteraturrapporter blev mere end 220 sekund\u00e6re metabolitter med god aktivitet isoleret fra Aspergillus niger fra 2006 til 2016. De vigtigste metabolitter omfatter polyketidforbindelser, diketopiperazinderivater, alkaloider, aflatoksiner og kinolinderivater. Blandt dem har dimeriske naphthopyranonforbindelser moderat antibakteriel aktivitet mod forskellige patogene bakterier, udviser en vis h\u00e6mmende aktivitet mod forskellige kr\u00e6ftceller og har ogs\u00e5 mutagene, antioxidante og relaterede enzymh\u00e6mmende aktiviteter. Vores forskningsgruppes indledende unders\u00f8gelser har vist, at den endofytiske svamp Aspergillus fumigatus SAS10 fra mangroveplanterne i Dongzhaigang Nature Reserve, Hainan-provinsen, er rig p\u00e5 metabolitter under risfermentering og -dyrkning. Tidligere unders\u00f8gelser har opn\u00e5et 5 pyranonforbindelser og 2 esterforbindelser fra denne stamme. P\u00e5 baggrund af tidligere arbejde forts\u00e6tter denne artikel med at dykke ned i den kemiske sammens\u00e6tning og antibakterielle aktivitet af risfermenteringsmetabolitter for at opdage ledende forbindelser med unikke strukturer og betydelig antibakteriel aktivitet.<\/p>\n<p>&nbsp;<\/p>\n<p>Vores forskningsgruppe unders\u00f8gte den kemiske sammens\u00e6tning af sekund\u00e6re metabolitter fra den endofytiske svamp Aspergillus fumigatus SAS10 i mangrover. Ved hj\u00e6lp af forskellige separations-, oprensnings- og identifikationsteknikker ekstraherede og isolerede vi syv dimere naphthopyranonforbindelser og en naphthopyranonvinkelmonomer fra fermenteringsbouillonen. Alle forbindelser var nyligt rapporterede sekund\u00e6re metabolitter fra den endofytiske svamp Aspergillus fumigatus i mangrover i det sydkinesiske hav, hvilket giver en ny kilde til erhvervelse af dimere naphthopyranonforbindelser. Baseret p\u00e5 forbindelsernes struktur og relateret forskning spekulerer vi i, at deres biosyntesevej er eddikesyre-malonsyre-vejen (se figur 2). Syv molekyler acetyl-CoA kan gennemg\u00e5 en r\u00e6kke reaktioner for at opn\u00e5 line\u00e6re naphthopyranon-for\u00e6ldrekerner (s\u00e5som forbindelser 8 og 11) og kantede naphthopyranon-for\u00e6ldrekerner (s\u00e5som forbindelser 9 og 10). Derefter fjernes en elektron p\u00e5 hydroxylgruppen i naphthopyranonmonomeren under p\u00e5virkning af Gip1-relaterede enzymer for at danne et bundet frit oxygenradikal. Efter elektronomlejring i det konjugerede system aktiveres kulstoffet p\u00e5 monomeren, og de to kulstofaktiverede monomerer kobles til sig selv eller til hinanden for at danne dimer 1-7.<\/p>\n<p>If\u00f8lge litteraturrapporter udviser denne type forbindelse god aktivitet i antibakterielle og antitumor-aspekter. Forbindelserne 1 og 4 viste en vis h\u00e6mmende aktivitet p\u00e5 den humane bugspytkirtelkr\u00e6ftcelle PANC-1, brystkr\u00e6ftcellen MDA-MB-231, den humane tyktarmskr\u00e6ftcelle Caco-2 og \u00e6ggestokkr\u00e6ftcellen SK-OV-3. Blandt dem viste forbindelser 1 st\u00e6rk cytotoksicitet over for PANC-1-celler med en IC50-v\u00e6rdi p\u00e5 8,25 \u00b1 2,20 \u03bc M; 2. 6 og 8 har gode antibakterielle (Bacillus subtilis, Escherichia coli og Pseudomonas fluorescens) og antifungale (Pseudomonas aeruginosa og Candida albicans) aktiviteter, hvor 2 par Bacillus subtilis og Pseudomonas fluorescens havde MIC'er p\u00e5 1,9 \u03bc g\/mL (positive kontroller var penicillin: 0.78 \u03bc g\/mL), og 6 par af Pseudomonas fluorescens og Trichophyton rubrum med MIC'er p\u00e5 7,8 \u03bc g\/mL (positive kontroller var penicillin: 0,78 \u03bc g\/mL og ketoconazol: 3,9 \u03bc g\/mL); 1. 4, 6 og 7 viste svage antibakterielle virkninger mod Candida albicans, Escherichia coli og Staphylococcus aureus (den positive kontrol for f\u00f8rstn\u00e6vnte var amphotericin B, mens den positive kontrol for de to sidstn\u00e6vnte var ampicillinnatrium). Vores antibakterielle aktivitet indikerer, at forbindelserne 1-8 har varierende grader af h\u00e6mmende aktivitet mod Staphylococcus aureus, Escherichia coli og to typer methicillinresistente Staphylococcus aureus. Blandt dem har forbindelse 6 en IC50 p\u00e5 32,42 \u03bc g\/mL mod methicillinresistent Staphylococcus aureus (MRSA N315). Derudover er den h\u00e6mmende aktivitet af dimerisk naphthopyranon mod Staphylococcus aureus (S. aureus ATCC 29213) og to methicillinresistente Staphylococcus aureus (MRSA N315 og MRSA NCTC 10442) generelt st\u00e6rkere end den for line\u00e6re naphthopyranonmonomerer. Den antibakterielle aktivitet af dimeriske naphthopyranonforbindelser med forskellige konfigurationer (line\u00e6re og kantede) varierer ogs\u00e5 til en vis grad.
