Ma, er en af kildeplanterne til Shanmaidong i Pharmacopoeia of the People's Republic of China (2010-udgaven). Steroide saponiner er dens vigtigste aktive ingrediens, og dens sapogenin-basisskelet tilh\u00f8rer derivatet af spirostan (best\u00e5ende af 27 kulstofatomer). I henhold til C25-konfigurationen i spirostanstrukturen og F-ringens cykliske tilstand kan den opdeles i f\u00f8lgende fire hovedtyper: spirostanol (C25 er S-konfiguration), isosterostanol (C25 er R-konfiguration) og furostanol (F-ringen er et \u00e5bent k\u00e6dederivat).), modificerede spirostanolalkoholer (F-ringen er en femleddet tetrahydrofuranring). Fjorten typer steroide saponiner er blevet isoleret fra Ophiopogon japonicus, hvor de vigtigste glykosider er diosgenin og ruscogenin, som begge er C25 (S)-isomerer. Blandt dem er indholdet af saponiner C (Ls-S3) i Ophiopogon japonicus relativt h\u00f8jt. Steroide saponiner har flere farmakologiske aktiviteter, og bortset fra nogle f\u00e5, der kan syntetiseres kemisk, udvindes de hovedsageligt direkte fra traditionelle kinesiske l\u00e6gemidler gennem kemisk ekstraktion. Xiangmaidongs steroide saponiner er ingen undtagelse. Men p\u00e5 grund af problemer som lange produktionscyklusser, kontinuerlige dyrkningshindringer i marken, lavt saponinindhold, h\u00f8je udvindingsomkostninger og betydelig milj\u00f8forurening er den b\u00e6redygtige produktion og forsyning af Xiangmaidong steroide saponiner begr\u00e6nset. Derfor er det n\u00f8dvendigt at finde nye m\u00e5der at fremstille saponiner p\u00e5 i store m\u00e6ngder.<\/p>\n<p>Der har tidligere v\u00e6ret unders\u00f8gelser, som rapporterede om isolering af endofytiske bakterier, der kunne producere steroide saponiner fra Dianzhong- og Huazhong-bygninger. Inspireret af dette fors\u00f8gte vores forskerteam at isolere og opdage endofytiske bakteriestammer, der metaboliserer og producerer steroide saponiner fra Xiangmaidong. Ved hj\u00e6lp af renkulturmetoder og 16SrDNA-PCRDGGE-teknologi analyserede og identificerede vi fordelingen af endofytiske bakterier i pr\u00f8ver (knolde eller r\u00f8dder) af Xiangmaidong i l\u00f8bet af dens f\u00f8rste fremkomst, hurtige ekspansion og modne h\u00f8stperioder. Samtidig blev farvereaktion, TLC og HPLC-metoder brugt til forel\u00f8bigt at evaluere saponinproduktionsevnen hos de opn\u00e5ede rene kulturstammer for at opn\u00e5 m\u00e5lstammerne og give teoretisk st\u00f8tte til \"fabriks-, skala- og hurtig\" produktion af steroide saponiner.<\/p>\n<p>Ved hj\u00e6lp af traditionelle mikrobielle renkulturmetoder isolerede denne unders\u00f8gelse 15 sl\u00e6gter og 22 arter af endofytiske bakterier fra rodknolde og fibr\u00f8se r\u00f8dder af Xiangmaidong i forskellige v\u00e6kststadier. Blandt dem var der 4 arter af Bacillus og 4 arter af Staphylococcus, 2 arter af Pseudomonas og de resterende 12 sl\u00e6gter, herunder Salmonella, Enterobacter, Pseudoxanthomonas, Morganella, Agrobacterium, Microbacterium, Porphyromobacter, Acinetobacter, Lysinibacter og Sphingosine. Der findes kun \u00e9n bakterieart i sl\u00e6gterne Sphingomonas, Sphingomonas og Curtobacterium.<\/p>\n<p>Gennem 16SrDNA PCR DGGE-identifikation fandt denne unders\u00f8gelse, at den endofytiske bakteriesamfundsstruktur i Xiangmaidong stort set var den samme i den indledende periode, den hurtige ekspansionsperiode og den modne h\u00f8stperiode for dens knolde, med lille forskel i stammegrupper. Gennem sekvenstilpasning blev 15 sl\u00e6gter og 21 bakteriearter identificeret fra de isolerede sekvenser, herunder 3 arter af Bacillus og 3 arter af Staphylococcus, 2 arter af Serratia og 2 arter af Pseudomonas, og de resterende 11 sl\u00e6gter, herunder Pseudomonas, Pectobacterium, Bordetella, Stenotrophomonas, Porphyromobacter, Curtobacterium, Enterobacter, Methylobacterium, nitrogenfiksering. Der er kun \u00e9n stamme af Azotobacter, Microbacterium og Pseudoclavibacter. Sammenlignet med de 22 stammer af rene endofytiske bakterier, der er n\u00e6vnt ovenfor, har DGGE identificeret 7 nye sl\u00e6gter: Serratia, Pectobacterium, Bordetella, Stenotrophomonas, Methylobacterium, Azotobacter og Pseudomonas.<\/p>\n<p>Denne unders\u00f8gelse evaluerede saponinproduktionsevnen hos rene dyrkbare endofytiske stammer og identificerede forel\u00f8bigt tre bakteriestammer, Pseudoxanthomonas mexicana, Staphylococcus caprae og Microbacterium testaceum, med betydelige egenskaber ved at producere steroide saponiner. If\u00f8lge HPLC-detektionsresultaterne viste disse tre endofytiske bakterier alle karakteristiske toppe p\u00e5 tidspunkterne 15,98, 57,64 og 75,19 minutter, hvilket indikerer, at mindst tre saponiner i deres fermenteringsprodukter kan have den samme struktur, hvilket kr\u00e6ver yderligere eksperimentel verifikation. P\u00e5 nuv\u00e6rende tidspunkt er der to hovedstrategier til p\u00e5visning af steroide saponiner: den ene er direkte p\u00e5visning, og den anden er indirekte p\u00e5visning gennem kvalitativ og kvantitativ analyse af spaltede saponiner. Forskere har brugt HPLC-metoden til at bestemme indholdet af dioscin i Dioscorea nipponica, arrowroot, sisal og liljeknopper. P\u00e5 grund af manglen p\u00e5 standardiserede og isolerede rene pr\u00f8ver af steroide saponiner fra Xiangmaidong i dette eksperiment blev de forberedte steroide saponiner fra Xiangmaidong-rod (blanding) midlertidigt brugt som positive kontroller. Ved at sammenligne, om karakteristiske toppe p\u00e5 samme tidspunkt kunne vises under de samme detektionsbetingelser, blev glykosidproduktionsegenskaberne for endofytiske bakterier forel\u00f8bigt identificeret. I fremtiden vil vores laboratorium yderligere adskille og rense fermenteringsproduktets saponinekstrakt og bruge massespektrometri, kernemagnetisk resonans og andre metoder til mere pr\u00e6cis karakterisering.<\/p>\n<p>Baseret p\u00e5 DGGE-analysen af den h\u00f8je variable region af 16SrDNA V6 \uff5e V8 og kombineret med screeningsresultaterne af rene dyrkede endofytiske bakterier, der er i stand til at producere steroide saponiner, fandt dette eksperiment et f\u00e6nomen: i den hurtige ekspansionsperiode og modne h\u00f8stperiode af Xiangmaidongs rodknolde kan mexicansk pseudogul pseudomonas P. p\u00e5vises fra dens rodknolde Tilstedev\u00e6relsen af mexicana blev ikke p\u00e5vist i den indledende fase af rodknolde.
