Tuberkulose for\u00e5rsaget af l\u00e6gemiddelf\u00f8lsomme stammer kan behandles med f\u00f8rstelinjemedicin som rifampicin, isoniazid, ethambutol og pyrazinamid, men fremkomsten af multiresistent tuberkulose (MDR-TB) og ekstensivt l\u00e6gemiddelresistent tuberkulose (XDR-TB) har medf\u00f8rt vanskeligheder for behandlingen af tuberkulose. P\u00e5 grund af den begr\u00e6nsede beskyttende effekt af BCG-vaccination er udviklingen af antituberkulosemedicin med nye mekanismer det vigtigste middel til at kontrollere forekomsten af tuberkulose.<\/p>\n<p>Naturprodukter er en af de vigtigste kilder til nye l\u00e6gemiddelkandidatmolekyler. I det sidste \u00e5rhundrede er et stort antal l\u00e6gemiddelkandidatmolekyler blevet opdaget fra naturprodukter. Forskere i plantekemi har udf\u00f8rt omfattende forskning i den kemiske sammens\u00e6tning af l\u00e6geplanter og etableret flere databaser over naturprodukter eller planters kemiske sammens\u00e6tning. For eksempel indsamler den kinesiske naturproduktdatabase (CNPD) omfattende oplysninger om den kemiske sammens\u00e6tning af kinesisk urtemedicin, herunder mere end 10.000 kemiske komponenter og involverer over 4500 typer kinesisk urtemedicin.<\/p>\n<p>I de senere \u00e5r har vi foretaget blindscreening af over 200 kinesiske urteekstrakter mod Mycobacterium tuberculosis og fundet ud af, at den mindste h\u00e6mmende koncentration (MIC) for over 80 kinesiske urteekstrakter mod Mycobacterium tuberculosis in vitro er mindre end 2 mg\/mL. Sammens\u00e6tningen af kinesisk urtemedicin er kompleks, og det kr\u00e6ver ofte gentagen adskillelse af ingredienser og verificering af aktivitet at belyse de aktive ingredienser. For hurtigt at opdage aktive molekyler mod tuberkulose fra disse naturl\u00e6gemidler foreslog vi ideen om \"omvendt sporingsseparation\" baseret p\u00e5 databasen med naturproduktingredienser. I dette eksperiment blev Guijianyu og kanel valgt som forskningsobjekter, og metoden med omvendt sporingsseparation blev brugt til at udf\u00f8re forskningsarbejdet: (1) ved hj\u00e6lp af databaser som China Natural Product Chemical Composition Database til at s\u00f8ge efter de kemiske komponenter i Guijianyu og kanel; (2) Gennem litteraturgennemgang blev det konstateret, at \u00e6blesyren i Guijianyu og kanelaldehydet i kanel har anti-tuberkuloseaktivitet. Ved at bruge \u00e6blesyre og kanelaldehyd som styrende molekyler blev Guijianyu- og kanelekstrakter derfor gradvist opdelt i dele, der indeholder styrende molekyler, og dele eller komponenter uden styrende molekyler; (3) In vitro-anti-tuberkulosetest afsl\u00f8rede betydelige forskelle i aktiviteten af forskellige dele eller komponenter af ekstraktet fra Guijianyu. Komponenterne med h\u00f8jere indhold af \u00e6blesyre havde st\u00e6rkere aktivitet, mens dem med lavere indhold havde svagere aktivitet. Derfor kan det konkluderes, at \u00e6blesyre er det aktive molekyle i Guijianyu mod tuberkulose. Bevis ved hj\u00e6lp af samme metode, at kanelaldehyd er et aktivt molekyle i kanel mod Mycobacterium tuberculosis; (4) Ved hj\u00e6lp af kombinationsprincippet blev der foretaget strukturelle \u00e6ndringer af \u00e6blesyre og kanelaldehyd for at opn\u00e5 mere aktive derivater.<\/p>\n<p>Denne artikel viser fordelene ved separationsmetoden med omvendt sporing til at bekr\u00e6fte de aktive ingredienser i kinesisk urtemedicin og giver et videnskabeligt grundlag for den kliniske anvendelse af disse to urter, hvilket giver nye ideer til at opdage molekyler mod tuberkulosebakterier fra kinesisk urtemedicin.<\/p>\n<p>If\u00f8lge litteraturen har \u00e6blesyren i Guijianyu og kanelaldehydet i kanel anti-tuberkuloseaktivitet. Er det s\u00e5 de st\u00e6rkeste molekyler med anti-tuberkuloseaktivitet i henholdsvis disse to l\u00e6geurter? Det er et sp\u00f8rgsm\u00e5l, der optager os meget. I dette eksperiment foreslog vi en separationsmetode med omvendt sporing, hvor vi brugte \u00e6blesyre som et styrende molekyle til at spore og isolere antituberkulosekomponenter fra Guijianyu og kanelaldehyd som et styrende molekyle til at spore og isolere antituberkulosekomponenter fra kanel. Resultaterne viste, at der ikke var nogen st\u00e6rkere komponent end \u00e6blesyre i anti-tuberkulosekomponenterne i Guijianyu, og kanelaldehyd var det mest aktive molekyle mod tuberkulose i kanel. Det kan ses, at separationsmetoden med omvendt sporing har fordelene ved at spare tid, arbejdskraft og midler i udforskningen af aktive ingredienser i kinesisk urtemedicin, is\u00e6r antibakterielle aktive ingredienser. Selvf\u00f8lgelig er denne metode afh\u00e6ngig af en st\u00e6rk database over naturlige produktingredienser som fundament.<\/p>\n<p>Derivaterne opn\u00e5et ved strukturel modifikation af forbindelser som \u00e6blesyre og cinnamaldehyd har st\u00e6rkere anti-tuberkuloseaktivitet. Forbindelse 1 har en MIC-v\u00e6rdi p\u00e5 1 \u03bc g\/mL, hvilket er sikkert og ikke-toksisk for pattedyrsceller. Det kan bruges som en ledende forbindelse til yderligere strukturel optimering af anti-tuberkulosebakterier.<\/p>\n<p>For at unders\u00f8ge den mulige virkningsmekanisme for forbindelse 1 blev der anvendt en blind screeningsmetode til at forankre forbindelse 1 med 20 proteiner fra Mycobacterium tuberculosis, hvilket indikerede, at forbindelse 1 har en st\u00e6rk affinitet for \u03b2-ketoacyl ACP-syntase III (FabH) fra Mycobacterium tuberculosis. Derfor blev virkningen af forbindelse 1 p\u00e5 ekspressionen af FabH-genet i Mycobacterium tuberculosis bestemt. Ved subh\u00e6mmende og h\u00e6mmende koncentrationer kunne det strukturelle gen for \u03b2-ketoacyl ACP-syntase III i Mycobacterium tuberculosis blokeres af forbindelse 1, hvilket f\u00f8rte til nedregulering af ekspressionen. \u03b2-ketoacyl ACP-syntase III er et n\u00f8gleenzym i syntesen af langk\u00e6dede fedtsyrer i cellev\u00e6ggen hos Mycobacterium tuberculosis. N\u00e5r det h\u00e6mmes, vil det for\u00e5rsage \u00e6ndringer i Mycobacterium tuberculosis' morfologi, hvilket yderligere f\u00f8rer til beskadigelse af cellev\u00e6gsstrukturen og for\u00e5rsager bakteriel h\u00e6velse og opl\u00f8sning. Gennem mikroskopiske billeder kan det ses, at tuberkulosebakteriernes morfologi underg\u00e5r betydelige \u00e6ndringer efter intervention med forbindelse 1, hvilket indikerer, at forbindelse 1 for\u00e5rsager skade p\u00e5 tuberkulosebakteriernes cellev\u00e6g. Virkningen af cellev\u00e6gs\u00f8del\u00e6ggelsen p\u00e5 Mycobacterium tuberculosis' vitalitet er d\u00f8delig. Ved at m\u00e5le den bakteriedr\u00e6bende kurve fandt vi ud af, at forbindelse 1 (4 \u03bc g\/mL) dr\u00e6bte alle Mycobacterium tuberculosis i kultursystemet inden for 72 timer. Derfor spekulerer vi i, at antituberkulosemekanismen for forbindelse 1 er relateret til interventionen af \u03b2-ketoacyl ACP-syntase III. Da dette eksperiment ikke gav afg\u00f8rende bevis for, at forbindelse 1 binder sig til m\u00e5let, skal virkningsmekanismen for forbindelse 1 stadig unders\u00f8ges n\u00e6rmere.<\/p>\n<p>Den gamle bog fra det \u00f8stlige Han-dynasti, \"The Emergency Prescription for Back up the Elbow\", foreslog f\u00f8rst brugen af sp\u00f8gelsespilfjer til at kurere sp\u00f8gelsesinjektion og liginjektion.
